The Effect of Psychological Stress and Heat Shock Stress on the Emotion and Learning Ability of Mice

Abstract:ObjectiveTo explore the effect of psychological stress and heat shock stress on the emotion and learning ability of mice, and to find out whether the heat shock stress has a protective role on the negative behavioral response resulting from psychological stress or not. Methods40 mice from reproduction of mice of HSF1 knocked out was assigned into five groups: basic group(O); control group(A); heat-shock-stressed group(B); psychological-stressed group(C); heat-shock-stressed plus psychological-stressed group(D).One month after stress, the anxiety degree and learning ability were measured, then compare the difference between the results got from the five groups. The correlation between the level of anxiety and the learning ability. Results The results are included in the following: (1)The level of emotional arousal in C group is increased and it is higher than in both B(P=0.008) and D(P=0.008) groups; (2) The ratio of the central squares of mice scrawl across shows that the ratio in the B group is significantly higher than that in the A(P=0.007) and both in the C(P=0.001) and D(P=0.027) are lower than that in the B.(3) The frequency of the open arms entrance is significantly increased in the B(P=0.007) group;(4) The time spent in the maze explore is significantly decreased both in the B(P=0.047) and C groups(P=0.013);(5) The time spent in the maze explore is negatively correlate to both the central squares entrance frequency(r=-0.472,P0.05) and the ratio that the central squares to the total(r=-0.452,P0.05), the number of maze that the mice pass is positively correlate to the central squares entrance frequency(r=-0.384,P0.05). Conclusions Psychological stress increases the level of the anxiety and heat shock stress decreases the anxiety in mice . The heat stress reduces the emotional arousal resulting from psychological stress. Both kinds of stresses increase the spatial learning ability.
