Detection of COL4A5 gene mutations in Alport syndrome by analyzing cDNA of skin fibroblasts

Abstract:SUMMARY Objective:To establish a simple and fast approach that can be used for systematic gene analysis in patients with X linked Alport syndrome and to determine whether mutations in the COL4A5 gene turn off its transcription. Methods: The systematic analysis of the entire coding region of type Ⅳ collagen α5 chain mRNA in 21 unrelated Chinese X linked Alport syndrome patients (16 males and 5 females) and 2 male controls was carried out. The mRNA was extracted from the cultured skin fibroblast and analyzed by using reverse transcription polymerase reaction (RT PCR) and direct sequencing. Meanwhile, genomic DNA was detected for mutations by using PCR and direct sequencing. Results: We extracted enough total RNA from cultured skin fibroblasts, and the amplification of the entire α5(Ⅳ) chain cDNA was optimized. The five cDNA PCR products of a control and 1/5 cDNA PCR products (fragment E) of 8/21 X linked Alport syndrome patients were sequenced so far, and the sequences corresponded exactly to the published sequence data. Four noval mutations were detected both at the RNA level and at the genomic DNA level. Conclusion: The mutations of α5(Ⅳ) chain in X linked Alport syndrome patients can be detected by analyzing α5(Ⅳ) chain mRNA, which was extracted from the cultured skin fibroblasts and, four mutations detected in this study arenovel mutations. Although there were COL4A5 gene mutations,COL4A5 gene did not turn off transcription.
