Effect of Angiotensin-Converting Enzyme Inhibitor on Vascular Cell Adhesion Molecule-1 Expression in a Rabbit Model of Early Atherosclerosis

Abstract:Objective To evaluate the effect of angiotensin converting enzyme inhibitor fosinopril on the expression of vascular cell adhesion molecule 1(VCAM 1) in a rabbit model of high cholesterol diet induced atherosclerosis Methods Thirty two rabbits were divided into three groups: normal diet group, high cholesterol group, and fosinopril group (5 mg·kg -1 ·d -1 ) The latter two groups were fed with 1% cholesterol diet for 12 weeks to establish the early atherosclerosis model The plaque areas were determined by oil red O staining; blood samples were collected for cholesterol level and the susceptibility of low density lipoprotein(LDL) oxidation; the expression level of VCAM 1 mRNA in aortic tissue were evaluated by RT PCR Results There was no difference in serum cholesterol level between the two high cholesterol groups ( P 0 05),although it was much higher than the normal diet group As compared with the high cholesterol group, the rabbits in fosinopril group showed a reduced size of aortic lesion [(26±5 4)% vs (41±9 6)%, P 0 05], an increase of the lag time of LDL oxidation induced by CuSO 4 (150 min vs 240 min, P 0 001), and a down regulation of VCAM 1 mRNA expression, the ratio of VCAM 1/GAPDH mRNA was decreased (0 90±0 35 vs 1 40±0 51, P 0 05) Conclusion Chronic fosinopril ameliorates the severity of early atherosclerosis probably through the reduction of LDL oxidation and the suppression of VCAM 1 mRNA expression
