Marrow Stem Cells Seeded into Polyglycolic Acid Scaffold to Restore New Bone in Vivo

Abstract:Objective New bone restoration by marrow stem cells (MSCs) seeded into polyglycolic acid (PGA) scaffold was studied.Methods Marrow stem cells (MSCs) were cultured,expanded and induced in vitro.Cells were seeded into PGA non-woven mesh at the density of 5×10 7/ml in 50μl cell suspension.Cell growth on the scaffold was observed by phase-contrast microscope and scanning electronic microscope.After 5 days in vitro incubation,four cell-polymer complexes were implanted subcutaneously into nude mice back.Two months after operation,bone formation was observed by gross inspection and histological observation.Results MSCs adhered and spreaded well on the surface of PGA scaffold,observed by phase contrast microscope and scanning electronic microscope.Hard tissue was restored successfully subcutaneously in nude mice back 2 months after implantation.And histological examination demonstrated that newly restored tissue was constituted of bone.Conclusion It is an efficient method to restore new bone in extraskeletal site by seeding MSCs into PGA scaffold.
