Alteration of Renal Hemodynamic in Rats Treated with Aristolochic Acid

Abstract:Objective:Aristolochic acid Ⅰ,an ingredient of the Chinese herbs mutong ,has been associated not only with chronic,but also acute forms of renal failure in clinical settings.The hemodynamic changes in AAI induced nephrotoxicity has seldom been investigated.In this study,the acute renal hemodyanamic changes were investigated after acute intravenous administration of AAI in rats. Methodology:Male SD rats (250~300g) were randomized into four groups with 6 rats in each group.Intravenous AAI(the treatment groups,1 0,5 0,or 25 0 mg/kg) or saline(the control group) were administreted after anesthesia.The mean arterial blood pressure(MABP) was detected with a manometer.Renal plasma flow(RPF) was calculated as the renal clearance of para aminohippuric acid(PAH).Glomerular filtration rate(GFR) was calculated as inulin clearance.Renal vascular resistance(RVR) was calculated as the division of MABP by renal blood flow(RBF). Results:There were no significant changes of MABP,RPF,GFR,and RVR observed in the control group.MABP was found not significantly influenced after administration of AAI in different dosage groups of rat.Significant increment of urine volume,RPF,GFR,and significant decrement of RVR were observed after the intravenous administration of AAI ( P 0 01,compared to the pre administration data).The changes of urine volume,RPF,and GFR in all AAI groups peaked within the first 60 min after AAI administration and gradually decreased to normal range thereafter. Conclusion:AAI, at the dosage of 1~25 mg/kg,does significantly influence the renal hemodynamic parameters urine volume, RPF GFR,and RVR in rats.These hemodynamic changes gradually returned to normal range 60 minutes after the administration of AAI
