Abstract:The concentration of 50% scavenging ratio (SC50) and the scavenging rate constants (k) and stiochiometric factor (n) were determined when (-)-epigallocatechin gallate [(-)-EGCG] scavenging superoxide anion free radical (O2.-) and hydroxyl radical (.OH). The mechanism of scavenging active free radicals of (-)-EGCG and the promotion of (-)-EGCG free radical and its structure were analyzed in vitro. Our results suggest that the centers of scavenging reaction of (-)-EGCG are B, D, and A cycles, and each (-)-EGCG traps six O2.- or .OH. It is in accord with the value of stiochiometric factor n = 6.
Mechanism of Scavenging Reactive Oxygen Radicals of Tea Catechins

Abstract:Tea catechins, have received considerable attention in recent years due to their numerous potentially beneficial medicinal properties. A common discussed mechanism of their biological and pharmacological effects is the antioxidative activities of tea catechins. In this paper, we evaluated the scavenging effects of four catechins both on superoxide anions (O2-) and hydroxyl free radical (@OH). In addition their synergic effects, structure-activity relationship, as well as stoichiometric factors and rate constants for the reactions of O2-and @OH with EGCG were discussed.

Abstract:Free radicals and oxidants play a dual role as both toxic and beneficial compounds, since they can be either harmful or helpful to the body. They are produced either from normal cell metabolisms in situ or from external sources (pollution, cigarette smoke, radiation, medication). When an overload of free radicals cannot gradually be destroyed, their accumulation in the body generates a phenomenon called oxidative stress. This process plays a major part in the development of chronic and degenerative illness such as cancer, autoimmune disorders, aging, cataract, rheumatoid arthritis, cardiovascular and neurodegenerative diseases. The human body has several mechanisms to counteract oxidative stress by producing antioxidants, which are either naturally produced in situ, or externally supplied through foods and/or supplements. This mini-review deals with the taxonomy, the mechanisms of formation and catabolism of the free radicals, it examines their beneficial and deleterious effects on cellular activities, it highlights the potential role of the antioxidants in preventing and repairing damages caused by oxidative stress, and it discusses the antioxidant supplementation in health maintenance.

Abstract:Free radicals have acquired growing importance in the fields of biology and medicine. They are produced during many different endogenous and exogenous processes. Mitochondria are the main source of endogenous reactive oxygen species (ROS) produced at cell level. The overproduction of free radicals can damage macromolecules such as nucleic acids, proteins and lipids. This leads to tissue damage in various chronic and degenerative diseases. Antioxidants play a crucial role in the body’s defense against free radicals. This review concerns the main properties of free radicals, their sources and deleterious effects. It highlights the potential role of the dietary supplementation of antioxidants and discusses unsolved problems regarding antioxidant supplements in the prevention and therapy of diseases.

Abstract:Tea catechins, the most important compounds in tea polyphenols, can efficiently scavenge superoxide anion free-radical (O₂-·), hydroxyl radical. (·OH) The mechanism of scavenging active oxygen free radicals was investigated by ESR spin trapping technique and Chemiluminescence. Results showed that various tea catechins constitute an antioxidant cycle in accordance with the decreasing order of the first reductive potential, and produce the effect of cooperative strength each other. Esterificated catechins could scavenge active oxygen free radicals more effectively than the non-esterificated ones. When ·OH and O₂-·- were scavenged by (-)-epigallocatechin gallate [(-)- EGCG], the stoichiometric factors were 6, and the rate constants of scavenging reaction reached 7.71x10(6) and 3.52x10(11) L mmol-¹ S-¹, respectively. In the mean time, tea catechins could scavenge superoxide anion fiee radical (O₂-·) and hydroxyl radical (·OH) in a dose dependent manner. But at higher concentration or pH value, tea catechins can induce the prooxidant.

Abstract:Biochar as an emerging carbonaceous material has exhibited a great potential in environmental application for its perfect adsorption ability. However, there are abundant persistent free radicals (PFRs) in biochar, so the direct and indirect PFRs-mediated removal of organic and inorganic contaminants by biochar was widely reported. In order to comprehend deeply the formation of PFRs in biochar and their interactions with contaminants, this paper reviews the formation mechanisms of PFRs in biochar and the PFRs-mediated environmental applications of biochar in recent years. Finally, future challenges in this field are also proposed. This review provides a more comprehensive understanding on the emerging applications of biochar from the viewpoint of the catalytic role of PFRs.

Abstract:Puerarin is a C-glycoside of daidzein, one of the major bioactive ingredients isolated from the root of Pueraria lobata, which has a wide spectrum of pharmacological effects. Although puerarin is well-known for its effective antioxidant activity, there is seldom a systematic theoretical study on its radical scavenging activity. Herein, the free radical scavenging ability of puerarin was investigated systematically by density functional theory (DFT) calculations. The reaction activity was compared with daidzein as well. Three reaction pathways: hydrogen atom transfer (HAT), single electron transfer followed by proton transfer (SET-PT), and sequential proton loss electron transfer (SPLET) were discussed and compared by thermodynamic parameters such as bond dissociation enthalpy (BDE), ionization potential (IP), proton dissociation enthalpy (PDE), proton affinity (PA), and electron transfer enthalpy (ETE). The reaction kinetics of puerarin with special radicals •OH and •OOH were also studied. The results obtained may be of great significance for better understanding the relationship between the antioxidant properties and structural design of puerarin, as well as other antioxidants.

Abstract:【Objective】 The study provides a theoretical basis for medical care,health,nutrition and other functions of components of nonessential oils in Lavender mint leaves.【Method】 The components of nonessential oils in Lavender mint leaves are distilled respectively by water extraction,methanol-water extraction,ethyl acetate extraction and ethanol extraction,then compared and evaluated by Chemiluminescence systems of superoxide anion free radical,hydroxyl radical,peroxynitrite and DNA oxidative damage caused by hydroxyl radical and colorimetric systems of DPPH free radical.【Result】 The main active ingredients of components of nonessential oils in Lavender mint leaves exist in polar and weak polar solvent extracts.The experiments reveal that the four different extractions of the components of nonessential oils in Lavender mint leaves can not only directly scavenge superoxide anion free radical,hydroxyl radical,peroxynitrite and DPPH free radical,but also has a better ability to obviously mitigate and delay DNA oxidative damage caused by hydroxyl radical.【Conclusion】 The components of nonessential oils in Lavender mint leaves are very effective and multifunctional crude antioxidants and free radical scavengers with a good application prospect.

Abstract:<p>Free radicals were generally regarded as highly reactive, transient and harmful species. In fact, some of the free radicals can also be inactive, long-lived and beneficial for our health. These properties of free radicals provide future possibilities for their application in various fields. Owning to their open-shell electronic structure, free radicals exhibit unique advantages in biomedical applications, such as high reactivity, photoacoustic and photothermal conversion ability, molecular magnetic. In this review, recent progress on free radicals and their applications in cancer theranostics are presented. Typical materials that exhibit controlled generation of free radicals and their applications for photodynamic therapy (PDT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), gas therapy, hypoxic cancer treatment, photothermal therapy (PTT), photoacoustic imaging (PAI) and magnetic resonance imaging (MRI) are summarized and discussed.</p>

Abstract:OBJECTIVE To study the scavenging effects of curculigoside,a main compound isolated from Curculigo orchioides Gaertn.,on hydroxyl and superoxide anion radicals.METHODS Colorimetric method and electron spine resonance(ESR) technique were applied,respectively,to determine the radical clearance ratio of curculigoside on hydroxyl and superoxide anion radicals. RESULTS As the data showed,curculigoside exhibited effective antioxidant activities on both hydroxyl and superoxide anion radicals,but the radical scavenging ratios were slightly lower than those of(-)-Epigallocatechin gallate(EGCG).CONCLUSION Curculigoside has the capability to scavenge radicals and serves as one of the active natural antioxidants.

Abstract:Objective To explore the scavenging effect of lipoic acid (LA) on free radicals produced in the digestive system of high-fat fed mice. Method Mice were randomly assigned to four groups. The control group consumed ordinary diet,The other three groups were fed with high-fat diet, high-fat diet plus 0.05% or 0.1% LA. After 6 w, the free radicals and anti-oxidation indices of whole blood and gastrointestinal system were examined. Results As compared with control group, the levels of free radicals in blood, stomach, pancreas, duodenum,jejunum and ileum were significantly increased and the anti-oxidation indices significantly decreased in high-fat diet group, and all of them recovered nearly to the control when 0.1% LA was added to the high-fat diet but no effect when 0.05% LA was added. Conclusion High-fat diet can significantly increase the levels of free radicals produced in digestive system and decrease the anti-oxidation capacity of mice. The optimal dose of LA can availably eliminate free radicals and increase the anti-oxidative capacity of mice fed high-fat.

Abstract:Objective:To study the effect of oxygen free radicals on hepatic ischemia-reperfusion injury during liver transplantation.Methods:Plasma LPO,SOD,TAC and NO of18patients were measured preop-eratively,an hepatic phase and1hour after reperfusion.Results:The plasma LPO and TAC declined,SOD and NO elevated after reperfusion.Conclusion:Oxygen free radicals play an important role in hepatic ischemia-reperfusion injury during liver transplantation.After reperfusion,protective mechanism was activated.Blood transfusion and some drug may alleviate the injury caused by free radicals and protect body.

Abstract:Free radicals-induce oxidized injury should be studied contrapuntally due to the differences of radicals species and contents.We previously found Kochia scoparia extracts showed good anti-oxidant activity and ethanol extracts showed higher capacity than water extracts.In this paper,free radical scavenging activities (DPPH · and ABTS+ · and hydroxyl radical) of different concentrations (40％,50 ％,60 ％) of ethanol extracts from Kochia scoparia were compared,and the total saponins were also considered.We found that different ethanol extracts showed differences between free radicals scavenging capacity,and each extracts exerted greatly different scavenging effect on these radicals.As ethanol concentration increased,DPPH · scavenging activities increased while hydroxyl radicals scavenging activities decreased.The total saponins content of 60％ ethanol extracts is 3 times higher than that of 40％ ethanol extracts,but the hydroxyl radicals scavenging activities are lower by 30 ％.The 60％ ethanol extracts showed the highest content of total saponins but exerted lowest scavenging capacity.These conclusions may provide more evidence for antioxidants selection and Kochia scoparia development.

Abstract:Excessive oxygen free radicals can lead to aging, cancer, and other diseases. Therefore, searching for effective antioxidants to scavenge oxygen free radicals has become the focus of modern medicine. In this study, the molecular mechanism of Licorice Green Tea Beverage (LGTB) in scavenging oxygen free radicals was investigated by means of network pharmacology, molecular docking and experimental verification. Network pharmacology studies have shown that paeonol, eugenol, cinnamaldehyde, swertisin, rutin, glycyrrhetinic acid, oleic, pelargonidin-3-O-glucoside and quercetin, kaferempol were the main active components of LGTB, and SOD and CAT are important targets for LGTB in scavenging oxygen free radicals. The results of molecular docking showed that these representative compounds had good affinity to SOD and CAT target proteins. In vitro free radical scavenging experiments showed that LTGB had significant scavenging effects on both DPPH and ABTS radicals, and had strong total reducing power. In vitro cell experiments showed that LGTB could protect HaCaT cells from oxidative stress induced by H2 O2 . The mechanism of LGTB was related to the increase of SOD and CAT activity. Western blotting showed that LGTB could inhibit PI3K/AKT/HIF-1 signaling pathway and improve the antioxidant capacity of HaCaT cells. In vivo experiments showed that LGTB could significantly increase mouse visceral index, increase serum SOD and GSH-Px activity, decrease the content of MDA, and improve liver and kidney pathological state. This study reported the molecular mechanism of LTGB scavenging oxygen free radicals, which provided scientific basis for the treatment and clinical research of aging and other diseases caused by excessive free radicals. PRACTICAL APPLICATIONS: Free radicals are produced by the normal response of cells during aerobic respiration and perform various functions, such as signaling and providing protection against infection. However, excessive free radicals can lead to aging, cancer, and other diseases. The antioxidant can overcome the harm caused by excessive free radicals. In this study, we investigated the molecular mechanism of scavenging oxygen free radicals of Licorice Green Tea Beverage (LGTB) through network pharmacology and molecular docking, and its efficacy was verified by free radical scavenging experiment in vitro, HaCaT cell oxidative stress injury induced by H2 O2 , D-galactose to establish an aging model in mice and Western blotting experiment. It not only elucidates its mechanism at the system level, but also proves its validity at the biological level. It provides the theoretical basis and experimental evidence for the follow-up research and promotion of the product.

Abstract:Reactive oxygen and nitrogen species (RONS) are essential for normal physiological processes and play important roles in cell signaling, immunity, and tissue homeostasis. However, excess radical species are implicated in the development and augmented pathogenesis of various diseases. Several antioxidants may restore the chemical balance, but their use is limited by disappointing results of clinical trials. Nanoparticles are an attractive therapeutic alternative because they can change the biodistribution profile of antioxidants, and possess intrinsic ability to scavenge RONS. Herein, we review the types of RONS, how they are implicated in several diseases, and the types of nanoparticles with inherent antioxidant capability, their mechanisms of action, and their biological applications.
