Effect of Sertraline on Behavior and Expression of NR1 in Hippocampus of Post-Myocardial Infarction Depression Rats

Abstract:Objective:To observe the effect of sertraline on behavior in vivo in a rat model of post-MI depression,and to investigate the underlying molecular mechanism.Methods:Eighty SpragueDawley rats(150-200g)were randomly assigned to five groups:normal control rats(control), myocardial ischemia rats(MI),depression rats developed by chronic unpredictable mild stress(CUMS),myocardial ischemia rats with CUMS(model),and myocardial ischemia rats with CUMS treated with intragastric administration of 5mg/kg sertraline(sertraline).Behavior measurement was done before and during experiment.Electrophysiology study in vivo and molecular biological detection were carried on after 4-week CUMS.Results:After 4-week CUMS,depression-like behaviors were observed in CUMS and model groups,and these behaviors were relieved significantly after sertraline administration(P0.05).Expression of NR1in hippocampus was reduced by 12±5%,30%±11%,and 57%±9%respectively in MI group,CUMS group,and model group,which could be increased by sertraline(17%±6%).Conclusion:Sertraline can increase the expression of NR1in hippocampus and improve depression-like behaviors in post-myocardial infarction depression rats.
