Parathyroid Hormone Inhibits Expression of Membrane-Type Matrix Metalloproteinase-1 and is Invoved in RANKL Transduction in Osteoblast-Like MG-63 Cells

Abstract:Objective To characterize the effects of parathyroid hormone (PTH) on the expression of membrane-type 1 matrix metalloproteinase-1 (MT1-MMP) and receptor activator of NF-κB ligand (RANKL) in human osteoblast-like cell line MG-63 in order to elucidate the mechanism by which PTH acts on bone. Methods MG-63 cells were treated with PTH (1-34). Northern blotting and western blotting were used to show the actions of PTH(l-34) on expression of MT1-MMP, RANKL mRNA and protein of MG-63 cells, separately. Results PTH (1-34) inhibited MT1-MMP mRNA and protein expression in a dose-and time-dependent manner in MG-63 cells. The PKA antagonist H-89 blocked PTH(1-34)-mediated decreases in MT1-MMP protein synthesis. Forskolin, a PKA agonist, decreased MT1-MMP expression, which was similar to the action of PTH on MT1-MMP expression in MG-63 cells. Staurosporine, a PKC inhibitor, also blocked the inhibition by PTH. We suggest that both PKA and PKC pathways might be involved in MT1-MMP down-regulation by PTH. Furthermore, we found that PTH (1-34) induced RANKL mRNA expression in a dose-and time-dependent manner in MG-63 cells, and this effect of PTH on RANKL mRNA expression was nearly paralleled to those of MT1-MMP down-regulation. It implies the correlation between MT1-MMP and RANKL expression. Conclusions Decreased MT1-MMP expression by PTH is involved in RANKL signaling in osteoblasts, and PTH plays a role in activation of bone resorption.
