Proliferative Responses of T Cell Lines from Multiple Sclerosis Patients to Different Myelin Compositions

Abstract:Objective To compare the proliferative responses of T cell line (TCL) to myelin basic protein (MBP), proteolipid protein (PLP) and their synthetic peptides in multiple sclerosis (MS) patient. Methods Human T cell lines, from mononuclear cells of HLA-DR15 type, were established using intact myelin by short period culture in vitro and then their proliferative responses to 11 antigens, such as MBP, PLP and their synthetic peptides, were observed. Results The recognized myelin components in either MS or control displayed polymorphism. TCLs from MS recognized 7 antigens of 11 (5 antigens recognized by control). When their responses were compared with MS and control, the difference was statistically significant to MBP, P30-49, P95-117 and P178-191. However, there was no significant difference between their average positive wells. Conclusions Our results suggest that MBP and PLP might play some important role in initiating demyelinating diseases, and that M87-106, P40-60, P95-117 and P185-206 might be candidates for antigen epitope relevant to MS.
