The comparison among three methods of inducing an experimental autoimmune thyroiditis animal model

Abstract:ObjectiveTo adopt three different methods fo r establishing the model of experimental autoimmune thyroiditis (EAT) in mice an d to compare the effectiveness and simplicity of these three methods. This study is aiming at providing an ideal EAT model for further investigating the pathoge nesis of autoimmune thyroid diseases.MethodsThree different methods of ① injections of het erogenous mice thyroglobulin (Tg) emulsified with complete Freund’s adjuvant (C FA) into experimental mice (considered as Tg+CFA group) ② transplantation of he terogenous thyroid to experimental mice peritoneal cavity (considered as thyroi d transplantation group)③ transfer of lymphocytes activated by heterogenous Tg in vitro to experimental mice (considered as transfer of lymphocytes group) were u sed separately to induce EAT in mice. Four weeks after immunization, sera were s creened for anti-Tg-antibody and the histological grade of EAT was assessed by blind evaluation of thyroid specimen. Unimmunized mice were used as control. ResultsIn the Tg+CFA group, the level of anti-Tg anti body was four times higher than that of the control and there were 5 cases whose lymphocytic infiltration index was ≥2 that took up about 50% of the experimen tal mice.The thyroid tissues from the other two experimental groups showed no si gnificant differences from the control.ConclusionsEAT can be characterized as high level of a nti-Tg antibody and lymphocytic infiltration in the thyroid tissue. From this s tudy, we may conclude that the method of injections of heterogenous mice thyrogl obulin (Tg) emulsified with complete Freund’s adjuvant (CFA) into experimental mice can induce EAT with more effectiveness and simplicities than the other two methods. It is an ideal method for inducing EAT model in further investigating t he pathogenesis of autoimmune thyroid diseases.
