Abstract:The activation of hydroxyl group is a key step in the synthesis of ligand-polymer for affinity partition. Using thionyl chloride as the chloridating agent and polyethylene glycol (PEG) as the model polymer, the conventional process was improved by introducing triethylamine(Et 3N) into the reaction system.The precipitate of Et 3N·HCl formed in the system promotes the reaction into a thermodynamically beneficial process, which significantly reduced the dosage of thionyl chloride and simplified the synthesis procedure.The mole ratio of reaction materials(SOCl 2,Et 3N and PEG),reaction temperature and reaction time were investigated and optimized.A high value of the synthetic yield was reached at 1.54 mol Cl ·(mol PEG) -1 .The comparisons to the conventional thionyl chloride method and epichlorohydrin method were also discussed.

Abstract:Acyl chloride is an important functional group in acylation reactions, though its compounds are difficult to store and transport due to their high reactivity. In this research, phenanthro[9,10-d]imidazole is introduced into an acyl chloride agent of TPEPCl to enhance the stability via the delocalization effect. Meanwhile, rarely reported acyl chloride crystals have been achieved which could further prove its stability. The tetraphenylethylene group is also employed to visually monitor the changes of TPEPCl via fluorescent variation with the help of aggregation-induced emission (AIE) characteristics. The reactivity of TPEPCl could be stimulated by a strong alkaline solution or high temperature. Crystallography analyses of acyl TPE-PI derivatives are also discussed and the relationship between the delocalization structure and stability is determined. These findings help to stabilize, control and monitor the reactivities of active functional materials for applications.

Abstract:Modifications at polypeptide side chains have been found to affect conformational and properties,which is a vital strategy to prepare functional polypeptides.This contribution describes the synthesis of homo and co-polypeptides containing unsaturated bonds and thioether moieties,poly(S-allylcysteine)(PAC).The pendant vinyl groups provide an active binding site which are able to be further modified with cysteine by thiol-ene click reaction.The sulfoxide from thioether oxidation bestows the polypeptide water solubility and enzymatic catalytic sites.The self-assembly micelles of PEG-b-PAC are ruptured along with oxidation to release the Nile red payloads.Moreover,poly(S-allyl-cysteine sulfoxide),aka.polyalliin,is the oxidation product of PAC which undergoes side chain cleavage catalyzed by alliinase enzyme.The polypeptides are biocompati-ble as confirmed by cell viability assays and may find applications in drug delivery,biosensing and nanoreactor.

Abstract:A new efficient synthesis of (2S,3R)-3-hydroxy-3-methylproline (3) is reported. During the course of a recent study on the Lewis acid promoted intramolecular opening of an epoxide by a carbamate NH, a highly concerted rearrangement was unexpectedly observed. Further investigations of substrate generality show that delta-carbamate-alpha,beta-epoxide esters commonly underwent similar rearrangements with the aid of Lewis acids. Retrosynthetic analysis of such a C-2-N disconnection can lead to an efficient synthesis of (2S,3R)-3-hydroxy-3-methylproline (3) in high enantio purity. Stereochemistries were established by a Sharpless asymmetric dihydroxylation and a diastereoselective reductive amination.

Abstract:The introduction of a 4-mercaptan substituent was recently reported to be effective to improve the reactivity of C-terminal prolyl thioesters in native chemical ligation (NCL). To elucidate the origin of the high reactivity of 4-mercaptoprolyl thioesters, a theoretical study was performed on the transthioesterification of NCL herein with the aid of density functional theory (DFT) methods. The calculation results support a transthioesterification mechanism involving two stages. The first stage is the intramolecular transthioesterification of 4-mercaptoprolyl thioesters to form a bicyclic thiolactone intermediate and the second stage is the intermolecular transthioesterification of the thiolactone intermediate with N-terminal cysteine. The two stages proceed both via nucleophilic attack of the thiolate and proton-donor-assisted thiolate release. The thiolate release in the first stage is the rate-determining step of the whole transthioesterification of 4-mercaptoprolyl thioesters. Based on the mechanistic details, further analysis was conducted to gain a deeper insight with natural bond orbital (NBO) analysis, non-covalent interaction plots (NCIplots), distortion analysis, etc. The n -> pi* interaction between the N-carbonyl oxygen and thioester carbonyl was found to be detrimental to the first stage while a previously unnoticed n -> pi* interaction between the amide nitrogen and the thiolactone carbonyl slightly benefits the second stage. The entropy effect was the main driving force for the first stage because this stage is an intramolecular one-to-two process overall. On the other hand, the release of the ring stain precluded in the bicyclic thiolactone intermediate is mainly responsible for the high reactivity of the thiolactone in the second stage compared with other prolyl thioesters. Meanwhile, the intramolecular hydrogen bond between the thiolactone carbonyl oxygen and the protonated amine of N-terminal cysteine also contributes to the transthioesterification.

Abstract:The copolymers of N-isopropylacrylamide (NIPAM) and cholesteryl acrylate (CHA) in different molar ratios were synthesized and characterized. The surface activity and micellar behavior of the copolymers in aqueous solution were studied by surface tension method and fluorescence probe method. The Lower Critical Solution Temperature(LCST) of the copolymers was determined by fluorescence probe and turbidity measurement method. Results showed that the incorporation of hydrophobic comonomer CHA led to a lower LCST than PNIPAM and the LCST decreased with the increase of the ratio of CHA. The copolymers in aqueous solution had obvious aggregation behavior with the increase of their concentrations. It was evident that the introduction of a small amount of CHA units into the polymer backbone could lead to a very dramatic effect on the amphiphilic properties of the copolymers. The inner core of the micelles formed by self-aggregates of CHA segments of the copolymers was observed to encapsulate hydrophobic compounds, such as cholesterol (a drug model). With the increase of cholesterol amount in the sample solutions with the same polymeric concentrations, the average size of the formed micelles shifted to a larger value, which indicated that more cholesterol molecules were encapsulated in the micelles.
Orbital Interactions in Native Chemical Ligation Reaction of Proline Thioesters

Abstract:A systematic theoretical study was carried out to investigate the origin of the relatively low reactivity of peptide-prolyl-thioesters in the native chemical ligation (NCL) reaction. Mechanistic calculations were performed on the two NCL reactions of peptide-prolyl-thioester (Path-Pro) and peptide-alanyl-thioester (Path-Ala). The results show that both include three steps: intermolecular thiol-thioester exchange, transthioesterification, and a final intramolecular S -> N acyl migration. The calculations indicate that the first step is the rate determining step of both pathways. Path-Pro is kinetically disfavored, so the peptide-prolyl-thioester is found to be less reactive in NCL reaction. This conclusion is consistent with the experimental observations. Further examination of the rate determining steps of these two pathways shows that the n ->pi(star) interaction of proline N-alpha carbonyl increases the LUMO orbital energy of peptidyl-prolyl-thioester, decreases the interaction energy between proline carbonyl and the sulphur atom in aryl thiol, and finally increases the total energy barrier.

Abstract:N-acylation of l-prolinol (2) with enoyl chloride provided l-cinnamoylprolinol (3a) and l-crotonylprolinol (3b), respectively. The attachment of (3a) or (3b) to Merrifield resin and the utilization of such a system for asymmetric Michael additions of Grignard reagents to α,β-unsaturated carbonyl compounds were studied. The stereoselectivities have been increased compared with those observed for the same reactions in solution. On the other hand, it has been proved by the FT-IR difference spectrum of the Merrifield resin and polymer-supported chiral auxiliary that l-prolinol derivatives have been coupled to the resin.

Abstract:We reported that for a series of amines with different structures and functional groups, the deprotection of the dialkylphosphoryl group from the N-alkyl-N-dialkyl- phosphoramidates can be achieved under very mild conditions as compared to the corresponding sulfonyl derivatives. From the kinetic study, we find that the diisopropylphosphoramidates derivatives have substantial stability in the acid media. The O, O-diisopropyl-N-phenyl-phosphoramidates has a half life of 53 min in 6M HCl at 50℃. Also, the P-N bond persists during the Lewis acid catalyzed Freidel-Crafts acylation of N-dialkylphosphoryl-N-substituted phenethylglycine (4) to give the stable 2, 3, 4, 5-tetrahydro-7, 8-dialkoxyl-N-diisopropylphosphoryl- 3-benzazepine-1-one (5). 5 can be quantitatively transformed into the amine HCl salt under very mild condition. Thus. we have shown that diisopropyl phosphite can be used as an economical amino protection reagent, the steric hindrance of the diisopropyl group slows down the rate of acid hydrolysis, therefore, increases its stability in the acid media. This chemical property will be useful in organic synthesis.

Abstract:Prolyl thioesters have shown significantly lower reactivities in native chemical ligation (NCL) in comparison to that of the alanyl thioester. This report describes a mild and efficient internal activation protocol of peptidyl prolyl thioesters in NCL without using any thiol-based additives, where the introduction of a 4-mercaptan substituent on the C-terminal proline significantly improves the reactivity of prolyl thioesters via the formation of a bicyclic thiolactone intermediate. The kinetic data indicate that the reaction rate is comparable to that of the reported data of alanyl thioesters, and the mechanistic studies suggest that the ligation of two peptide segments proceeds through an NCL-like pathway instead of a direct aminolysis, which ensures the chemo-selectivity and compatibility of various amino acid side chains. This 4-mercaptoprolyl thioester-based protocol also allows an efficient one-pot ligation-desulfurization procedure. The utility of this method has been further demonstrated in the synthesis of a proline-rich region of Wilms tumor protein 1.

Abstract:Aim To synthesize N-((2-oxo-3-substituted-oxazolidin-5-yl)methyl)acetamide by a new way.Meth- ods The intermediate of glyeidylacetamide was prepared orderly by amination and cyclization of epichloeohydrin in the presence of protective agent of bezaldehyde.Reaction of N-substitutrd ethyl carbamate and glycidylaeetamide are carried out in the presence of catalyst to provide N-((2-oxo-3-substituted-oxazolidin-5-yl)methyl)acetamide.Re- suits Four N-((2-oxo-3-substituted-oxazolidin-5-yl)methyl)acetamides were synthesized.The total yield is 50%.The structure of the compounds was confirmed by IR,H NMR and elementary analysis.Conclusion The methods were mild and available for synthesis N-((2-oxo-3-substituted-oxazolidin-5-yl)methyl)acetamide.

Abstract:Peptides can be used as effective molecular tool for covalent modification of proteins and play important roles in ligand directed covalent modification. Tyr-selective protein modifications exert a profound impact on protein functionality. Here, we developed a general strategy that involves nucleophilic addition of alkyne for tyrosine modification. The terminal alkyne of propargyl sulfonium is motivated by the sulfonium center to react with phenolic hydroxyl. This approach provides a straightforward method for tyrosine modification due to its high yield in aqueous solution at physiological temperature. In addition, cyclic peptides could be obtained via adjusting pH to 8.0 from peptides consisting of tyrosine and methionine modified by propargyl bromide, and the resulting cyclic peptides are proved to have better stability, excellent 2-mercaptopyridine resistance and improved cellular uptakes. Furthermore, molecules made from the propargylated sulfonium have the potential to be used as warheads against tyrosine containing biomolecules. Collectively, we develop a direct and uncomplicated technique for modifying tyrosine residues, the strategy concerned can be widely utilized to construct stable peptides and biomolecules imaging.
Synthesis, Characterization, and Antifungal Property of Hydroxypropyltrimethyl Ammonium Chitosan Halogenated Acetates

Abstract:Hydroxypropyltrimethyl ammonium chitosan halogenated acetates were successfully synthesized from six different haloacetic acids and hydroxypropyltrimethyl ammonium chloride chitosan (HACC) with high substitution degree, which are hydroxypropyltrimethyl ammonium chitosan bromacetate (HACBA), hydroxypropyltrimethyl ammonium chitosan chloroacetate (HACCA), hydroxypropyltrimethyl ammonium chitosan dichloroacetate (HACDCA), hydroxypropyltrimethyl ammonium chitosan trichloroacetate (HACTCA), hydroxypropyltrimethyl ammonium chitosan difluoroacetate (HACDFA), and hydroxypropyltrimethyl ammonium chitosan trifluoroacetate (HACTFA). These chitosan derivatives were synthesized by two steps: first, the hydroxypropyltrimethyl ammonium chloride chitosan was synthesized by chitosan and 3-chloro-2-hydroxypropyltrimethyl ammonium chloride. Then, hydroxypropyltrimethyl ammonium chitosan halogenated acetates were synthesized via ion exchange. The structures of chitosan derivatives were characterized by Fourier transform infrared spectroscopy (FTIR), ¹H Nuclear magnetic resonance spectrometer (¹H NMR), 13C Nuclear magnetic resonance spectrometer (13C NMR), and elemental analysis. Their antifungal activities against Colletotrichum lagenarium, Fusarium graminearum, Botrytis cinerea, and Phomopsis asparagi were investigated by hypha measurement in vitro. The results revealed that hydroxypropyltrimethyl ammonium chitosan halogenated acetates had better antifungal activities than chitosan and HACC. In particular, the inhibitory activity decreased in the order: HACTFA > HACDFA > HACTCA > HACDCA > HACCA > HACBA > HACC > chitosan, which was consistent with the electron-withdrawing property of different halogenated acetates. This experiment provides a potential idea for the preparation of new antifungal drugs by chitosan.

Abstract:A lithocholic acid derivative having high compatibility with phospholipid was synthesized and incorporated to the phospholipid bilayer membrane. A significant influence of it on the molecular packing and the stability enhancement of the vesicle suspension were clarified by a fluorescence depolarization method and a turbidity measurement, respectively.

Abstract:Self-stratifying epoxy-acrylate composite emulsions were synthesized by core-shell emulsion polymerization using reactive emulsifiers.The structure and size of emulsion particles were characterized by transmission electron microscope and laser diffraction particle size analyzer.The cured latex films were characterized by attenuated total reflectance infrared spectroscopy and scanning electron microscopy.The stability of emulsions,as well as the pencil hardness and water resistance of their cured films were tested.A most stable emulsion containing epoxy resin as much as 40.00wt.% was obtained when the amount (with respect to the solid content of emulsion) was 7.00wt.% for emulsifier,0.36wt.% for stabilizer (hydroxypropyl methylcellulose),and 6.00-8.00wt.% for methacrylic acid (as functional monomer).Its cured film presents a stratified structure (acrylate resin on top,epoxy resin at the bottom,and the mixture of them in the middle) and is good at pencil hardness and water resistance.

Abstract:Effects of the different reaction conditions on the synthetic method of iso-propyl chloroacetate have been reviewed. Different catalysts that consist of inorganic salt like ( Ce(SO 4 ) 2 ∙4H 2 O and NaHSO 4 ) and PCl 3 have been introduced also. The reaction conditions include the reaction time, the molar ratio of chloroacetic acid to isopropanol , the amount of the catalyst and the number of reusable catalyst . The optimized reaction time, the molar ratio of chloroacetic acid to isopropanol and the amount of the catalyst are beneficial to improve the yield of iso-propyl chloroacetate.

Abstract:An improved synthetic process of captopril intermediate,D-3-acetylthio-2-methylpropanoyl-L-proline,was carried out using potassium hydroxide and potassium dihydrogen phosphate buffer as a condensating agent.Add potassium dihydrogen phosphate into a raw material of the L-proline,then,drop potassium hydroxide to form potassium hydroxide and potassium dihydrogen phosphate buffer as a condensating agent,the title compound was synthesized by dropping another raw material of 3-acetylthio-2-methyl-propanoyl chloride into above prepared solution.The pH of reaction system keeps in from 7.5 to 8.5,the reaction process is simple and efficient,and the average yield of product was improved from 33% to 39.5%.

Abstract:To improve the properties of chitosan,this research used chitosan and chloroacetic acid to synthesize a new type chitosan derivative-N,O-carboxymethyl chitosan.The experiment results(orthogonal test)showed that the optimum condition for making N,O-carboxymethyl chitosan was:reaction temperature 70℃,the mole ratio of sodium hydroxide and chloroacetic acid was 2.4,reaction time was 4h,amount of isopropyl alcohol was 50ml.Analyzed the chemical and physical properties of N,O-carboxymethyl chitosan,the result showed that the isoelectric point of N,O-CMC migrated toward the acid with the increasing degree of substitution;N,O-carboxymethyl chitosan with high degree of substitution had a better moisture content and superior moisture absorption than hyaluronic acid.In the condition of high humidity(RH81%),the hygroscopicity of N,O-CMC with 1.36 degree of substitution was 1.8 times of hyaluronic acid.
