Effect of Nonylphenol on the Apoptosis of Rat Stertoli Cells in Vitro

Abstract:Objective To study the effect of nonylphenol(NP)on the apoptosis of rat stertoli cells in vitro Methods Sertoli cells were successfully separated,purified and cultured from 28-D SD rats The effect of NP added was tested at 0 2,1,3,5 μg/L for 24,48 and 72 h intervals Then the vitality and apoptosis of sertoli cells were determined by MTT and Flow Cytometry Semi- quantitation RT-PCR was used to detect the changes of mRNA expression of Fas and Fasl in the sertoli-germ cells Results NP induced a concentration and time dependent decrease in sertoli cell vitality Apoptosis of sertoli cells significantly increased with increasing NP concentration Both Fas and FasL were expressed with relatively high expression levels in NP exposure groups Conclusions The apoptosis of sertoli cells may be the major reason for the adverse effects of NP on spermatogenesis in mammals
