The Relation Between Fimc Gene and Pathogenicity of Avian Escherichia Coli

Abstract:Twenty strains of avian E.coli were respectively inoculated into 1-day-old chicks to examine their pathogenicity, and fimC gene was amplified by PCR with their genome DNAs as template respectively. The result showed that 95% high pathogenic strains were fimC~+, no or low pathogenic strains were fimC~-. According to the related informations, the fimC gene almost only exist in avian pathogenic E.coli, it could be regarded as a virulent mark for identification of avian pathogenic E.coli. Amplified product of E.coli O2 strain was cloned into pMD18-T vector, the recombinant plasmids were identifid with enzymes and sequence analysis, the homogeneity of the nucleotide sequence was 97.9%, and the homogeneity of amino acid sequence was 96%, compared with the reference sequence,which conformed that the amplified product was fimC gene.
