Influence of Methionine-depletion and Its Combination with Chemotherapy in an Experimentation on Human Primary Gastric Cancer Cells

Abstract:Objective: To elucidate whether human primary gastric cancer and gastric mucosa epithelial cells can grow normally in a methionine (Met)-depleted environment, ie. Met-dependence, and whether such status can enhance the killing effect of chemotherapeutic agents. Methods: Fresh human gastric cancer and mucosal tissues were processed to form monocellular suspensions, which were then cultured in the Met-free but homocysteine-containing (Met -Hcy +) medium, or jointly applied to different chemotherapeutical drugs. The proliferation of the cells was examined by cell counter, flow cytometry (FCM) and microcytotoxicity assay (MTT). Results: ① The growth of human primary gastric cancer cells in Met -Hcy + was suppressed, manifested by the decrease of total cell counts, the decline in the percentage of G 0G 1 phase cells and the increase of S phase cells; however, the growth of gastric mucosal cells was not disturbed. ② If Met -Hcy + medium was used in combination with chemotherapeutical drugs, the number of surviving gastric cancer cells dropped significantly. Conclusions: ①Human primary gastric cancer cells are Met-dependent in vitro; yet, gastric mucosal cells have not shown the same characteristics. ② Met -Hcy + environment may enhance the killing effect of chemotherapy on human primary gastric cancer cells.
