Study on the Expression of Sex Hormone-Binding Globulin Mrna in Tissue of Uterine Leiomyoma

Abstract:Objective To investigate the expression of sex hormone-binding globulin (SHBG) mRNA and the relationship between SHBG and uterine leiomyoma.Methods 30 samples of uterines by hysteromyomectomy were selected in the study. The RT-PCR and Southern blot were used to detect the expression of SHBG mRNA in tissues of uterine leiomyoma and normal myometrium.Results There was no significant difference in SHBG wild-type (SHBGw) mRNA between the tissues of uterine leiomyoma and normal myometrium(P0.05), however, the SHBG exon VII splicing variant (SHBGv) levels in uterine leiomyoma was significantly reduced,as compared with those in normal myometrium (P0.05).There was no obvious relationship between the levels of SHBG mRNA and the menstrual phase (P0.05).Conclusion The expressions of both SHBGv and SHBGw can be found in both the tissues of uterine leiomyoma and normal myometrium.SHBGw may not be related to the high concentration of hormone in local myometrium,however, the low levels of SHBGv may be ralated to the genesis of uterine leiomyoma.
