Abstract:Objective To study DNA damage induced by aminophenols in human peripheral blood lymphocytes and mouse spleen cells in vitro. Methods Isolated human peripheral blood lymphocytes and mouse spleen cells were exposed to the tested substances for 1h respectively. The comet assay was performed and the rates of damaged cells and arbitrary units were calculated. Results All of three kinds of aminophenols caused DNA damage to two kinds of cells. The DNA damage scoring was increased with the increase of doses of the tested substances and significant difference was observed at high concentrations compared with the solvent controls. The genotoxicity of p-aminophenol was stronger than o-aminophenol and m-aminophenol. Conclusion The results of this study indicated that the human peripheral blood lymphocytes were more sensitive to the aminophenols than mouse spleen cells. The comet assay of human peripheral blood lymphocytes might be a suitable and sensitive test for biomonitoring genotoxicity on human population.
