The Supernatant of Bone Marrow from SLE Patients Promotes the Senescence of Mesenchymal Stem Cells

Abstract:Patients with systemic lupus erythematosus(SLE)showed over activation of many immune cells,which can be modulated by mesenchymal stem cells(MSCs),so whether MSCs can effectively regulate immune response is critical for the autoimmune disease.MSCs from SLE patients have been demonstrated to be enhanced senescence and impaired immune modulation.The objective of this study is to explore whether local environment of bone marrow affects the senescence of MSCs.Umbilical cord derived MSCs were cultured in DMEM/F12 containing 10% bone marrow supernatant of SLE patients or controls.MSCs were stained with SA-β-gal,and the percentages of SA-β-gal positive cells were evaluated.The mRNA levels of p53 and p21were detected by Real time PCR.The protein levels of p53/p21 were examined by Western blot.DNA damage response was detected by immunofluorescence analysis.Cell proliferation and apoptosis were evaluated by CCK8 assay and flow cytometry.The results demonstrated that MSCs in bone marrow supernatant from SLE patients showed more frequencies of SA-β-gal positive cells,as well as higher levels of p53/p21 than that of healthy controls.SLE supernatant also significantly elevated the levels of 53BP1 in cell nuclei.In addition,SLE supernatant inhibited the capacity of MSC proliferation but induced cell apoptosis.Therefore,the supernatant of bone marrow in SLE patients can promote the senescence of MSCs via accelerating DNA damage response.
