Comparative alkaline phosphatase physiological characteristics of Pseudo-nitzschia pungens and Aureococcus anophagefferens

Abstract:The batch culture experiment was used to study the differences in alkaline phosphatase(AP) physiological characteristics between two marine microalgae, Pseudo-nitzschia pungens and Aureococcus anophagefferens, and to analyze the regulation of extracellular phosphorus(DIP) and intracellular phosphorus(PP) to algal AP expression. The results showed that AP of both P. pungens and A. anophagefferens was inducible. The expression of alkaline phosphatase activity(APA) was co-regulated by DIP and PP. The response of P. pungens APA was more sensitive than A. anophagefferens. P. pungens expressed abundant APA when DIP was still high(1.80 μmol·L–1) and PP was replete(222.19 fmol·cell–1) while A.anophagefferens APA didn't show a significant increase until both DIP and PP were exhausted(0.26 μmol·L–1 and 4.49 fmol·cell-1, respectively). The AP of P. pungens was almost cell-bounded, while 6.1%-20.9% of A. anophagefferens AP released into the culture medium. The maximal APA of A. anophagefferens was 73 times higher than P. pungens APA in the same cell volume, so the ability to hydrolyze dissolved organic phosphorus by AP was different for P. pungens and A. anophagefferens.
