Fas and FasL expressions on the different stages of large intestine malignant transformation and their significance in apoptosis escape of cancer

Abstract:Objective:This article attempts to explore the molecular mechanism underlying the Fas counterattack by observing the expression of Fas and FasL and apoptosis and further analysing the relationships between them.Methods:Using insitu hybridization,and TUNEL techniques,it was examined a number of paraffin embeded tissues including37cases of large intestinal adenocarcinomas,26cases of malignantly transformed adenomas,30cases of tubulovillous adenomas,and24cases of tubular adenomas for Fas mRNA,FasL mRNA and DNA fragment.6cases of non-tumor mucosas are used as control group.Results:With the progression of the malignant transformation,Fas mRNA expressions increase slightly in benign adenomas,and then decrease significantly in malignant diseases.Meanwhile,FasL mRNA exˉpressions show an increasing tendency.Apoptotic lymphocyte densities show an increasing tenˉdency,too.Apoptotic epithelial cell densities increase in benign tumors and then decrease in malignant tumors.There is a positive correlation(r=0.672,P0.001)between FasL expres_ sions and apoptotic lymphocyte densities in adenocarcinomas.Conclusion:With the progression of the large intestine malignant transformation,the Fas counterattack is forming progressively.Cancer cell expressing FasL induce tumor infiltrating lymphocytes to undergo apoptosis to esˉcape from organic immune surveillance through Fas-FasL interactions.However,the cancer cells may downregulate Fas expression by themselves to resist the death signals transduced by FasL expressed by the immune effector cells and cancer cells,forming the Fas resistance mechanism---the prerequisite of the Fas counterattack,and therefore inhibiting the apoptosis of the cancer cells.Thus,together with Fas resistance,Fas counterattack leads to the formation of the large intestine cancer in the end.
