Correlation between urinary protein excretion and morphometric changes of glomerular foot process in rats with puromycin aminonucleoside nephropathy

Abstract:Objective:To investigate the correlation between urinary protein excretion and morphometric changes of podocyte foot processes in rat puromycin aminonucleoside (PAN) nephropathy and to understand the mechanism of proteinuria. Methodology:A total of SD 30 rats were enrolled in this study. They were divided randomly into control group (15 rats, group A) and experimental group (15 rats, group B). The nephropathy model of rats was established by the injection of PAN. Urine protein excretion in 24h was determined by biuret method. Morphometric methods were applied, and transmission electron microscopy as well as image analysis was used to study the changes of foot processes. Results:①The proteinuria was gradually increased at the 5th day after PAN injection, and went to the peak level at the 10th day group B, which was significant different from that in group A (P0.01). At the 15th day, the amount of proteinuria in group B began to decrease, and was still higher than that in group A (P0.05). At the 20th day, the amount of urinary protein decreased to the normal level in group B.② In the process of development of PAN nephropathy, the foot process became broader and then fusion. After 10 days, the foot processes began to recover. Between the group B and A, there were significantly differences (P0.01) in the morphometric parameters indicated the foot process width, volume density, and specific surface (ratio of surface to volume) of foot process at the different time points. The surface density was different from day 2 to 15 between the group B and A (P0.05). ③There was a positive correlation between urinary protein excretion and podocyte foot process width (r=0.9294, P=0.02), and volume density (r=0.9559, P=0.011). However, there was a negative correlation between proteinuria excretion and specific surface(r=-0.9388, P=0.018). Conclusion:There was a correlation between morphological changes of podocyte foot process and the urinary protein excretion in rats PAN nephropathy. The morphological changes presented prior to proteinuria, but the recovery of proteinuria was earlier than that of foot process morphology.
