Abstract:A superoxide dismutase (SOD) was purified from seeds of the Chinese black soybean(Glycine max cv). The purification procedure comprised ammonium sulfate precipitation, ion-exchange chromatography on CM-Sephadex C-50, affinity chromatography on Affi-gel blue gel and HP LC on DEAE 650-C. The final enzyme was purified 15.40 fold with a specific activity of 4619.3U/mg protein and a recovery of enzyme was 29.41% from (NH4)2SO4 precipitation. The SDS-PAGE of the purified protein exhibited a molecular mass of 31.0kDa and could be depolymerized spontaneously to a band at about 14.5kDa in non-reducing conditions, and a diffusion band at about 15.5kDa in reducing conditions, indicating a double-strand protein with intra-molecular disulfide bridges. The isoelectric point was determined to be 5.6, indicating it's an acidic protein. The enzyme is a type of copper plus zinc superoxide dismutase (Cu, Zn-SOD) because it was greatly inhibited by hydrogen peroxide, insensitive to chloroform–ethanol, and the maximum ultraviolet absorbance was found to be 266nm. Thermostability experiments showed that the activity of the purified enzyme was stable after exposure to temperatures below 50°C, and the activity could be well maintained between pH 6.0 and 8.0. Moreover, the SOD exerted an obvious protective effect against oxidative stress of UVC radiation in normal human liver cell strain L-02 cells.

Abstract:Cordyceps militaris mycelium produced mainly Cu, Zn containing superoxide dismutase (Cu, Zn-SOD). Cu, Zn-SOD activity was detectable in the culture filtrates, and intracellular Cu, Zn-SOD activity as a proportion protein was highest in early log phase culture. The effects of Cu2+, Zn2+, Mn2+ and Fe2+ on enzyme biosynthesis were studied. The Cu, Zn-SOD was isolated and purified to homogeneity from C. militaris mycelium and partially characterized. The purification was performed through four steps: (NH4)2SO4 precipitation, DEAE-sepharose™ fast flow anion-exchange chromatography, CM-650 cation-exchange chromatography, and Sephadex G-100 gel filtration chromatography. The purified enzyme had a molecular weight of 35070±400Da and consisted of two equal-sized subunits each having a Cu and Zn element. Isoelectric point value of 7.0 was obtained for the purified enzyme. The N-terminal amino acid sequence of the purified enzyme was determined for 12 amino acid residues and the sequences was compared with other Cu, Zn-SODs. The optimum pH of the purified enzyme was obtained to be 8.2–8.8. The purified enzyme remained stable at pH 5.8–9.8, 25°C and up to 50°C at pH 7.8 for 1.5h incubation. The purified enzyme was sensitive to H2O2, KCN. 2.5mM NaN3, PMSF, Triton X-100, β-mercaptoethanol and DTT showed no significant inhibition effect on the purified enzyme within 5h incubation period.

Abstract:A simple and cost-effective way to purify superoxide dismutase (SOD) from garlic was proposed. The stepwise isolation and purification procedure consists of phosphate buffer and chloroform–ethanol extraction, acetone precipitation followed by ion exchange chromatography on DEAE column. The specific enzyme activity was 4124U/mg protein with the recovery rate of 53.3% and concentration factor of 192, respectively. SDS-PAGE analysis showed high consistency of garlic SOD with cattle blood SOD. The enzyme activity of garlic SOD was greatly inhibited by cyanide and hydrogen peroxide while it could be well maintained at pH values ranging from 4 to 11. Only slight decrease of enzyme activity was identified at temperatures lower than 50°C or by treatment with ultrasonic wave for 30min. The ultraviolet absorbance of garlic SOD was found to be 269nm.

Abstract:Superoxide dismutases (SODs) were purified from sea buckthorn and chestnut rose by ammonium sulfate precipitation and anion-exchange chromatography, and the detection methods of water-soluble tetrazolium-1 (WST-1), nitrobluetetrazolium (NBT) and pyrogallol autoxidation (PA) for SOD activity were compared. WST-1 method was selected due to its coefficient of variation (CV) <6% in this study. Two SODs exhibited similar characteristics. Their molecular mass and isoelectric point were about 30 kDa and 4.8 to 5.0 estimated by electrophoresis, and the K-m was 0.05 to 0.08 mmol/L, respectively. Dynamic light scattering analysis suggested their hydrodynamic radius distributes from 60 to 1500 nm. The activity of two SODs was unchanged at <80 degrees C or pH 2 to 9 or in simulated human gastric fluid. Their circular dichroism spectra suggested a main beta-sheet structure, the fluorescence spectra reflected that the tryptophan residues of two SODs is partially exposed, these structures were rather stable at pH 2 to 9 or 50 to 90 degrees C. Practical Application Superoxide dismutase (SOD) is an important antioxidant enzyme. SODs from sea buckthorn and chestnut rose were stable at high temperature or low pH or simulated gastric fluid. This result can provide a new approach for the potential application of SOD in the food and pharmaceutical fields.

Abstract:OBJECTIVE To prepare Cu,Zn containing superoxide dismutase from Cordyceps militaris Mycelia.METHODS The homogenization of mycelia was prepared by sonicator,(NH_(4))_(2)SO_(4) precipitation,ion-exchange chromatography.RESULTS The optimized sonic power and time were 400 W and 14 min for 10 g wet weight mycelium,respectively.The purification was performed through three steps:(NH_(4))_(2)SO_(4) precipitation(55%～99% saturation),DEAE-sepharose~(TM) fast flow anion-exchange chromatography,CM-52 cation-exchange chromatography.After this procedure,a specific activity of 13 759.1 U·mg(pro)~(-1) was reached,corresponding to 214.1 fold purification with a yield of 22.4%.The purity was homogeneous by SDS-PAGE analysis.The purified enzyme was sensitive to H_(2)O_(2) and KCN.The maximum absorption was observed at 266 nm.The amino acid compositions were similar to that of other sources Cu and Zn-SOD.CONCLUSION The method established in this study is effective and feasible,the homogeneous protein is obtained by SDS-PAGE analysis.

Abstract:After fermenting, SOD from Bacillus cerus was dpurified by a stepwise procedure consisting of (NH_(4))_(2)SO_(4) fractionation, sephades G-75 gel filtration and DEAE-52-cellulose chromatography. This procedure resulted in a single band on sodium dodecyl sulfte-polyacrylamide gels indicating that the prearation was homogeneous. The subunit molecular weight of this protein was estimated at 17783 daltons. The nature analysis by the spectrum of ultraviolet and the inhibition of cyanide, H_(2)O_(2) and chloroform-ethanol indicated that the pure preparation was Fe-SOD. The relative activity wa s1753.8u.

Abstract:Superoxide dismutases ( SODs) are one family of important antioxidant enzymes involved in scavenging superox-ide anion free radical in organisms. In the present paper, the expression primer was designed by Cu/Zn-SOD gene se-quence of Siniperca chuatsi ( named as ScCu/Zn-SOD, accession numbers: KJ558392. 1 ) . A truncated signal peptide sequence with 460 bp was amplified. After identification of the sequence, the constructed recombinant expression plasmid (pET-30a+ScCu/Zn-SOD) was transformed into BL21(DE3), and was expressed by induction with IPTG. The solu-ble recombinant protein of ScCu/Zn-SOD ( designated as rScCu/Zn-SOD) was obtained by optimized expression condi-tion, and the concentration and activity of rScCu/Zn-SOD was measured after the purification of recombinant protein. The results showed that the expression of ScCu/Zn-SOD could be induced under 20℃ and 37℃ conditions. The rScCu/Zn-SOD was mainly aggregated to form inclusion bodies at 37 ℃. The expression quantity of rScCu/Zn-SOD could be im-proved by reduction induction temperature and supplement Cu2+ /Zn2+. The addition of 0. 5 mmol/L CuSO4 and 0. 1 mmol/L ZnCl2 in culture medium was optimal under 20 ℃ and 0. 5 mmol/L IPTG condition, and the expression quantity of rSc-Cu/Zn-SOD was obviously increased. The results can provide basic data for the research on functional characterization of SOD protein.

Abstract:We report cDNA cloning, expression, purification, and characterization of a novel Cu/ Zn superoxide dismutase (SOD) from Jatropha curcas leaves. The full-length cDNA of this SOD contained a 496-bp open-reading frame (ORF) encoding 162 amino acid residues. The recombinant plasmid containing the SOD coding sequence was introduced into Escherichia coli, and the SOD was expressed as a fusion protein. The recombinant SOD was purified from a high-density fed-batch culture using a combination of immobilized metal ion affinity chromatography (IMAC) and Sephadex G25 desalting chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis indicated that the recombinant SOD was a monomeric protein with a molecular mass of approximately 16.4 kDa. Isoelectric focusing showed that this SOD was a basic protein with pI values of 7.04, 7.33, 8.62, and 8.77. The activity of the SOD was stable at 70 degrees C for 10 min, and in a broad pH range from 4 to 9. The presence of urea (up to 8 M), guanidinium chloride (up to 6 M), and 2-mercaptoethanol (up to 8 mM) had little effect on the activity. The activity decreased gradually with increasing concentrations of imidazole, hydrogen peroxide, and ethylenediaminetetraacetic acid (EDTA). Atomic absorption spectrometry showed the presence of 0.239 copper and 0.258 zinc atoms, respectively, in the SOD polypeptide.

Abstract:The cDNA of Cu, Zn containing superoxide dismutase from the Cordyceps militaris SH (cm-SOD) was overexpressed in Escherichia coli BL 21 (DE3) using the pET-21a expression vector. The recombinant cell overexpressed the protein corresponding to 35±3% of total bacterial protein in cytosol. The purification was performed through three steps: DEAE-FF, CM-52, and G-100. After this purification procedure, a specific activity of 27272.7U/mg of protein was reached, corresponding to 6.1-fold purification with a yield of 85.0%. The purity was homogeneous by SDS–PAGE analysis and 94.2±1.0% by CZE analysis. A subunit molecular mass of the recombinant enzyme was 15704Da with a Cu and Zn element. In addition, the dimeric and polymeric structures were observed on MALDI-TOF-MS. Isoelectric point value of 7.0 was obtained for the recombinant enzyme that was sensitive to H2O2 and KCN. The recombinant enzyme remained 80±2% residual activity at pH 7.8, at 50°C for 4h incubation. The properties: N-terminal amino acid sequence (the first 12 amino acid residues), pI, subunit molecular mass, thermo-stability of the purified recombinant SOD are similar to that of the native Cu, Zn-SOD from C. militaris (N-cm-SOD).

Abstract:At present, deep-sea enzymes are a research hotspot. In this study, a novel copper–zinc superoxide dismutase (CuZnSOD) was successfully cloned and characterized from a new species of sea cucumber Psychropotes verruciaudatus (PVCuZnSOD). The relative molecular weight of the PVCuZnSOD monomer is 15 kDa. The optimum temperature of PVCuZnSOD is 20 °C, and it maintains high activity in the range of 0–60 °C. It also has high thermal stability when incubated at 37 °C. PVCuZnSOD has a maximum activity of more than 50% in the pH range of 4–11 and a high activity at pH 11. In addition, PVCuZnSOD has strong tolerance to Ni2+, Mg2+, Ba2+, and Ca2+, and it can withstand chemical reagents, such as Tween20, TritonX-100, ethanol, glycerol, isopropanol, DMSO, urea, and GuHCl. PVCuZnSOD also shows great stability to gastrointestinal fluid compared with bovine SOD. These characteristics show that PVCuZnSOD has great application potential in medicine, food, and other products.

Abstract:A superoxide dismutase (SOD) with the molecular weight of 31,079 has been purified as a homodimer from Panax ginseng by employing neutral pH buffer extraction, ammonium sulfate precipitation, isoelectric point precipitation and ion exchange methods. The enzyme's specific activity determined by an improved Marklund method was 9480.43 U/mg. Metal analysis showed that the SOD contained iron with the stoichiometry of 0.9 ± 0.3 Fe/subunit and exhibited high thermal stability (70 °C) over the pH range from 4.0 to 9.0. Its maximum absorption wavelength was 278 nm and it was sensitive to hydrogen peroxide, trichloromethane-ethanol and urea. These results indicate that the enzyme is an iron SOD.

Abstract:The objective of this work is to develop a membrane-based cost-effective process for the separation and purification of superoxide dismutase (SOD) from garlic. It was found that a two-stage ultrafiltration process using a 100kDa MWCO regenerated cellulose membrane and a 50kDa MWCO polyethersulfone membrane could successfully be employed to isolate SOD by removing the bigger macromolecules and smaller molecules from the crude feedstock. The effects of solution pH and ionic strength on the transmission of SOD and the presence of impurity proteins with molecular weights close to SOD were quantified using the pulsed sample injection ultrafiltration technique. The effect of other parameters including system hydrodynamics and mass transfer, in terms of stirring speed, and permeate flux on the separation of SOD was also studied using parameter scanning ultrafiltration. Under optimized conditions, the purity of SOD obtained was greater than 98.0% after two-stage ultrafiltration process and the recovery of SOD from the feedstock was close to 90%. The resulting SOD product was analyzed by isoelectric focusing (IEF), SDS-PAGE and circular dichroism (CD), to confirm its isoelectric point, molecular weight and molecular secondary structure of the garlic SOD. The purity and activity as well as effects of metal ions (copper and zinc ions) on SOD specific activities were experimentally determined.

Separation, purification and properties of superoxide dismutase from germination of mung bean

Abstract:The different periods of germination of mung bean was used to prepare the crude SOD enzyme extract,and the highest specific activity during the period of crude enzyme solution was screened,then the solution was separated and purified by the thermal denaturation,ammonium sulfate fractionation,DEAE-Sepharose FF anion-exchange chromatography and SephacrylS 100 molecular sieve chromatography,finally the physical and chemical properties of the pure SOD enzyme were studied.The results showed the maximum specific activity of SOD on the seed germination 3 days of mung bean,and the purified SOD enzyme specific activity was 2256.4 U/mg(purification factor 3.9).As the structure of SOD in Mung bean,Cu/Zn SOD and MnSOD have a molecular weight of 68KD,high thermal stability,stability in the pH 5-8,and resistant to high concentrations of SDS,and its activity was able to inhibited by EDTA and completely inhibited by H2O2.

Abstract:PPO was purified from Cistanche deserticola, and its enzymatic characteristics were clarified. It was found that microwave treatment was an efficient way to inactivate PPO. Polyphenol oxidase (PPO) from Cistanche deserticola was obtained and purified through an acetone precipitation and anion exchange column, the enzymatic characteristics and inactivation kinetics of PPO were studied. The specific activity of PPO was 73135.15 ± 6625.7 U/mg after purification, the purification multiple was 48.91 ± 4.43 times, and the recovery was 30.96 ± 0.27
