Proliferation,Identification,and Detection of Endocytosis Ability of Dendritic Cells from Healthy Human Peripheral Blood

Abstract:Objective To induce and proliferate dendritic cells from human peripheral blood in vitro.Methods The mononuclear cells isolated from healthy human peripheral blood were cultured in RPMI Medium 1640 containing rhGM-CSF, IL-4, and TNF-α to generate dendritic cells. The morphologic characteristics of these cells were observed with light microscopes and the phenotypic figures were analysed with FACS. The function of these cells was reflected in mixed leukocyte reaction assay (MLR) and endocytosis ability was detected by the horseradish peroxidase (HRP) uptaken experiment.Results Peripheral blood mononuclear cells (PBMNCs) were cultured in present of rhGM-CSF, IL-4, and TNF-α for 12 days, these cells bear typical morphologic and phenotypic characteristics of dendritic cells, expressed MHCⅡ (HLA-DR) 93.5%, CD80 96%, CD86 92.6%, CD1a 60.4%, CD83 82.6%, and CD14 10.5%, gained the powerful capacity to stimulate allogeneic T cells. It showed the characteristics of mature DCs. In addition, these cells had active endocytosis ability which peaked at the sixth day in culture but deceased afterwards. Conclusion This is a reliable method to induce and proliferate DCs, which may facilitate further studies of DCs and its clinical application.
