Analysis of the polymorphisms of apoE gene and ACE gene in Alzheimer's disease and vascular dementia

Abstract:Objective To explore the relations between the polymorphisms of apolipoprotein E(apoE) gene and ACE gene and Alzheimer's disease(AD) and vascular dementia(VaD).Methods The polymorphisms of ACE gene and apoE gene were detected by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) technique in 26 patients with late-onset AD,54 patients with VaD and 68 normal controls. Results The frequencies of allele e2,e3 and e4 of apoE gene were 0.077,0.615 and 0.308 respectively in AD patients and 0.056,0.685 and 0.259 respectively in VaD patients. The frequency of ACE genotype DD increased significantly in patients with AD and VaD as compared with the controls.The frequency of allele D increased significantly in patients with AD as compared with the normal control.Conclusions The results indicated that ACE genotype DD and allele e4 of apoE gene might increase the risk of the AD and VaD.It is significant to monitor and intervene the risk factors of dementia for decreasing the incidence.
