Abstract:Two full-length genomic cDNA clones, pTA/FMDV and pCA/FMDV, were constructed that contained three point-mutants [A174G and A308G (not present in pTA/FMDV); T1029G] in the genome compared with the wild type A/AKT/58 strain of foot-and-mouth disease virus. These two viruses were rescued by co-transfection of pCA/FMDV with pCT7RNAP, which can express T7 RNA polymerase in BHK-21 cell-lines, or by transfection of the in vitro transcribed RNA. Their biological properties were analyzed for their antigenicity, virulence in suckling-mice (LD 50 ) and growth kinetics in BHK-21 cells. The in vivo rescued viruses showed high pathogenicity for 3-day-old unweaned mice (LD 50 =10 −7.5 ). However, the in vitro transcribed RNA derived from pTA/FMDV had lower pathogenicity for suckling-mice (LD 50 =10 −6 ), and the in vivo transcribed RNA recovered from pCA/FMDV co-transfected with pCT7RNAP showed no significant differences from the wild type virus. These data showed that recovery of the infectious foot-and-mouth disease virus directly from the use of in vivo techniques was better than from in vitro methods. Furthermore, the reverse genetic procedure technique was simplified to a faster one-step procedure based on co-transfection with pCT7RNAP. These results suggest that in vivo RNA transcripts may be more valuable for engineering recombinant foot-and-mouth disease virus than in vitro RNA transcripts, and may contribute to further understanding of the biological properties, such as replication, maturation and quasispecies, of the foot-and-mouth disease virus.
Amber codon is genetically unstable in generation of premature termination codon (PTC)-harbouring Foot-and-mouth disease virus (FMDV) via genetic code expansion

Abstract:The foot-and-mouth disease virus (FMDV) is the causative agent of FMD, a highly infectious and devastating viral disease of domestic and wild cloven-hoofed animals. FMD affects livestock and animal products' national and international trade, causing severe economic losses and social consequences. Currently, inactivated vaccines play a vital role in FMD control, but they have several limitations. The genetic code expansion technology provides powerful strategies for generating premature termination codon (PTC)-harbouring virus as a live but replication-incompetent viral vaccine. However, this technology has not been explored for the design and development of new FMD vaccines. In this study, we first expanded the genetic code of the FMDV genome via a transgenic cell line containing an orthogonal translation machinery. We demonstrated that the transgenic cells stably integrated the orthogonal pyltRNA/pylRS pair into the genome and enabled efficient, homogeneous incorporation of unnatural amino acids into target proteins in mammalian cells. Next, we constructed 129 single-PTC FMDV mutants and four dual-PTC FMDV mutants after considering the tolerance, location, and potential functions of those mutated sites. Amber stop codons individually substituted the selected amino acid codons in four viral proteins (3D, L, VP1, and VP4) of FMDV. We successfully rescued PTC-FMDV mutants, but the amber codon unexpectedly showed a highly degree of mutation rate during PTC-FMDV packaging and replication. Our findings highlight that the genetic code expansion technology for the generation of PTC-FMD vaccines needs to be further improved and that the genetic stability of amber codons during the packaging and replication of FMDV is a concern.
A Heat-Induced Mutation on VP1 of Foot-and-Mouth Disease Virus Serotype O Enhanced Capsid Stability and Immunogenicity

Abstract:Foot-and-mouth disease (FMD) is a highly contagious viral disease affecting cloven-hoofed animals that causes a significant economic burden globally. Vaccination is the most effective FMD control strategy. However, FMD virus (FMDV) particles are prone to dissociate when appropriate physical or chemical conditions are unavailable, such as an incomplete cold chain. Such degraded vaccines result in compromised herd vaccination. Therefore, thermostable FMD particles are needed for use in vaccines. This study generated thermostable FMDV mutants (M3 and M10) by serial passages at high temperature, subsequent amplification, and purification. Both mutants contained an alanine-to-threonine mutation at position 13 in VP1 (A1013T), although M3 contained 3 additional mutations. The selected mutants showed improved stability and immunogenicity in neutralizing antibody titers, compared with the wild-type (wt) virus. The sequencing analysis and cryo-electron microscopy showed that the mutation of alanine to threonine at the 13th amino acid in the VP1 protein (A1013T) is critical for the capsid stability of FMDV. Virus-like particles containing A1013T (VLPA1013T) also showed significantly improved stability to heat treatment. This study demonstrated that Thr at the 13th amino acid of VP1 could stabilize the capsid of FMDV. Our findings will facilitate the development of a stable vaccine against FMDV serotype O. IMPORTANCE Foot-and-mouth disease (FMD) serotype O is one of the global epidemic serotypes and causes significant economic loss. Vaccination plays a key role in the prevention and control of FMD. However, the success of vaccination mainly depends on the quality of the vaccine. Here, the thermostable FMD virus (FMDV) mutants (M3 and M10) were selected through thermal screening at high temperatures with improved stability and immunogenicity compared with the wild-type virus. The results of multisequence alignment and cryo-electron microscopy (cryo-EM) analysis showed that the Thr substitution at the 13th amino acid in the VP1 protein is critical for the capsid stability of FMDV. For thermolabile type O FMDV, this major discovery will aid the development of its thermostable vaccine.

Abstract:Background Foot-and-mouth disease virus (FMDV) uses a highly conserved Arg-Gly-Asp (RGD) triplet for attachment to host cells and this motif is believed to be essential for virus viability. Previous sequence analyses of the 1D-encoding region of an FMDV field isolate (Asia1/JS/CHA/05) and its two derivatives indicated that two viruses, which contained an Arg-Asp-Asp (RDD) or an Arg-Ser-Asp (RSD) triplet instead of the RGD integrin recognition motif, were generated serendipitously upon short-term evolution of field isolate in different biological environments. To examine the influence of single amino acid substitutions in the receptor binding site of the RDD-containing FMD viral genome on virus viability and the ability of non-RGD FMDVs to cause disease in susceptible animals, we constructed an RDD-containing FMDV full-length cDNA clone and derived mutant molecules with RGD or RSD receptor recognition motifs. Following transfection of BSR cells with the full-length genome plasmids, the genetically engineered viruses were examined for their infectious potential in cell culture and susceptible animals. Results Amino acid sequence analysis of the 1D-coding region of different derivatives derived from the Asia1/JS/CHA/05 field isolate revealed that the RDD mutants became dominant or achieved population equilibrium with coexistence of the RGD and RSD subpopulations at an early phase of type Asia1 FMDV quasispecies evolution. Furthermore, the RDD and RSD sequences remained genetically stable for at least 20 passages. Using reverse genetics, the RDD-, RSD-, and RGD-containing FMD viruses were rescued from full-length cDNA clones, and single amino acid substitution in RDD-containing FMD viral genome did not affect virus viability. The genetically engineered viruses replicated stably in BHK-21 cells and had similar growth properties to the parental virus. The RDD parental virus and two non-RGD recombinant viruses were virulent to pigs and bovines that developed typical clinical disease and viremia. Conclusions FMDV quasispecies evolving in a different biological environment gained the capability of selecting different receptor recognition site. The RDD-containing FMD viral genome can accommodate substitutions in the receptor binding site without additional changes in the capsid. The viruses expressing non-RGD receptor binding sites can replicate stably in vitro and produce typical FMD clinical disease in susceptible animals.
Cloning of VP1 Gene of Foot-and-Mouth Disease (FMDV) Type O and Its Expression in Escherichia coli

Abstract: In this study, a special primer pair containing BamHⅠand HindⅢaccording to complete genome of foot-and-mouth disease (FMDV) Type O was designed to amplify VP1 gene from plasmid pMD-P1 of FMDV type O. The interest fragment was inserted into PMD18-T vector and the recombinant plasmid was indentifed and the sequence was detected . The target gene VP1 was subcloned into PET32A vector digested by BamHⅠand HindⅢ respectively. Positive clone with interest gene was identified by restriction analysis、PCR and DNA sequencing. Then the recombinant plasmid was transformed into Escherichia coli BL21(DE3) for VP1 expression. The interest gene was induced to express in E.coli with different concentration IPTG. The bacteria containing PET32A-VP1 was collected and examined by SDS-PAGE and western-blotting. Result showed that the structural protein was expressed in E.coli and the amount of expression was higher. Melecular weight of the protein was 41 ku.The protein can be recognized by the positive serum of FMDV, the amount of protein is about 30% of the total bacteria protein. This study showed that the structural protein VP1 amount of expression was higher in E.coli and the protein has antigenicity.

Abstract:Objective To construct and identify recombinant adenoviruses expressing FMDV VP1 gene.Methods FMDV VP1 gene was cloned into pAdeno Vator-CMV5-IRES-GFP,named pA5-VP1.pA5-VP1 was Linearized by PmeⅠ,and then Linearized pA5-VP1 and pAdeno VatorΔE1/E3 cotransformed into E.coli BJ5183 competent cells by electroporation.The homologous recombinant plasmid pAd5-VP1 was selected and linearized by PacⅠto expose the encapsidation signal.Linearized recombinant virus plasmids were transfected into 293A cells by LipofectamineTM2000,and recombinant viruses was named rAd5-VP1.Infection titer and rate were evaluated through monitoring green fluorescent protein(GFP) expression.Expression of FMDV VP1 was testified by PCR and western-blot.Results Recombinant adenoviruses expressing FMDV VP1 was constructed successfully,and the filter of rAd5-VP1 was 1011.87 TCID50/ml.PCR result indicated that VP1 expresses in HEK-293A cells.Western-blot result indicated that Recombinant adenoviruses could react with FMDV positive serum.Conclusion Recombinant adenoviruses rAd5-VP1 had specific antigenicity of FDMV.And some biological characteristics of the recombinant viruses were stable after passaged 10 times.

Abstract:【Objective】The aim of the study was to develop a reverse genetics platform of infectious foot-and-mouth disease (FMDV) in vivo, which is a base for the construction and function researches of viruses. 【Method】 The full-length genome of FMDV Asia1/JS/China/2005 strain was assembled into a mammalian expression vector downstream of a T7 promoter. The recombinant plasmids, pFMDV-A were linearized with NotI enzyme and cotransfected into BHK-21 cells with pcDNAT7P plasmids that could express T7 RNA polymerase to research reverse genetics of FMDV. 【Result】 The transfected cells were serially passaged, and the third passage showed apparent cytopathogenicity effect (CPE) within 48 h, the CPE were observed after 10 h in fourth passage. The results of the RT-PCR, indirect immunofluorescence, and electron microscope showed that FMDV was rescued successfully in vivo from the full-length cDNA clone of FMDV. The recovered virus contained genetic tags. Repeat experiments also obtained recovered virus with the established method. The rescued virus showed a similar pathogenicity in suckling mouse (LD50) compared to its wild-type virus. 【Conclusion】 Plasmid-based reverse genetics system was developed in authors’ laboratory for efficient and stable generation of infectious FMDV.

Abstract:One pair of primers were designed and synthesized to amplify the VP1-2A gene of FMDV.The predicted 1 055 bp fragment had been obtained by RT-PCR from the RNA of FMDV YN strain.The result of cloning and sequencing analysis showed that the homology of nucleotide sequence of the VP1-2A gene between the strain and the referenced sequence of type O foot-and-mouth disease is between 77.65%～93.00%,and the homology of amino acids is between 87%～97%.
Prokaryotic Expression and Activity Analysis of VP1 Gene of Foot-and-Mouth Disease Virus Serotype O

Abstract:The objective of this research is to get the FMDV VP1 protein which be of antigenancy activity.The recombinant expression vector pET-VP1 was constructed by cloning VP1 gene of FMDV into the prokaryotic expression plasmid pET32a from PMD18T-VP1.After the recombinant plasmid was verified by PCR and sequenced analysis,the result showed that VP1 gene was successfully cloned into expression plasmid pET32a,The protein corresponding to the VP1 gene was expressed after induction with IPTG.SDS-PAGE and Western-blot with the product of bacterial culture in different time showed that the molecular mass of the protein was approximately 45 ku,which was identified by positive sera of FMDV;The protein was refolded by dialysis method,and was assayed by ELISA,the result showed that it demonstrates high activity.So we can say that VP1 protein was expressed efficiently in E.coli,and the renaturation VP1 protein can be developed as diagnosis antigen or vaccine.

Abstract:Rapid evolution is widespread in the viral kingdom and a major concern for developing universal vaccines. The isolation of large numbers of viral sequence variants at highly variable regions in viral proteins remains a daunting challenge. Foot-and-mouth disease virus (FMDV) is a picornavirus and an underlying cause of a highly contagious disease of cloven-hoofed animals which often results in substantial economic losses. This study aimed at developing a combined method for the isolation of novel sequences to cope with rapid viral variations at the G-H loop of FMDV VP1 protein. DNA primer sets harbouring random nucleotides scattered in the G-H loop region, carrying additionally coding sequences for a T cell epitope and the carboxyl terminus, were designed and assembled by asymmetric PCR. After eliminating insert-free vector background and enriching positive clones, 100 novel sequences targeting the rapid viral variations of the G-H loop were obtained from 2 plasmid libraries. The method allowed radical changes in amino acid sequence, and our study has identified critical steps in the batch discovery of viral sequence variants, paving the way for future research on the possibility of developing a polyvalent universal vaccine.
Gene Cloning and Expression of the VP1 and 2VP1 Gene of FMDV

Abstract:The C-terminal half of VP1 gene of FMDV was amplified by PCR. The PCR products were cloned into pMD18-T and sequenced. The squence result indicated that the C-terminal half of VP1 gene was about 285 bp and coded 75 amino acids.The homology analysis of the gene with O/HKN/12/91 and O/PEN/TAW/99 in GenBank indicated that it was relatively conserved. Then the PCR products were inserted into pGEX-KG and transformed BL21(DE3) bacteria. The expression of GST-2VP1 and GST-VP1 was induced with IPTG and confirmed by SDS-PAGE, the recombinant protein had a molecular weight of 45 ku and 37 ku respectively.Their activity were confirmed by Western blot. Based on expressed GST-2VP1 protein as antigen, the ELISA to detect antibody against VP1 was developed and was primarily used to detect serum samples.

Abstract:VP1 gene of foot-and-mouth disease virus(FMDV) is the major immunogenic gene as it contains T cell and B cell epitopes.In this study,we constructed a recombinant expressing plasmid(KG2VP1) including two VP1 genes(2VP1) of Asia 1 FMDV,and the 2VP1 gene was expressed in E.coli BL21.The fusional protein(GST2VP1) which was purified by Sephadex-G200 sieve chromatography demonstrated the reactionogenicity by Western blot.And the mice test indicated that the GST2VP1 can induce similar levels of ELISA and neutralization antibody like inactivated vaccine.We provide foundation for researching the immunogenicity of FMDV serotype Asia 1 and further application of GST2VP1 in future.

Abstract:According to the complete genome of foot-and-mouth disease virus(FMDV)type O,a pair of special primers was designed to amplify VP1 gene.The VP1 gene was amplified by RT-PCR and subsequently inserted into the expression vector pGEX-6p-1 and induced by IPTG.Then SDS-PAGE showed the expressed protein was 51 kD in molecular weight.Then the product was purified by GSTrap FF columns.The product was detected through Western-blot that showed the protein has antigenicity.It provided fundamental data and materials for further investigation on diagnosis method of FMDV.

Abstract:The research for molecular epidemiology of foot-and-mouth disease virus (FMDV) usually bases on the a-nalysis of VP1 gene sequence.This article described a strain of FMDV Asia 1 type that was isolated from pigs.The VP1 gene of the strain was amplified by RT-PCR and was sequenced.Alignment with other strains of sequence of VP1 gene,we got the homology ranging from 83.3% to 86.4% when compared to the strains of FMDV type Asia 1 from China,while got 81.4% to 98.4% homology when compared to the strains from other countries.There was a variance between the isolate and other strains,and only 2 strains shared the homology over 90%.

Abstract:Foot-and-mouth disease virus (FMDV), the most acid-unstable virus among picornaviruses, tends to disassemble into pentamers at pH values slightly below neutrality. However, the structural integrity of intact virion is one of the most important factors that influence the induction of a protective antibody response. Thus, improving the acid stability of FMDV is required for the efficacy of vaccine preparations. According to the previous studies, a single substitution or double amino acid substitutions (VP1 N17D, VP2 H145Y, VP2 D86H, VP3 H142D, VP3 H142G, and VP1 N17D + VP2 H145Y) in the capsid were introduced into the full-length infectious clone of type O FMDV vaccine strain O/HN/CHN/93 to develop seed FMDV with improved acid stability. After the transfection into BSR/T7 cells of constructed plasmids, substitution VP1 N17D or VP2 D86H resulted in viable and genetically stable FMDVs, respectively. However, substitution VP2 H145Y or VP1 N17D + VP2 H145Y showed reverse mutation and additional mutations, and substitution VP3 H141G or VP3 H141D prevented viral viability. We found that substitution VP1 N17D or VP2 D86H could confer increased acid resistance, alkali stability, and thermostability on FMDV O/HN/CHN/93, whereas substitution VP1 N17D was observed to lead to a decreased replication ability in BHK-21 cells and mildly impaired virulence in suckling mice. In contrast, substitution VP2 D86H had no negative effect on viral infectivity. These results indicated that the mutant rD86H carrying substitution VP2 D86H firstly reported by us could be more adequate for the development of inactivated FMD vaccines with enhanced acid stability.

Abstract:Previous studies have shown that the FMDV Asia1/YS/CHA/05 high-fidelity mutagen-resistant variants are attenuated (Zeng et al., 2014). Here, we introduced the same single or multiple-amino-acid substitutions responsible for increased 3Dpol fidelity of type Asia1 FMDV into the type O FMDV O/YS/CHA/05 infectious clone. The rescued viruses O-DA and O-DAMM are lower replication fidelity mutants and showed an attenuated phenotype. These results demonstrated that the same amino acid substitution of 3Dpol in different serotypes of FMDV strains had different effects on viral fidelity. In addition, nucleoside analogues were used to select high-fidelity mutagen-resistant type O FMDV variants. The rescued mutagen-resistant type O FMDV high-fidelity variants exhibited significantly attenuated fitness and a reduced virulence phenotype. These results have important implications for understanding the molecular mechanism of FMDV evolution and pathogenicity, especially in developing a safer modified live-attenuated vaccine against FMDV.
Sequences Outside That of Residues 93–102 of 3A Protein Can Contribute to the Ability of Foot-and-mouth Disease Virus (FMDV) to Replicate in Bovine-Derived Cells

Abstract:Foot-and-mouth disease (FMD) is a highly contagious and economically devastating disease of cloven-hoofed animals. During 2010 and 2011, there was an epidemic of the Mya-98 lineage of the Southeast Asia (SEA) topotype in East Asia, including China. Changes in the FMDV 3A protein have been previously reported to be associated with the inability of FMDV to grow in bovine cells and cause disease in cattle. In this paper, we report the generation of a full-length infectious cDNA clone of FMDV O/SEA/Mya-98 strain O/GZSB/2011 for the first time along with two genetically modified viruses with deletion at positions 93-102 and 133-143 in 3A based on the established infectious clone. All the recombinant viruses grew well and displayed growth properties and plaque phenotypes similar to those of the parental virus in baby hamster kidney (BHK-21) cells, porcine kidney (PK-15) cells, and primary fetal porcine kidney (FPK) cells. While the recombinant viruses rvGZSB and rvSB Delta 133-143 exhibited similar growth properties and plaque phenotypes with the parental virus in primary fetal bovine kidney (FBI() cells, the recombinant virus rvSB Delta 93-102, containing deletion at positions 93-102 in 3A, grew at a slower rate and had a smaller plaque size phenotype in FBK cells than that of the parental virus. Therefore, the results suggest that the deletion at positions 93-102 of 3A protein does not affect FMDV replication efficiency in BHK-21, PK-15 and FPK cells, but affects virus replication efficiency in FBK cells, although, cannot alone account for the inability to replicate in bovine cells. Abstract:Inactivated foot-and-mouth disease virus (FMDV) vaccines have been used widely to control foot-and-mouth disease (FMD). However, the virions (146S) of this virus are easily dissociated into pentamer subunits (12S), which limits the immune protective efficacy of inactivated vaccines when the temperature is higher than 30 °C. A cold-chain system can maintain the quality of the vaccines, but such systems are usually not reliable in limited-resource settings. Thus, it is imperative to improve the thermostability of vaccine strains to guarantee the quality of the vaccines. In this study, four recombinant FMDV strains containing single or multiple amino acid substitutions in the structural proteins were rescued using a previously constructed FMDV type O full-length infectious clone (pO/DY-VP1). We found that single or multiple amino acid substitutions in the structural proteins affected viral replication to different degrees. Furthermore, the heat and acid stability of the recombinant viruses was significantly increased when compared with the parental virus. Three thermally stable recombinant viruses (rHN/DY-VP1Y2098F, rHN/DY-VP1V2090A-S2093H, and rHN/DY-VP1V2090A-S2093H-Y2098F) were prepared as inactivated vaccines to immunize pigs. Blood samples were collected every week to prepare sera, and a virus neutralization test showed that the substitutions S2093H and Y2098F, separately or in combination, did not affect the immunogenicity of the virus, but the Y2098F mutation increased the thermostability significantly (p < 0.05). Therefore, the rHN/DY-VP1Y2098F mutant should be considered for use in future vaccines.

Abstract:According to the host range specificity,Papaya Ringspot Virus (PRSV) is classified into two types,called type W and P. The VPg gene of PRSV was involved in genome replication. To obtain the VPg gene of PRSV-W,the VPg gene of PRSV-P was modified into a VPg gene of PRSV-W by long primer extension and overlap extension PCR. The results showed that nine sites of VPg gene from PRSV-P were mutated and the VPg gene of PRSV-W was attained successfully. The research was benefit to further investigate both type of PRSV host specificity for infection of papaya.

Abstract:The objective of this research is to obtain the VP1 protein with high activity of FMDV serotype A.The recombinant expression vector pET-A-vp1 was constructed by cloning vp1 gene which was amplified by PCR into the prokaryotic expression plasmid pET28a from pMD19A-T-vp1.The E.coli BL21(DE3)bacteria which contained recombinant plasmid pET-A-vp1 were induced by IPTG.SDS-PAGE showed that VP1 protein was expressed in the form of inclusion body,and its molecular weight is about 29ku.Based on optimizing IPTG concentration and expression time,the largest amount of expression of VP1 protein was induced by 0.3 mmol/L IPTG for 6 hours at 37 ℃.Western-Blot analysis showed that the expression of VP1 protein can be specific recognized by positive serum of FMDV serotype A.After purification by washing inclusion and refolding VP1 by urea concentration gradient dialysis,ELISA test showed that VP1 had a high activity.
