Abstract:Dendritic cells(DC) are the most powerful professional antigen presenting cells.Lots of studies have demonstrated that the DC vaccines play an important role in antitumor immunity.With the developments of molecular biology,immunology and genetic engineering,rapid progress has been made in DC vaccines and application of DC-based immunotherapy.In this review,the DC biological property,the relationship between the development of cancer and dendritic cell,the production of dendritic cell tumor vaccines and their use in clinical trials were discussed.It may provide valuable information with respect to DC primary study and clinical trials.

Abstract:Cancer is one of the top killers which threatens global people's health. Although current treatments such as surgery, radiotherapy and chemotherapy have been improved, prognosis remains unsatisfactory. Developing new therapeutic strategy is still an urgent demand. Dendritic cell vaccines based immunotherapy activated immune cells can specifically kill tumor cells by recognition of tumor-associated antigens without injuring normal cells. Abstract:Although tumor immunotherapy has been proposed for many years,the consensus denoting it as an essential approach for fighting against cancer is reached only in recent years. Tumor immunotherapy can be categorized as active and passive ones. In order to successfully cure cancer,safe and efficient active immunotherapy is required. Dendritic cells (DCs)are not only the bridge linking innate and adaptive immunity,but also the key determinants of the quality of adaptive immunity:immunity versus immune tolerance. Therefore,the safe and efficient DC-based tumor-specific and broad-spectral tumor vaccine has an irreplaceable important position in tumor immunotherapy. Because of the high heterogeneity of DCs, the research on DC-based tumor vaccine has encountered a bottleneck. Here,we reviewed the progress in research on DC-based tumor vaccine and related problems needed to be resolved with the incorporation of our experiences.

Abstract:Dendritic cell which is found as the most powerful professional antigen presenting cell (APC) at present,can activate resting T cell and make it proliferate,induce the production of antigen-specific cytotoxic T lymphocytes( CTL),thus it plays an antineoplastic immune effect.In recent years,in tumor vaccine researcher,People take a variety of strategies to prepare the DC tumor vaccine which can clear tumor cells by changing the immune system to the immune status of the tumor and strengthening active immunity against tumor.This will almost no damage to normal tissues.So it would bring prospects for clinical application.Now,several DC tumor vaccines are reviewed in this paper.

Abstract:Objective To explore the anti-tumor effect of dendritic cells (DC) vaccine and new immunotherapy method. Methods Cytokine interleukin-18 (IL-18) an melanoma-associated antigen gp100 were transfected into bone marrow-derived DC (IL-18/gp100-DC), which were used as vaccine to induce the protective in a B16 melanoma model. And the effect of DC vaccine were detected through ELISA, RT-PCR, FACS and MLR. Results DC vaccines were successful constructed. The effect of anti-tumor immunity in IL-18/gp100-DC was significant higher than that of IL-18-DC or gp100-DC. Conclusion The effect of anati-tumor was significanly enhanced with DC vaccine co-expressing Th1 cytokine IL-18 and gp100.

Abstract:As the most potent professional antigen presenting cells, dendritic cells (DCs) have the ability to activate both naive CD4 and CD8 T cells. Recognized for their exceptional ability to cross-present exogenous antigens to prime naive antigen-specific CD8 T cells, DCs play a critical role in generating CD8 T cell immunity, as well as mediating CD8 T cell tolerance to tumor antigens. Despite the ability to potentiate host CD8 T cell-mediated anti-tumor immunity, current DC-based cancer vaccines have not yet achieved the promised success clinically with the exception of FDA-approved Provenge. Interestingly, recent studies have shown that type 1 conventional DCs (cDC1s) play a critical role in cross-priming tumor-specific CD8 T cells and determining the anti-tumor efficacy of cancer immunotherapies including immune checkpoint blockade (ICB). Together with promising clinical results in neoantigen-based cancer vaccines, there is a great need for DC-based vaccines to be further developed and refined either as monotherapies or in combination with other immunotherapies. In this review, we will present a brief review of DC development and function, discuss recent progress, and provide a perspective on future directions to realize the promising potential of DC-based cancer vaccines.

Abstract:Dendritic cell(DC) play a key role in systemic immune response against tumor for their ability to initiate primary immune response. The infiltration of DC with different phenotype in head and neck squamous cell carcinoma(HNSCC) patients correlates to tumor prognosis closely. This review will discuss the correlation of tumor prognosis with different subtypes of DC and their infiltration in tumor tissue. Followed by the decreased amount,abnormal maturity and dysfunction of DC in HNSCC patients,including DC in tumor tissue,lymph node and peripheral blood. And then discuss reasons for DC dysfunction and the present research on dendritic cell vaccine.

Abstract:Abstract As crucial antigen presenting cells, dendritic cells (DCs) play a vital role in tumor immunotherapy. Taking into account the many recent advances in DC biology, we discuss how DCs (1) recognize pathogenic antigens with pattern recognition receptors through specific phagocytosis and through non-specific micropinocytosis, (2) process antigens into small peptides with proper sizes and sequences, and (3) present MHC-peptides to CD4 + and CD8 + T cells to initiate immune responses against invading microbes and aberrant host cells. During anti-tumor immune responses, DC-derived exosomes were discovered to participate in antigen presentation. T cell microvillar dynamics and TCR conformational changes were demonstrated upon DC antigen presentation. Caspase-11-driven hyperactive DCs were recently reported to convert effectors into memory T cells. DCs were also reported to crosstalk with NK cells. Additionally, DCs are the most important sentinel cells for immune surveillance in the tumor microenvironment. Alongside DC biology, we review the latest developments for DC-based tumor immunotherapy in preclinical studies and clinical trials. Personalized DC vaccine-induced T cell immunity, which targets tumor-specific antigens, has been demonstrated to be a promising form of tumor immunotherapy in patients with melanoma. Importantly, allogeneic-IgG-loaded and HLA-restricted neoantigen DC vaccines were discovered to have robust anti-tumor effects in mice. Our comprehensive review of DC biology and its role in tumor immunotherapy aids in the understanding of DCs as the mentors of T cells and as novel tumor immunotherapy cells with immense potential.

Abstract:With the mass amplification of dendritic cells (DC) in vitro and the fast development of fusion technique, dendritic/tumor fusion cell-based vaccine has been a focus of tumor immunotherapy worldwide.In the presence of costimulatory signals, fusions of DCs and tumor cells can process and present tumor associated antigens(TAAs) and tumor specific antigens(TSAs), and subsequently induce specific antitumor cytotoxic T Lymphocytes(CTL) and lead to the inversion of tolerance induction. This strategy might be developed into a new way of tumor therapy.

Abstract:Dendritic cells are well known as the strongest antigen-presentation cells at present, which exert the effects of capturing, processing and presenting antigen, as well as sensitizing naive T cells. In vitro,cultureted dendritic cells stimulated by tumor antigen could induce the generation of cytotoxic T lymphocytes and lead to tumor-specific killing. Based on the finding, dendritic cells vaccine has been developed and some important application progress has been gained in immunotherapy for gynecological cancers at present. Therefore,more attention is focused on dendritic cells vaccine for gynecological cancers.

Abstract:Dendritic cell (DC) vaccines play an important role in anti-tumor immunotherapy. Tumor-associated cells or cytokines in the tumor microenvironment (TME) can inhibit the antigen-presenting function of DC. Immunogenic cell death (ICD) can enhance the uptake and presentation of tumor antigens by DC. This study investigates the maturation mechanism of DC induced by low-temperature plasma (LTP), as well as the therapeutic and protective effects of LTP-induced DC vaccine in a tumor model. DC2.4 that is co-cultured with LTP-treated B16F10 (LTP-B16) or with these supernatants exhibited decreased phagocytic activity, increased production of cytokines (IL-12, IL-6, TNF-alpha, and IL-1 beta), and increased expression of cell surface activation markers (CD80, CD86, and MHC II). The expression of CD80+/CD86+ is decreased after pre-treatment with TLR4 and NF-kappa B (p65) inhibitors, respectively. In vivo, trials indicated that the LTP-induced DC vaccine-induced anti-tumor immunity and, when combined with cisplatin, synergistically reduced tumor growth. Abstract:Dendritic- cells (DCs) have received considerable attention as potential targets for the development of anticancer vaccines. DC-based anticancer vaccination relies on patient-derived DCs pulsed with a source of tumor-associated antigens (TAAs) in the context of standardized maturation-cocktails, followed by their reinfusion. Extensive evidence has confirmed that DC-based vaccines can generate TAA-specific, cytotoxic T cells. Nonetheless, clinical efficacy of DC-based vaccines remains suboptimal, reflecting the widespread immunosuppression within tumors. Thus, clinical interest is being refocused on DC-based vaccines as combinatorial partners for T cell-targeting immunotherapies. Here, we summarize the most recent preclinical/clinical development of anticancer DC vaccination and discuss future perspectives for DC-based vaccines in immuno-oncology.

Abstract:Dendritic cell (DC) vaccination in cancer patients aims to induce or augment an effective antitumor immune response against tumor antigens and was first explored in a clinical trial in the 1990s. More than two decades later, numerous clinical trials have been performed or are ongoing with a wide variety of DC subsets, culture protocols, and treatment regimens. The safety of DC vaccination and its ability to induce antitumor responses have clearly been established; however, although scattered patients with long-term benefit were reported, DC vaccines have not yet fulfilled their promise, perhaps mainly due to the lack of large-scale well-conducted phase II/III trials. To allow meaningful multicenter phase III trials, the production of DC vaccines should be standardized between centers which is now becoming feasible. To improve the efficacy of DC-based immunotherapy, it could be combined with other treatments.

Abstract:As the most potent antigen-presenting cells (APC),dendritic cells are important in launching both humoral and celluar immune responses against tumor. Although the high evaluation of DC in immnunotherapy for cancer by means of DC vaccines,more studies have indicated DC is a heterogeneous population and proved that DC subsets are prominent determinants for the effectiveness of immune responses. Different DC subsets display different receptors and surface molecules,and express different sets of cytokines/chemokines,which result in distinct immunological outcomes. Clinical trials with ex vivo generated DC vaccines also manifest unexpected immunological tolerance as well as allegic response. It is essential to study the biological aspects of human DC subsets,which may be a key to the generation of novel DC-based vaccines. In this article,the progress of studies on biology of dendritic cells including their origins,differentiation,function and application of DC subsets is reviewed.

Abstract:Dendritic cells (DCs) are the most potent antigen-presenting cells, and tumor antigen-loaded DCs (DC-vaccines) can activate tumor-specific cytotoxic T lymphocytes (CTLs) in lymphatic tissues. DC vaccination is a newly emerging and potent form of cancer immunotherapy and has clinically relevant mechanisms of action with great potential for the systemic treatment of cancers. However, clinical trials have demonstrated relatively poor therapeutic efficacy. The efficacy of DC-vaccines is strongly influenced by various techniques for the priming antigen loading onto DCs and their ability to migrate to the draining lymph nodes (LNs). Therefore, it is critical to improve DC-vaccines homing to draining LNs after administration in order to optimize DC-based therapy for individual patients. This review underlines 1) appropriate strategy to load tumor antigens onto DCs and 2) to optimize vaccine administration methods to ensure loaded DCs can migrate to LNs, in particular, Intraperitoneal (IP) injection. IP injection of DC-based vaccine may be a potential regimen for gastrointestinal tumors including hepatocellular carcinoma (HCC) and pancreatic adenocarcinoma (PDAC) since huge populations of LNs are present throughout the gastrointestinal track. Which might improve the subsequent migration to LNs.
Dendritic cell-based cancer vaccines:Current status of global clinical trials

Abstract:Dendritic cell(DC)-based cancer vaccines have been studied for several decades and a line of clinical trials has been completed or in process since Ralph M.Steinman,a Nobel Prize winner in Physiology or Medicine 2011,found DCs and their crucial role in adaptive immunity.Although three DC-based cancer vaccines(Sipuleucel-T,CreaVax RCC and Hybricell) have been so far approved for marketing,the DC-based immunotherapy has not yet been approved as a standard treatment for cancer.In order to make domestic peers comprehensively understood the current status of global clinical trials of DC vaccines in cancer,the general situation(regional and country distribution,involved cancer type,start year and clinical trial phase) of these clinical trials is introduced and 43 completed and published clinical trials(selecting subject,DC culturing method,vaccination schedule,effectiveness assessment method and trial result) are summarized on the basis of clinical trial registry websites accepted by International Committee of Medical Journal Editors(ICMJE) and PubMed web databases.Then the current development trend and existing problems of clinical trials using DC-based cancer vaccines are emphatically analyzed.Finally,several proposals to improve the clinical studies with DC vaccines against cancer are put forward as follows: perfection and refinement of our regulatory policy on clinical trials of DC-based cancer vaccines;close concern of international "in vivo DC targeting" strategy;grasping to establish DC culturing method,vaccination schedule and effect assessment standard;strengthening quality monitoring of DC-based cancer vaccines;attention to basic research and clinical trial registry of DC-based vaccines for cancer;improving the clinical trial protocols of DC-based cancer vaccines;and careful selection of subjects,effect assessment time points and synchronous application of immune monitoring during cancer treatment.The authors wish to arouse domestic peers' attention and stimulate discussion between them,and these problems will be studied and addressed as possible as we can in the future.
