Abstract:The deoxyglucose dithiocarbamate (DGDTC) was radiolabeled with 99mTc(V)‐glucoheptonate (GH), for the potential use as radiopharmaceuticals for tumor imaging. For labeling, 99mTcO‐DGDTC was prepared by ligand‐exchange reaction with 99mTc‐GH. The radiochemical purity of the 99mTcO‐DGDTC complex was over 90% by thin‐layer chromatography and high‐performance liquid chromatography, without any notable decomposition at room temperature over a period of 6 h. Its partition coefficient indicated that it was a hydrophilic complex. The ligand‐exchange reaction occured at neutral condition and under 100 °C for 15 min to achieve high radiochemical purity. In vitro cell studies showed there was an increase in the uptake of 99mTcO‐DGDTC as a function of incubation time and the cellular uptake of 99mTcO‐DGDTC was possibly mediated by way of a d‐glucose mechanism. The biodistribution of 99mTcO‐DGDTC in mice bearing S 180 tumor showed that the complex accumulated in the tumor with good uptake and excellent retention. As compared with other reported 99mTc radiolabeled glucose derivatives, 99mTcO‐DGDTC showed the highest tumor uptake and good tumor/muscle ratios. The tumor/muscle ratio of 99mTcO‐DGDTC uptake was higher than that of [18F] FDG uptake. Single photon emission computed tomography (SPECT) image studies showed there was a visible accumulation in tumor sites, suggesting 99mTcO‐DGDTC would be a promising candidate for tumor imaging.

Abstract:18F-2-fluoro-2-deoxy-d-glucose ([18F]FDG) has been the most used positron emission tomography imaging agent for clinical applications. Single photon emission computed tomography (SPECT) imaging is cheaper and used more widely for diagnostic use, but there is no SPECT tumor imaging agent for clinical applications comparable to [18F]FDG. Mannose is a C2 epimer of glucose and can also be transported into tumor cells via glucose transporters (GLUTs). To develop a novel SPECT tumor imaging agent with satisfactory tumor uptake and tumor/nontarget ratios, here a mannose derivative (CN7DM) was synthesized and radiolabeled with technetium-99m to prepare [99mTc]Tc-CN7DM. The six-coordinated structure of [99mTc]Tc-CN7DM was confirmed by the corresponding rhenium compound (Re-CN7DM). [99mTc]Tc-CN7DM was transported into cancer cells via GLUTs and may be trapped in the cancer cells by electrostatic attraction. The probe exhibited high uptake in tumors and low uptake in nontarget tissues in mice bearing different tumors, indicating that [99mTc]Tc-CN7DM exhibited promising potential for SPECT tumor imaging and warranted further clinical investigation.

Abstract:Sodium 3-(4-nitro-1H-imidazolyl) propyl dithiocarbamate (N4IPDTC) was synthesized and radiolabelled with a [(99)mTcO](3+), [(99)mTc N](2+) or [(99)mTc(CO)(3)](+) core to produce (99)mTcO-N4IPDTC, (99)mTcN-N4IPDTC and (99)mTc(CO)(3)-N4IPDTC, respectively. All of the three complexes were prepared with high radiochemical purity and had good in vitro stability over a period of 6 h. The partition coefficient results showed that (99)mTcO-N4IPDTC and (99)mTcN-N4IPDTC were lipophilic, while (99)mTc(CO)(3)-N4IPDTC was hydrophilic. The tumor cell experiments and the biodistribution in mice bearing S180 tumors showed that all of the complexes exhibited good hypoxic selectivity and accumulation in the tumor. Among them, (99)mTcO-N4IPDTC had the advantages of higher tumor uptake, and higher tumor/blood and tumor/muscle ratios. Planar scintigraphic imaging studies showed there was a visible accumulation in tumor sites, suggesting its potential usefulness as a tumor hypoxia imaging agent.

Abstract:DGDTC was successfully radiolabeled with [99mTc(CO)3(H2O)3]+ precursor to obtain 99mTc(CO)3–DGDTC in high yield. The biodistribution and SPECT studies in various tumor animals showed that 99mTc(CO)3–DGDTC exhibited very promising properties for tumor imaging.

Abstract:During the last four decades, nuclear medicine has undergone enormous growth, and positron emission tomography (PET) has been in the driving seat for most of the time. 18F-fluorodeoxyglucose (18F-FDG) is the most widely used agent for the detection of hibernating myocardium and metabolically active cancer tissue. But its cost and limited availability are the main limitations. For a long time different researchers and groups of pharmacists have tried to label glucose with a cheaper and long-acting radionuclide like 99mTc. However, they failed to achieve this goal owing to the chemical complexity of 99mTc and the lack of maintaining the physiological activity of diagnostic compounds. A pre-targeting strategy based on strain-promoted [3 + 2] azide-alkyne cycloaddition (SPAAC) reaction was applied to solve this problem. Functional click synthons were synthesized: 2-azido-2-deoxy-d-glucose (GlucN3) as a glucose analogue, and N- (2- (2- (2- (bis (pyridin-2-ylmethyl) amino) ethoxy) ethoxy) ethyl-2- (6H-11,12-didehydrodibenzo [a,e] cycloocten-5-ylideneaminooxy) acetamide (C7) as a 99mTc(CO)3 labeling and azido-binding group. The results of biodistribution experiments in mice bearing S180 tumor show the relatively high tumor/blood ratio (up to 2.95) and tumor/muscle ratio (up to 6.37), and both of them decreases significantly in the glucose blocking experiment. It indicates that GlucN3 behaves similarly to glucose and that in vivo SPAAC reactions can occur effectively. It is supposed that this pre-targeting strategy can indeed enhance target specificity and may be used for glucose metabolism imaging in tumor diagnosis.

Abstract:Objective To evaluate the pharmacokinetic characteristics and feasibility of imaging in tumor-bearing mice and rabbits of [99TcmN]2+ intermediate radiolabelled deoxyglucose dithiocarbamate(DGDTC).Methods The DGDTC was synthesized and radiolabelled with [99TcmN]2+ to form the 99 TcmN-DGDTC complex under the guideline of kit.Pharmacokinetics data of healthy rabbits were acquired by collecting radio counts of auricular vein and processed with software DAS2.1.1.Animal tumor model(10 mice with H22 transplanted cancer and 5 rabits with VX2 transplanted cancer) imaging was designed to two groups,99TcmN-DGDTC group and 99Tcm pertechnetate group(as a control).Scintigraphy was performed with blood flow,blood pool,and static acquisition continuously,till 2-4 hours after injection.Results The pharmacokinetics results were proved to be a two-compartment model,and t1/2 was 34.05 minutes.Scintigraphic results showed 99 TcmN-DGDTC complex could display tumor well in mice and rabbits tumor model.The count ratio of tumor to nontumorous tissue kept above 2 within 2 hours injected,suggesting the well tumor utilizing of 99 TcmN-DGDTC.Conclusion The pharmacokinetics results and scintigraphic findings supported the potential use of 99 TcmN DGDTC as a functional imaging agent.

Abstract:Objective To investigate the possibility of 99mTc-MAG3-Glucosc used for tumor imaging,99mTc-MAG3-Glucose was synthesized and its tumor localizing properties were studied.Methods β-N-mercaptoaeetyl-Gly-Gly-Gly-glucose ( MAG3-Glu)was labeled with 99mTc.The labelled compound was injected into the mice bearing S180 tumor,and biodistribution study and imaging were performed.The biodistribution of 18F-2-fluoro-desoxy glucose( 18F-FDG) at 5 h postinjection was also studied for comparison.Results 99mTc labeling yield was greater than 90％.The tumor uptakes at 1 h,3 h and 5 h postinjection were ( 2.45 ± 0.25 ) ％ ID/g,( 2.23 ± 0.53 ) ％ ID/g and ( 2.00 ±0.89)％ID/g,respectively.The T/NTs were greater than 1.7 for tumor over all other normal tissues excepted for lung,kidney and liver at 5 h postinjection.Tumor/blood,tumor/muscle and tumor/brain were 1.71,5.71 and 28.57,respectively.Tumor was visualized at 3 h postinjection,and the contrast became slightly sharper with time lasted.Compared with 18F-FDG,99Tc-MAG3-glu had lower tumor uptake and T/NTs for most tissues.But its T/NTs for tumor over muscle,heart,spleen and brain,especially for tumor over brain,were higher.Conclusions The results suggested the potential utility of 99mTc-MAG3-Glu as a suitable tumor imaging agent.The high tumor over brain uptake ratio indicated that 99Tc-MAG3-glu may have advantages in imaging brain tumor and the tumor near brain.
Preparation of New Technetium-99m Nns/X Complexes and Selection for Brain Imaging Agent

Abstract:Based on excellent experiment results of 99m TcO-MPBDA-CI, two new ligands MPTDA and MPDAA are synthesized. Then series of 99m TcO 3+ complexes are prepared through adding different halide anions, followed by tests of physical chemistry qualities and biodistribution experiments. And results of these experiments show that complexes formed with MPTDA and MPDAA have better lipophilicity than those formed with MPBDA, still maintain the good brain retention ability of this type of compounds, but radioactivity uptake in blood is higher than that of 99m TcO-MPBDA and ratios of brain/blood are reduced. Obvious affections are fetched out on brain uptake and retention if fluoride, bromide or iodide anions are added. Results of experiments can be explained in reason with theoretic computation. It is confirmed that 99m TcO-MPBDA-CI has potential to develop a new type of brain imaging agent considering integrated factors such as brain uptake, retention and toxicity.

Abstract:In order to visualize hypoxic tissues, a 99mTc complex containing a nitroimidazole group was evaluated. A new dithiocarbamate ligand, potassium 2-(4-nitro-1H-imidazolyl)ethyl dithiocarbamate (NIET) was synthesized. The corresponding nitridotechnetium complex was formed by a ligand exchange method. The dependence of the radiolabeling yield of 99mTcN-NIET on the amount of added stannous chloride and other parameters was screened by the normal procedure. Under optimized labeling conditions, the radiolabeling yield of 99mTcN-NIET is about 90%. The biodistribution of 99mTcN-NIET in mice bearing S180 tumor demonstrated some uptake in tumor and a tumor-to-muscle ratio of 3.14 at 4 hours after injection.

Abstract:Introduction: Achieving an ideal Tc-99m labeled nitroimidazole hypoxia marker is still considered to be of great interest. Metronidazole xanthate (MNXT) ligand was synthesized and radiolabeled with Tc-99m-glucoheptonate (GH) to form the (TcO)-Tc-99m-MNXT complex, for the potential use as a novel probe for imaging tumor hypoxia. Methods: For labeling, (TcO)-Tc-99m-MNXT was prepared by ligand-exchange reaction with Tc-99m-GH. The radiochemical purity of the (TcO)-Tc-99m-MNXT complex was measured by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). The distribution coefficient and stability of the complex was investigated. The structure of the (TcO)-Tc-99m-MNXT complex was verified by preparation and characterization of the corresponding stable rhenium complex. The cellular uptake of the (TcO)-Tc-99m-MNXT complex was determined in murine sarcoma 5180 cell lines under hypoxic and aerobic conditions. The biodistribution and single photon emission computed tomography (SPECT) image studies of the (TcO)-Tc-99m-MNXT complex were performed in mice bearing S 180 tumor. Results: The radiochemical purity of the (TcO)-Tc-99m-MNXT complex was over 90%. It had good in vitro stability and its distribution coefficient indicated that it was a hydrophilic complex. When Tc-99m and Re complexes were coinjected in HPLC, both radioactivity (for Tc-99m complex) and UV detectors (for Re complex) showed nearly identical HPLC profiles, suggesting their structures are similar. The tumor cell experiment and the biodistribution in mice bearing S 180 tumor showed that the (TcO)-Tc-99m-MNXT complex had a good hypoxic selectivity and accumulated in the tumor with high uptake and good retention. Single photon emission computed tomography (SPE') image studies showed that the tumor detection was observable. Conclusions: (TcO)-Tc-99m-MNXT is prepared from a kit without the need for purification and shows high tumor uptake, tumor/blood and tumor/muscle ratios, suggesting that it would be a promising candidate for imaging tumor hypoxia. Abstract:Tumor hypoxia can decrease the efficacy of clinical therapy due to resistance toward radiation damage and chemotherapy, thus detection of tumor hypoxia by radiolabeled hypoxia markers is important for the control of tumor. Radiopharmaceuticals with two bioreductive groups, such as propylene amine oxime-bisnitroimidazole or monoamine-monoamide dithiol (MAMA) -bisnitroimidazole, have potential to improve hypoxia selectivity. In order to obtain radiopharmaceuticals with better features, we synthesized two novel [99mTcN]2+ complexes with bisnitroimidazole moieties and MAMA ligand for targeting tumor hypoxia. Their physicochemical characters and biodistribution were also investigated. Both the [99mTcN]2+ complexes show good stability and hydrophilicity. They show faster clearance from blood and soft tissues, better tumor retention and favorable tumor-to-tissue ratios compared with a control complex without nitroimidazole group. In addition, both of them show more favorable biodistribution patterns than the corresponding [99mTcO]3+ complexes. These results indicate that the 99mTcN-labeled MAMA-bisnitroimidazole complexes would have potential to image tumor hypoxia in vivo.

Abstract:Twelve new peptide or pseudo-peptide chelators have been synthesized in the course of our continuing investigation of 99mTc-labelled peptides for application for renal imaging agents. All compounds were characterized on the basis of, IR, 1HNMR, 13CNMR spectroscopy, as well as MS and elemental analysis. All peptides yield stable 99mTc complexes using Sn(II) reactive coupling and exhibit renal uptake. Linear regression analysis between Logarithm of renal uptake value (RU) and the parameters obtained by the ZINDO/1 method was performed. Some equations were obtained which showed that molecular polar and charge have some relationship with their renal uptake.
99 Tcm-labeled Glucose Metabolism Imaging Agents Based on in Vivo Copper-free Click Chemistry

Abstract:In order to develop an ideal 99Tcm-labeled glucose metabolism imaging agent, MAMA-conjugated azadibenzocyclooctyne derivative and four 2-azido glucose derivatives were synthesized. Two potential candidates (N3-DG-1 and N3-DG-2) were selected according to the in vitro kinetic study experiment. The 2-azido glucose derivatives were pretargeted in mice bearing S180 tumor, then 99Tcm-labeled MAMA-azadibenzocyclooctyne derivative was administrated to accomplish the in vivo copper-free click reaction and imaging of glucose metabolism. The biodistribution results showed that the tumor/blood and tumor/muscle ratios of N3-DG-2 were higher than those of N3-DG-1, indicating the combination of 99Tcm-labeled MAMA-azadibenzocyclooctyne derivative and N3-DG-2 was more suitable for glucose metabolism imaging.

Abstract:Currently, a popular strategy for designing novel radioprobes as bone‐imaging agents is based on the concept of bifunctional radiopharmaceuticals. Considering the dithiocarbamate ligand can act as a suitable bifunctional linking agent to attach technetium‐99m (99mTc) to corresponding target molecules, in this study, alendronate dithiocarbamate (ALNDTC) was synthesized and radiolabeled with [99mTc≡N]2+ core by ligand exchange reaction to produce 99mTcN‐ALNDTC complex, for the potential use as a novel probe for bone imaging. The radiochemical purity of the complex was over 90%. The complex was stable in vitro and could bind to hydroxyapatite. The partition coefficient result indicated it was hydrophilic, and an evaluation of biodistribution in mice indicated that the complex exhibited a higher bone uptake than did 99mTc‐labeled methylenediphosphonate (99mTc‐MDP). Further, single photon emission computed tomography imaging study indicated clear accumulation in bone, suggesting that 99mTcN‐ALNDTC would be a promising candidate for bone imaging.
Radiolabeling and Preliminary Evaluation of 99mTc-Labeled DNA Cube Nanoparticles as Potential Tracers for SPECT Imaging

Abstract:Purpose: DNA nanostructures, with the advantages of structural designability and spatial addressability, have shown a great potential in the field of drug delivery and bio-medicine. Herein, we aimed to prepare technetium-99m radiolabeled DNA cube nanoparticles (99mTc-DCN) and expect to build a DCN-based drug carrier and nuclear medicine imaging platform. Methods: DCN could be readily assembled with 6 designed DNA oligonucleotides at an equal mole ratio in a single annealing procedure. 99mTc-MAG3-ssDNA (A20) was obtained by labeling MAG3-ssDNA (A20) with technetium-99m by using a stannous chloride reduction method. 99mTc-DCN was prepared by hybridize DCN with side chains (T20-DCN) with 99mTc-MAG3-ssDNA (A20). The biodistribution study and SPECT/CT imaging were conducted on KM mice. Results: DCN was successfully assembled, and as-prepared DCN was characterized by native polyacrylamide gel electrophoresis, atomic force microscope and fluorescence resonance energy transfer. The size of DCN was about 5 nm. The radiolabeling yield of 99mTc-MAG3-ssDNA (A20) was approximately 90% by radio thin-layer chromatography. T20-DCN mixed with 99mTc-MAG3-ssDNA (A20) in PBS could generate 99mTc-DCN upon hybridization. The retention time (RT) of 99mTc-MAG3-ssDNA (A20) was at ~22 min, and the RT of as-prepared 99mTc-DCN was at ~12 min by radio-HPLC. The results from biodistribution study and SPECT/CT imaging showed that a significant proportion of DCNs were metabolized through the liver and kidney. Intestine exhibited a relatively indicative signal as well, which might be explained by the enterohepatic circulation of DCN via the liver and gallbladder. Conclusion: We have successfully prepared 99mTc-DCN as a SPECT/CT imaging probe via the side-chain hybridization strategy. The probe was metabolized mainly by the liver and excreted primarily to the bladder. Due to the superior properties of DNA cube nanoparticles, we believe DCN may potentially be translated into a preclinical setting for diagnosis and treatment of cancer-related diseases.

Abstract:Hypoxia, as a common phenomenon in solid tumors, is of interest for its relationship with resistance to tumor therapies and malignant progression of tumor. The noninvasive nuclear medical imaging technique using hypoxia markers is an important method for the detection of tumor hypoxia. The aim of current study is designing tumor hypoxia markers with hypoxia selectivity and improved properties. Two MAG(3)-based bisnitroimidazole compounds were synthesized and purified by semipreparative HPLC. Both the MAG(3) derivatives were labeled with Tc-99m-oxo-technetium core via stannous tartrate exchange method in high yields (> 95%). The Tc-99m-MAG 3 complexes were stable at 37 degrees C, 4 h after preparation, and were more hydrophilic than Tc-99m-MAMA complexes. As biodistribution results showed, clearances of background activity for both the complexes were fast and they were excreted mainly through the hepatobiliary tract and part of renal tract. Although tumor uptakes of Tc-99m-MAG(3)-B2NIL were lower than those of Tc-99m-MAG(3)-B4NIL, tumor-to-blood ratios of Tc-99m-MAG(3)-B2NIL showed an increasing trend and were better than those of Tc-99m-MAG(3)-B4NIL after 2 h due to their different blood clearances. Tumor-to-muscle ratios of Tc-99m-MAG(3)-B2NIL and Tc-99m-MAG(3)-B4NIL were similar. Comparing with Tc-99m-MAMA complexes, Tc-99m-MAG(3)-B2NIL with better tumor-to-blood ratios exhibits improved feature for hypoxia imaging, though it has lower tumor uptake than Tc-99m-MAMA complexes.

Abstract:This work reports the synthesis, radiolabeling and biological studies of 99mTc(HYNIC–MFL)(tricine)(TPPTS) in tumor-bearing mice. The novel melphalan derivative was successfully synthesized by conjugation of HYNIC to melphalan. The ligand could be labeled by 99mTc in high yield to get 99mTc(HYNIC–MFL)(tricine)(TPPTS), which was very hydrophilic and was stable at room temperature. Biodistribution studies in tumor-bearing mice showed that 99mTc(HYNIC–MFL)(tricine)(TPPTS) accumulated in the tumor with favorable uptake and retention. The good accumulation in tumor tissue, high tumor/muscle ratios and satisfactory scintigraphic images highlight the potential of 99mTc(HYNIC–MFL)(tricine)(TPPTS) for tumor imaging.

Abstract:We report the design, synthesis and biological evaluation of a novel 99mTc 4-(4-cyclohexylpiperazine-1-yl)-butan-1-one-1-cyclopentadienyltricarbonyl technetium ([99mTc]5) as a potential SPECT tracer for imaging of σ2 receptors in tumors. [99mTc]5 was prepared in 25±5% isolated radiochemical yield with radiochemical purity of >99% via double-ligand transfer (DLT) reaction from the ferrocene precursor 2b (4-(4-cyclohexylpiperazine-1-yl)-1-ferrocenylbutan-1-one). The corresponding Re-complex 4 and the ferrocenyl complex 2b showed relatively high affinity towards σ2 receptors in in vitro competition binding assay (Ki values of 4 and 2b were 64.4±18.5nM and 43.6±21.3nM, respectively) and moderate to high selectivity versus σ1 receptors (Kiσ1/Kiσ2 ratios were 12.5 and 95.5, respectively). The logD value of [99mTc]5 was determined to be 2.52±0.33. Biodistribution studies in mice revealed comparably high initial brain uptake of [99mTc]5 and slow washout. Administration of haloperidol 5min prior to injection of [99mTc]5 significantly reduced the radiotracer uptake in brain, heart, lung, and spleen by 40–50% at 2h p.i.. Moreover, [99mTc]5 showed high uptake in C6 glioma cell lines (8.6%) after incubation for 1h. Blocking with haloperidol to compete with [99mTc]5 significantly reduced the cell uptake. Preliminary blocking study in C6-brain-tumor bearing rats showed that [99mTc]5 binds to σ receptors in the brain-tumor specifically. These results are encouraging for further exploration of 99mTc-labeled probes for σ2 receptor tumor imaging in vivo.

Abstract:1-(3-1,2,4-Nitrotriazole-1-yl)-propanhydroxyiminoamide and 1-(6-nitrobenzoimidazole-1-yl)-propanhydroxyiminoamide were synthesized and radiolabeled with 99mTc. The 99mTc labeled complexes continuously accumulated in hypoxic murine sarcoma S180 cells in vitro but not in aerobic cells. Biodistribution results in mice bearing S180 tumor indicated that the tracers could localize in tumor and eliminate from it slowly. These results suggested that the 99mTc labeled nitrobenzoimidazole and nitrotriazole might be the novel tumor hypoxia markers.
