Mechanism of signal transduction of blood-brain barrier damage after brain injury in rats

Abstract: Objective To explore the mechanism of signal transduction of blood brain barrier (BBB) damage in brain injury. Methods Evans blue dye exudation in the brain was observed with an fluorescence microscope and measured quantitatively with a spectrophotometer after brain injury in the rats treated with H7, a protein kinase C(PKC) inhibitor and forskolin, a re- agent that elevates cellular content of cAMP. Results Exudation of Evans blue dye was significant in the injured group and decreased significantly in the treatment group with H7 and forskolin treatment. Conclusion Forskolin and H7 can significantly prevent BBB opening, demonstrating that cAMP and PKC take an important role in BBB damage after brain injury.
