Clinical Application of Flow Cytometry for Measurement of Platelet-Associated IgG

Abstract:Objective:To investigate the characteristics of measurement of platelet-associated IgG (PAIgG) by flow cytometry (FCM) and its significance in the diagnosis of idiopathic thrombocytopenic purpura (ITP).Method:PAIgG was measured by using FCM in 47 patients with ITP, 13 cases with non-immune thrombocytopenia, 10 cases with AIHA, 18 cases with other immune system diseases and 31 healthy volunteers, in comparison with ELISA. Result:With FCM measurement, the level of mean fluorescence intensity (MFI) and percentage positive platelets in patients with ITP was significantly higher than that in control subjects (P 0.01 ) and an apparent peak shape abnormality was observed in histogram of fluorescence intensity (log) in 11 out of 47 cases of ITP. The positive percentage of PAIgG by FCM in ITP ( 87.2 %) was slightly higher than that by ELISA ( 83.0 %). The rate of accordance of these two methods was 85.1 %. However,these two methods showed low specificity. Conclusion:FCM analysis is a rapid, simple and sensitive method for the measurement of PAIgG in patients with immune thrombocytopenia. It may be suitable to clinical diagnostic application.
