Abstract:A novel lotus plumule polysaccharide (LPPS) was purified, characterised and cultured with RAW264.7 macrophages to evaluate its anti-inflammatory characteristics. LPPS was purified using Sepharose 6B gel filtration and dissolved into two major components, fraction-1 (F1) and fraction-2 (F2). The molecular weights of native F1 and F2 were approximately distributed at >2,000 and 25.7kDa, respectively. The total protein and carbohydrate constituent ratios in LPPS, F1, and F2 were 30.0±0.9% vs. 70.0±0.9%, 30.1±2.6% vs. 69.9±2.6%, and 96.5±6.1% vs. 3.5±6.1% (w/w), respectively, suggesting that F1 may be a major proteo-polysaccharide component and F2 a glycoprotein constituent in LPPS. Pro-/anti-inflammatory (IL-6/IL-10) cytokine secretion ratios by lipopolysaccharide-stimulated RAW264.7 macrophages were significantly decreased by F1 and F2 treatments, particularly by F2, in a dose-dependent manner under a preventive experimental model. This study suggests that purified components, F1 and F2 from LPPS, have strong anti-inflammatory effects on LPS-induced inflamed macrophages in a preventive manner.

Comprehensive Reviews in Food Science and Food Safety

Abstract:Nelumbo nucifera Gaertn., also known as the sacred lotus, is extensively cultivated in Southeast Asia, primarily for food and as an herbal medicine. This article reviews studies published between 1995 and 2017, on flavonoid and phenolic acid profiles and contents of 154 different cultivars of lotus. So far, some 12 phenolic acids and 89 to 90 flavonoids (47 flavonols, 25 to 26 flavons, 8 flavan-3-ols, 4 flavanons, and 5 anthocyanins) have been isolated from different parts of the lotus plant, including its leaves (whole leaf, leaf pulp, leaf vein, and leaf stalk), seeds (seedpod, epicarp, coat, kernel, and embryo), and flowers (stamen, petal, pistil, and stalk), although not all of them have been quantified. Factors affecting flavonoids and phenolic acid profiles, including types of tissues and extracting factors, are discussed in this review, in order to maximize the application of the lotus and its polyphenols in the food industry. Health promotion activities, attributed to the presence of flavonoids and phenolic acids, are described along with toxicology studies, illustrating appropriate usage and safe consumption dosages of lotus extracts. This review also presents the controversies and discusses the research gaps that limit our ability to obtain a thorough understanding of the bioactivities of lotus extracts.

Abstract:Nelumbo nucifera Gaertn. (lotus) is a widely distributed plant with a long history of cultivation and consumption. Almost all parts of the lotus can be used as foodstuff and nourishment, or as an herb. It is noteworthy that the polysaccharides obtained from lotus exhibit surprisingly and satisfying biological activities, which explains the various benefits of lotus to human health, including anti-diabetes, anti-osteoporosis, antioxidant, anti-inflammatory, anti-tumor, etc. Here, we systematically review the recent major studies on extraction and purification methods of polysaccharides from different parts (rhizome, seed, leaf, plumule, receptacle and stamen) of lotus, as well as the characterization of their chemical structure, biological activity and structure-activity relationship, and the applications of lotus polysaccharides in different fields. This article will give an updated and deeper understanding of lotus polysaccharides and provide theoretical basis for their further research and application in human health and manufacture development.

Abstract:Lotus Receptaculum Nelumbinis has been sparking wide research interests due to its rich phenolic compounds. In the present work, ultrasonic-assisted extraction coupled with glycerol was employed to extract phenolic compounds from Receptaculum Nelumbinis and the process was optimized using a response surface methodology with Box-Behnken design (BBD). The optimal conditions for the total phenolic content (TPC) extract were obtained: glycerol concentration of 40%, an extraction temperature of 66 °C, ultrasonic time of 44 min, and the solvent-to-solid ratio of 55 mL/g. Under these optimum extraction conditions, the extraction yield of TPC was 92.84 ± 2.13 mg gallic acid equivalents (GAE) /g. Besides, the antioxidant activities demonstrated the ability of free radical scavenging by four different methods that included 2,2-Diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and reducing activity (RA) were 459.73 ± 7.07, 529.97 ± 7.30, 907.61 ± 20.28, and 983.66 ± 11.80 μmol TE/g, respectively. Six phenolic compounds were identified by ultra-high pressure liquid chromatography combined with triple-time-of-flight mass spectrophotometry (UPLC-Triple-TOF/MS) from the extracts. Meanwhile, Fourier transform infrared (FTIR) was conducted to identify the characteristic functional groups of the extracts and thus reflected the presence of polyphenols and flavonoids. Scanning electron microscopy (SEM) illustrated the microstructure difference of four treatments, which might explain the relationships between antioxidant activities and the structures of phenolic compounds.

Abstract:Lotus seedpod, lotus seed coat, lotus seed epicarp, and lotus plumule, which are common lotus wastes produced in the food processing industry, are often directly discarded or underutilized, resulting in the obvious waste of lotus resources. Since lotus wastes contain multiple bioactive compounds such as polyphenols, alkaloids, and polysaccharides that can promote human health, the development and utilization of bioactive compounds from lotus wastes has attracted increasing attention. At present, both conventional and modern extraction methods have been widely used in the extraction of bioactive compounds from lotus wastes. Meanwhile, some new extraction methods have also been used to extract bioactive compounds from lotus wastes to increase the extraction rate and have shown significant effects. Polyphenols, alkaloids, and polysaccharides in lotus wastes exhibit bioactivities in many fields, especially in anti-glycation, anti-proliferative, anti-oxidant, and anti-inflammatory. However, most bioactive studies are still in-vitro stage, which need to be further explored by in-vivo experiments in the future. This paper reviews the recent advances in extraction methods and biological activities of the main chemical compounds in lotus waste, which is expected to provide a reference for the development and utilization of lotus waste resources.
Study on Technics of Lotus Root Beverage with the Function of Cool and Thirst-Quenching

Abstract:Objective With lotus root as main raw materials, florists chrysanthemum juice, honey and xylitol as auxiliary materials, to develop a kind of cool and thirst-quenching of lotus root beverage, explore the various factors that influences the quality of lotus root beverage and find the best formula. Methods The optimal color protection conditions (the additive amount of citric acid, the time and temperature of blanching), optimal enzymatic hydrolysis conditions (the additive amount of cellulase and amylase, enzymatic hydrolysis time, temperature and pH) and the additive amount of lotus root juice, tribute chrysanthemum juice, honey and xylitol were explored by single factor experiments. On this basis, the optimal formula of lotus root beverage was determined by L9(34)orthogonal test. Results The optimal color protection conditions were showed as follows: the additive amount of citric acid was 0.3%, blanching time was 3 min and blanching temperature was 85 ℃; the optimal enzymatic hydrolysis conditions were showed as follows: the additive amount of cellulase and amylase were 0.3%, enzymatic hydrolysis temperature was 65 ℃, enzymatic hydrolysis time was 2.5 h and pH was 5.5. The optimal formulas showed as follows: lotus root juice was 50%, tribute chrysanthemum juice was 25%, honey was 4% and xylitol was 12%. Conclusions The lotus root beverage is a kind of beverage with cool and thirst-quenching, rich flavor and excellent taste, which is suitable for consumers to drink.

Abstract:Objective: To optimize the extracting process of procyanidins and nuciferine from the lotus leaf simultaneously.Method: Orthogonal design was used to optimize the extracting process of procyanidins and nuciferine with the transfer rate as the evaluation index, The effect of several important factors such as the pH of extracting solvent,the volume fraction of acid ,the extraction time,the material-liquid ratio and the extract times of procyanidins and nuciferine were studied. Result.. The optimal extraction process was as follows: 0.3% (v/v)of phosphate water solution ; the material- liquid ratio was 1 : 25 ; two times with 90min per time. The transfer rate of procyanidins was 89.2%, while the transfer rate of nuciferine was 69.9%. Conclusion: The optimum extraction processes were stable, economical ,feasible and adapts for industrialized production.

Abstract:Nelumbo nucifera Gaertn. has a relatively wide geographical distribution and biological diversity; various lotus parts have excellent food and medicinal values. Lotus seeds, which are currently the oldest known plant seeds, contain many functional ingredients. They can be eaten raw or cooked, and are often added to foods as ingredients or supplements. Many naturally occurring ingredients isolated from lotus seeds are certified to be multiple functional compounds, such as polyphenols, protein, polysaccharides. Proteins and carbohydrates are the main nutrients of lotus seeds. Low fat content and good proportion of amino acids confer to lotus seeds unique nutritional values that have attracted increasing attention around the world: multiple studies have assessed the functional components of lotus seeds. The bioactivity of ingredients from lotus seeds in vitro and in vivo include antioxidant activity, hypoglycemic, immunomodulatory, antibacterial, anti-inflammatory, analgesic effects as well as gastrointestinal regulation. Lotus seeds show prospective application in function food area and traditional medicine research. Furthermore, structure–activity relationship of functional compounds from lotus seeds will attracts much more interests in recent years. This work briefly reviews the nutrition composition, physiological functions and processing methods of lotus seeds, describing the impact of the latter on nutrient preservation. In addition, this review addresses the recent progresses made in this area and discusses the potential applications and limitations.

On The Processing Technology of Low-sugar Sulfur-free Preserved Lotus Root

Abstract:This paper studies the process,such as color protecting,hardening and drying,in making lowsugar sulfur-free preserved lotus root. Results show that to make persevered lotus root of high quality,the optimal color fixative is 0. 5% Nacl,0. 3% Citric acid and 0. 2% Vitamin C,the optimal hardening concentration of CaCl2 is 1. 5%,the optimal precooked time is 9 min,the optimal glucose concentration is 50%,and the optimal drying time is 8 hours at 55 ℃ 。

Processing Characteristics and Flavour of Full Lotus Root Powder Beverage

Abstract:BACKGROUND: Lotus root beverage is commonly made from raw lotus root (RLR). However, RLR production is strictly limited, because it is prone to decomposition and browning after its short harvest season. In this study an innovative beverage was prepared from full lotus root powder (FLRP) as a substitute for RLR in an attempt to solve this problem.RESULTS: The components of FLRP basically corresponded to those of RLR, but there was some loss of heat-labile compounds. Using differential scanning calorimetry, a gelatinisation temperature range of 57.08-67.80 degrees C was determined for FLRP with an average particle size distribution of 70 mu m. The optimal conditions for enzymatic treatment of FLRP beverage were determined by response surface methodology as an enzyme concentration of 2.2 g kg(-1) at 53 degrees C for 86 min. Turbidity decreased from 1082 to 280 nephelometric turbidity units following enzymolysis. Properties of FLRP beverage were also studied and a qualitative comparison of flavour compounds between RLR and FLRP beverages was made by electronic nose.CONCLUSION: Basic flavour compounds were consistent and flavour radar plots had approximately the same shape, area and proportion when all ingredients were identical apart from FLRP and RLR. Therefore, in terms of flavour, FLRP beverage appears to be a feasible substitute for RLR beverage. (c) 2010 Society of Chemical Industry
Evaluation of Pre-Harvest Nutrient Composition and Functional Active Substances in Various Lotus Roots

Abstract:This study investigated the impact of variety and harvest time on the visual appearance, nutritional quality, and functional active substances of six lotus root cultivars: "Xinsanwu", "Wuzhi No. 2", "Baiyuzhan", "Huaqilian", "Elian No. 6", and "Elian No. 5". Samples were collected monthly from December 2023 to April 2024. A nutrient analysis revealed a decrease in the water content with a delayed harvest. The total soluble solids and soluble sugar content peaked towards the end and middle-to-late harvest periods, respectively. Starch levels initially increased before declining, while the soluble protein exhibited a triphasic trend with an initial rise, a dip, and a final increase. The vitamin C (Vc) content varied across cultivars. Functional active substances displayed dynamic changes. The total phenolics initially decreased, then increased, before ultimately declining again. The total flavonoid content varied by both cultivar and harvest time. The phenolic acid and flavonoid content mirrored the trends observed for total phenolics and total flavonoids. Gastrodin was the most abundant non-flavonoid compound across all varieties. "Wuzhi No. 2" and "Baiyuzhan" displayed higher levels of functional active substances and starch, while the Elian series and "Xinsanwu" cultivar exhibited a greater content of Vc, soluble sugar, and soluble protein. Specific harvest periods yielded optimal results: "Wuzhi No. 2" (H1 and H5), "Huaqilian" (H2), and "Baiyuzhan" (H3 and H4) demonstrated a high nutrient and functional active substance content. Overall, the lotus roots harvested in period H4 achieved the highest score. Overall, this study provides the foothold for the rapid identification of superior lotus root cultivars and the valorization of lotus root by-products via advanced processing methods. Additionally, it offers valuable insights for market participants and consumers to select optimal varieties and harvest times based on their specific needs.

Abstract:Lotus (Nelumbo nucifera), belonging to the family of Nelumbonaceae, is a beautiful aquatic perennial plant. It has been used as an ancient horticulture plant and famous agricultural crop for thousands of years. Modern phytochemical and pharmacological experiments have proved that polysaccharide is one of the most pivotal bioactive constituents of lotus. Hence, the systematic review covering the fundamental research advances and developing prospects of N. nucifera polysaccharides (NNPs) is an urgent demand to provide theoretical basis for their further research and application. The present review summarizes current emerging research progresses on the polysaccharides isolated from lotus, and it focuses on advanced extraction and purification methods, unique structural features, engaging biological activities, potential molecular mechanisms, as well as the relationship of structure and activity of NNPs. This review sheds light on the potential values of NNPs in affording functionally bioactive agents in food industry or therapeutically effective medicines for health care. In addition, this review will provide valuable insights for further commercial product development and promising industrial application of NNPs in both of the fundamental research communities and food or pharmaceutical industries in future.

Abstract:Through the research of color-protecting of lotus root slice, preparing of lotus root pulp milk, steril-ization conditions of lotus root pulp milk, the fermentation liquid preparing technology of lotus root yogurt was determined as follows:using the composite color-protecting stabilizer composed of 0.20%V C, 0.25%citric acid and 0.20%L-cysteine processing fresh lotus root for 30 minutes, mixing the color-protecting lotus root slices and water according to 1∶1.5(g/mL)ratio, adding 0.10%citric acid, beating, mixing lotus root pulp and milk according to 4∶6 (volume ratio), 90℃sterilizing for 15 minutes. Experiments showed that the fermenta-tion liquid of lotus root pulp milk prepared by this technology was milky white, delicate taste and the mixing fra-grance of yogurt and lotus root was fine after inoculation and fermentation.
Comparison among Different Varieties of Lotus Root in Tntegrated Lotus Root-fish Farming System

Abstract:In this paper, several main cultivars of lotus root were cultured in planting-breeding integrated system, then the growth and development, commodity features and nutritional quality of lotus root were studied. The research indicates that, the 5 cultivars used for experiments are suited for integrated cultivation. Elian 5 was suitable for cultivating as processing type lotus root and sale as fresh-eating lotus root for soup with high starch content and good commodity features. Elian 6 has good commodity features with moderate starch content and high levels of crude protein and amino acid, it could replace the old varieties of fresh-eating lotus root. Elian 7 has earlier maturity and good commodity features with moderate starch content, so it could be cultivated as early-maturing variety and saled as early fresh-eating lotus root.(The research screened different lotus root varieties for diverse uses, providing theoretical foundation for sustainable development of local lotus root planting-breeding integrated culture model.)

Abstract:AbstractLotus (Nelumbo nucifera) root has been used as an edible vegetable in East Asia for thousands of years. The present research was aimed to explore the physicochemical, nutritional and microbiological safety of lotus root fermented sugar syrup as a fermented food supplement or condiment for human health benefits. In this study, the physicochemical, nutritional and microbiological safety properties of lotus root syrup fermented with 57° Brix brown sugar at different time periods until 6 months (180 days) was investigated. There was a significant improvement as compared to 57° Brix brown sugar broth (as a control) in the total acceptability and physicochemical properties of lotus root sugar syrup samples such as pH and color improvement. The red color values of 180 days lotus root fermented sugar syrup samples were significantly enhanced (6.85 ± 0.58) when compared with the control (0.20 ± 0.15). In addition, the total protein content was increased from 8.27 ± 0.86 to 392.33 ± 7.19 μg/mL, along with the increase in fermentation time reaching to the level of consumption acceptability. All the lotus root fermented sugar syrup samples were subjected to microbiological analysis. It was found that the coliform, Bacillus cereus, Escherichia coli, Salmonella and Staphylococcus aureus counts were not detected in majority of the samples, confirming the high degree of hygiene processing of lotus root fermented sugar syrup samples for its use as a food supplement or condiment.

Abstract:Lotus is a member of a small family of Nelumbonaceae and is regarded as a sacred flower grown as a freshwater aquatic plant. All plant parts are utilized as food and medicine. Lotus seed (LS) is an important part of the lotus plant due to its nutritional and bioactive components. The presence of phenolic compounds imparts numerous bioactivities to the LS extracts. Nutritional composition is considered crucial for evaluating new ingredients for food applications. Hence, the review deals with the nutritional composition of the LS and bioactivities of LS extracts, viz., anti-cancer, anti-proliferation, anti-diabetic, anti-inflammatory, neuroprotective, antioxidant, and immunomodulatory action. LS also possesses a significant amount of industrially important components which are beneficial for the formulation of food products. The overall profile of the lotus seed makes them potential nutraceutical and bioactive food ingredients. Utilizing this low-cost horticultural waste for producing value-added products is a novel step in its sustainable utilization. The article aimed to comprehensively collate some of the vital information published on lotus seeds incurred in the agri-food sector and critically discussing the potentiality of tapping bioactive compounds from lotus seeds considered as wastes and/or by-products in the entire agriculture-based food sector.

Abstract:This comprehensive review systematically examines the multifarious aspects of Nelumbo nucifera , elucidating its ecological, nutritional, medicinal, and biomimetic significance. Renowned both culturally and scientifically, Nelumbo nucifera manifests remarkable adaptability, characterized by its extensive distribution across varied climatic regions, underpinned by its robust rhizome system and prolific reproductive strategies. Ecologically, this species plays a crucial role in aquatic ecosystems, primarily through biofiltration, thereby enhancing habitat biodiversity. The rhizomes and seeds of Nelumbo nucifera are nutritionally significant, being rich sources of dietary fiber, essential vitamins, and minerals, and have found extensive culinary applications. From a medicinal perspective, diverse constituents of Nelumbo nucifera exhibit therapeutic potential, including anti-inflammatory, antioxidant, and anti-cancer properties. Recent advancements in preservation technology and culinary innovation have further underscored its role in the food industry, highlighting its nutritional versatility. In biomimetics, the unique "lotus effect" is leveraged for the development of self-cleaning materials. Additionally, the transformation of Nelumbo nucifera into biochar is being explored for its potential in sustainable environmental practices. This review emphasizes the critical need for targeted conservation strategies to protect Nelumbo nucifera against the threats posed by climate change and habitat loss, advocating for its sustainable utilization as a species of significant value.
