Abstract:OBJECTIVE To study the optimum extraction parameters and components on ant oil from Polyrhachis vicina. METHOD The optimum condifious for supercritical CO2 fluid extraction (SFE-CO2), were investigated with orthogonal design, GC-MS was applied for analyzing. The components and their contents in the ant oil were analyzed by GC-MS, and the contents of lead, zinc and manganese in the oil were determined by ICP-AES. RESULT The optimum extraction parameters were achieved, temperature of 50 degrees C, pressure of 30 MPa and time of 2 hours. The extracting yield of the ant volatile oil was 11.4% - 14.3%. 51 Constituents were identified including 9-octadecenoic acid, ethyl oleate, cholesterol, n- Hexadecanoic acid, etc, and the content of various constituents was determined by orea normalization. The oil contained unsaturated fatty acid of 64.6%, lead of 0.80 microg x g(-1), zinc of 0.54 microg x g(-1) and manganese of 0.15 microg x g(-1). CONCLUSION The method showes advantages including faster and efficient of extraction, good quality and no solvent residues in the oil.

Abstract:The insecticidal activity of 1,3,4-oxadiazole-ring containing pyridylpyrazole-4-carboxamides against diamondback (Plutella xylostella) and noctuid (Spodoptera frugiperda) moths were evaluated. Only substitutions on the 1,3,4-oxadiazole moiety dictated bioreactivity, with 8q2 m-CF3Ph being the most effective against Plutella xylostella, showing 80%-93% mortality at a concentration of 0.5-500 mu g ml(-1) while the others remained inactive or less active (highest mortality being 63%). The larvicidal activity against Spodoptera frugiperda was inferior compared with that of Plutella xylostella, with the highest mortality being 33%. A crystal of 8q5 was obtained and analyzed, revealing the effects of intramolecular H-bonds and intermolecular pi-pi interactions.

Abstract:Unrestricted reproduction and spread of pest had caused great damage to the quality and yield of crops in recent years. Besides the use of traditional chemical pesticides, natural products also make a huge contribution against pests. Chasmanthinine, a diterpenoid alkaloid isolated from Aconitum franchetii var. villosulum , shown extremely antifeedant activity against Spodoptera exigua . Therefore, a series of novel Chasmanthinine derivatives were synthesized and their biological activity was studied in this work. Compound 33 showed the strongest antifeedant activity (EC 50 = 0.10 mg/cm 2 ) among all the test compounds. The mechanism research of 33 revealed that its antifeedant effect was related to the inhibition of carboxylesterase (CES), and proved the thiophene acyl group could form a strong binding effect with CES by molecular docking. Moreover, compound 10 exhibited the strongest cytotoxicity (IC 50 = 12.87 μM) against Sf9 cell line and moderate contact toxicity. The mechanism research indicated that compound 10 could induce Sf9 cells apoptosis. In summary, the results lay a foundation for the application of diterpene alkaloids in plant protection.

Abstract:In order to develop novel spirocyclic tetronic acid lead compounds, a series of new spirocyclic tetronic acid derivatives containing oxime ether moiety was synthesized and bioassayed. The structures of 16 target com-pounds were characterized by 1H NMR spectra, 13C NMR spectra and high-resolution mass spectrometer(HRMS). Preliminary bioassays indicated that most of the title compounds displayed excellent insecticidal activity against Aphis fabae and Nilaparvata lugens at 100 mg/L. In particular, compound 6k showed the similar activity(LC50= 6.87 mg/L) against A. fabae as the spirotetramat(LC50=4.56 mg/L) and had better effects(LC50=1.64 mg/L) against N. lugens in comparison with spirotetramat(LC50=7.90 mg/L). The study showed that compound 6k exhibited more promising and broad-spectrum insecticide activity and may serve as a new insecticidal agent for sucking pests.

Abstract:To explore insecticides targeting the γ-aminobutyric acid (GABA) receptor, two series of novel isoxazoline derivatives containing sulfonic and carboxylic esters were designed and synthesized. Their insecticidal activities against Plutella xylostella, Mythimna separata, and Aedes aegypti larvae and their structure-activity relationship were investigated. The sulfonate-containing isoxazoline derivatives (10k-q) exhibited promising insecticidal activities against the three insect larvae. Compound 10o displayed excellent activities with LC50 values of 8.32, 5.23, and 0.35 μg/mL at 48 h against P. xylostella, M. separata, and A. aegypti larvae, respectively, which were better than or similar to those of avermectin. Furthermore, compound 10o exhibited a faster insecticidal effect than avermectin against M. separata. The mode of action of 10o was preliminarily verified by molecular docking, theoretical calculations, and measurement of glutamate decarboxylase and glutamic pyruvic transaminase activities. Compound 10o is a novel insecticidal candidate acting on GABA receptors, which could guide the discovery of isoxazoline insecticides.

Abstract:A series of 4-phenyl-acyl-substituted 3-(2,5-dimethylphenyl)-4-hydroxy-1-azaspiro[4.5]dec-3-ene-2,8-dione derivatives were designed and synthesized, and their structures were characterized using (1)H NMR (or (13)C NMR), mass spectrometry, and elemental analysis. The bioactivities of the new compounds were evaluated. These compounds exhibited good inhibition activities against bean aphids (Aphis fabae) and carmine spider mite (Tetranychus cinnabarinus), and 4-phenyl acyl esters showed stronger bioactivity than 4-arylesterases and alkyl esters. The results showed that compound 8-I-e, which contains a para-methoxy group on the phenyl acyl, and compound 8-I-m, which contains a para-trifluoromethyl group on the phenyl acyl, displayed potent insecticidal activity against A. fabae and T. cinnabarinus respectively. The insecticidal activity showed a clear structure-activity relationship, confirming the importance of the flexible bridge. The DFT/B3LYP/6-31(d) level method was used to calculate molecular geometries and electronic descriptors. These factors included total energy, charge distribution, and the linear orbital level of the title compounds. Quantitative structure-activity relationship studies were performed on these compounds using quantum-chemical and physicochemical parameters as independent variables and insecticidal activity as a dependent variable. Insecticidal activity was most closely correlated (r > 0.8) with quantum chemical and physicochemical parameters.
Flavonoids from the Leaves of Sabina vulgaris Antoine

Abstract:Chemical constituents in the leaves of Sabina vulgaris Antoine were studied.Nine flavonoids were isolated and purified by extraction and different kinds of column chromatography,and their structures were elucidated as cupressuflavone(Ⅰ),cupressuflavone 4',4-dimethyl ether(Ⅱ),amentoflavone(Ⅲ),podocarpusflavone A(Ⅳ),catechin(Ⅴ),quercetin(Ⅵ),isoscutellarein 7-O-β-D-xylopyranoside(Ⅶ),myricetin 3-O-β-D-glucoside(Ⅷ),rutin(Ⅸ).CompoundsⅠ,Ⅱ,Ⅴ,Ⅶ,Ⅷ and Ⅸ were obtained from this plant for the first time.Compounds Ⅷ and Ⅸ were isolated from Sabina genus for the first time.All of isolated compounds were tested for their antioxidative activities by 1,1-diphenyl-2-picrylhydrazyl free radical(DPPH·)scavenging assay.It was found that compounds Ⅴ and Ⅵ showed antioxidative activity close to that of positive control,L-ascorbic acid,and the flavonoids glycosides(Ⅶ,Ⅷ and Ⅸ)are more active than the biflavones(Ⅰ,Ⅱ,Ⅲ and Ⅳ).

Abstract:BACKGROUND:Anthranilic diamide derivatives are among the most important classes of synthetic insecticides. Moreover, the 1,2,4-oxadiazole heterocycle, a bioisostere of amide, has been extensively used in pesticides. In order to discover novel molecules with high insecticidal activities, a series of anthranilic diamide analogues containing 1,2,4-oxadiazole rings were designed and synthesised.RESULTS:A series of novel anthranilic diamide derivatives containing 1,2,4-oxadiazole were obtained, and confirmed by 1 H and 13 C nuclear magnetic resonance and high-resolution mass spectrometry. The structure of 3-bromo-N-(4-chloro-2-methyl-6-{3-[(methylsulphonyl)methyl]-1,2,4-oxadiazol-5-yl}phenyl)-1-(3-chloropyridin-2-yl)-1H-pyrazole-5-carboxamide was further characterised by X-ray diffraction analysis. In addition, bioassays showed that most of the newly synthesised compounds displayed 100% mortality against Plutella xylostella at 100 mg L-1 , and compound 3IIl showed 90% larvicidal activities at a concentration of 0.5 mg L-1 . The LC50 value of 3IIl was 0.20 mg L-1 , which indicated that it may be used as a potential leading compound for further structural optimisation. Furthermore, brief comparative molecular field analysis (CoMFA) models were established to study the structure-activity relationships (SARs) of the title compounds.CONCLUSION:Compound 3IIl may be used as a potential leading compound for further structural optimisation, and SARs and CoMFA models could provide reliable clues for further structural optimisation. © 2016 Society of Chemical Industry.
Synthesis, insecticidal activities and structure-activity relationship study of dual chiral sulfilimines

Abstract:To investigate the "methyl" impact on bioactivity of sulfiliminyl dicarboxamides, a total of 16 novel N-cyano and N-trifluoroacetyl sulfiliminyl dicarboxamides containing m-heptafluoroisopropylated aromatic amino moiety were studied. Two series of sulfiliminyl substituents were designed, synthesized and evaluated against oriental armyworm (Pseudaletia separata Walker) for their insecticidal activities. Their chemical structures were established by corresponding [Formula: see text] NMR, HRMS and optical polarimetry. Bioassay results revealed that some of the title compounds showed potent insecticidal activities against oriental armyworm. Notably, compounds IIa, IIIa, IVa exhibited 100% activity at [Formula: see text], in particular, IIa showed a comparable control efficacy to that of the commercial product flubendiamide. The SAR of these N-cyano sulfiliminyl isomers can be summarized as follows (Sc, Ss) [Formula: see text] (Sc, Rs), while the N-trifluoroacetyl sulfiliminyl isomers is (Sc, Rs) [Formula: see text] (Sc, Ss). Comparative molecular field analysis indicated that an electropositive substituent, [Formula: see text] group in the benzene ring was very important for the improvement in biological activity. These results could hold promise for novel chiral sulfiliminyl RyR regulators.
Insecticidal Activities of Sophora flavescens Alt. towards Red Imported Fire Ants (Solenopsis invicta Buren)

Abstract:The red imported fire ant (Solenopsis invicta) is a worldwide invasive and dangerous insect that is controlled mainly by chemical insecticides. Plant-derived insecticidal compounds are generally better than synthetic insecticides for environmental compatibility and the biosafety of non-targets. The toxicity of the ethanol extract of Sophora flavescens roots against S. invicta was evaluated under laboratory conditions. The ethanol extract showed toxicity against minor and medium workers of S. invicta with 7-day LC50 values of 1426.25 and 2292.60 mg/L, respectively. By bioactivity-directed chromatographic separations using the minor worker as the test insect, two active compounds, matrine and sophocarpine, were isolated from the S. flavescens total alkaloids; their chemical structure was identified by 13C NMR data. Matrine showed toxicities against minor and medium workers with 7-day LC50 values of 46.77 and 71.49 mg/L, respectively, and for sophocarpine, 50.08 and 85.87 mg/L, respectively. The two compounds could substantially reduce the foraging response, food consumption, and aggregation of S. invicta workers at a sublethal concentration of 15 mg/L. The present research suggests that S. flavescens roots have potential as a natural control agent for red imported fire ants.

Novel Insecticidal Butenolide-Containing Methylxanthine Derivatives: Synthesis, Crystal Structure, Biological Activity Evaluation, DFT Calculation and Molecular Docking

Abstract:The design of novel agrochemicals starting from bioactive natural products is one of the most effective ways in the discovery and development of new pesticidal agents. In this paper, a series of novel butenolide-containing methylxanthine derivatives (Ia-Ir) were designed based on natural methylxanthine caffeine and stemofoline, and the derivatized insecticide flupyradifurone of the latter. The structures of the synthesized compounds were confirmed via 1H-NMR, 13C NMR, HRMS and X-ray single crystal diffraction analyses. The biological activities of the compounds were evaluated against a variety of agricultural pests including oriental armyworm, bean aphid, diamondback moth, fall armyworm, cotton bollworm, and corn borer; the results indicated that some of them have favorable insecticidal potentials, particularly toward diamondback moth. Among others, Ic and Iq against diamondback moth possessed LC50 values of 6.187 mg ⋅ L-1 and 3.269 mg ⋅ L-1, respectively, - 2.5- and 4.8-fold of relative insecticidal activity respectively to that of flupyradifurone (LC50=15.743 mg ⋅ L-1). Additionally, both the DFT theoretical calculation and molecular docking with acetylcholine binding protein were conducted for the highly bioactive compound (Ic). Ic and Iq derived from the integration of caffeine (natural methylxanthine) and butenolide motifs can serve as novel leading insecticidal compounds for further optimization.
Study on the Antifungal Components in the Root of Elsholtzia bodinieri Vaniot

Abstract:Three antimicrobial active components were isolated from the root of Elsholtzia bodinieri V.by bioactivity guided method.On the basis of spectral analysis and comparision with literature,their structures were identified as ludongnin 5(Ⅰ),quercetin(Ⅱ) and luteolin(Ⅲ),respectively.Among them,compoundⅠwas obtained from Elsholtzia genus for the first time.The antimicrobial experiments indicated that compoundsⅠ～Ⅲ showed the inhabitory and bactericidal activity against Bacillus subtilis,Staphylococcus aureus and Escherichia coli in varying degrees.The scientific foundations were provided more further to exploit and utilize this plant by the studying.
Sulfonate derivatives bearing an amide unit: design, synthesis and biological activity studies

Abstract:Pest disasters which occurs on crops is a serious problem that not only cause crop yield loss or even crop failure but can also spread a number of plant diseases.Sulfonate derivatives have been widely used in insecticide and fungicide research in recent years. On this basis, a series of sulfonate derivatives bearing an amide unit are synthesized and the biological activities are evaluated. The bioassay results showed that compounds A 8 , A 13 , A 16 , B 1 , B 3 , B 4 , B 5 , B 10 , B 12 − 20 , C 3 , C 5 , C 9 , C 10 , C 14 , C 15 , C 17 and C 19 showed 100% activity at a concentration of 500 μ g/mL against the Plutella xylostella ( P. xylostella ). Among them, B 15 which contains a thiadiazole sulfonate structure still shows 100% activity at 50 μ g/mL concentration against P. xylostella and had the lowest median lethal concentration (LC 50 ) (7.61 μ g/mL) among the target compounds. Further mechanism studies are conducted on compounds with better insecticidal activity. Molecular docking results shows that B 15 formed hydrophobic interactions π-π and hydrogen bonds with the indole ring of Trp532 and the carboxyl group of Asp384, respectively, with similar interaction distances or bond lengths as those of diflubenzuron. Moreover, chitinase inhibition assays are performed to further demonstrate its mode of action. In addition, the anti-bacterial activity of the series of compounds is also tested and the results showed that the series of compounds has moderate biological activity against Xanthomonas oryzae pv. oryzae ( Xoo ) and Xanthomonas oryzae pv. oryzicola ( Xoc ), with inhibition rates of 91%, 92% and 92%, 88% at the concentration of 100 μ g/mL, respectively. Our study indicates that B 15 can be used as a novel insecticide for crop protection.

Abstract:A series of anthranilic diamides analogs (3-11, 16-24) containing 1,2,4- or 1,3,4-oxadiazole rings were synthesized and characterized by H-1 NMR, MS and elemental analyses. The structure of 3-bromo-N-(2-(3-(4-bromophenyl)-1,2,4-oxadiazol-5-yl)-4-chloro-6-methylphenyl)-1-(3-chloropyridin-2-yl)-1H-pyrazole-5-carboxamide (18, CCDC-924454) was determined by X-ray diffraction crystallography. The insecticidal activities against Plutella xylostella and Spodoptera exigua were evaluated. The results showed that most of title compounds displayed good larvicidal activities against P. xylostella, especially compound 3-bromo-N-(4-chloro-2-methyl-6-(5-(methylthio)-1,3,4-oxadiazol-2-yl)phenyl)-1-(3-chloropyridin-2-yl)-1H-pyrazole-5-carboxamide (6), which displayed 71.43% activity against P. xylostella at 0.4 mu g mL(-1) and 33.33% against S. exigua at 1 mu g mL(-1). The structure-activity relationship showed that compounds decorated with a 1,3,4- oxadiazole were more potent than compounds decorated with a 1,2,4-oxadiazole, and different substituents attached to the oxadiazole ring also affected the insecticidal activity. This work provides some hints for further structure modification and the enhancement of insecticidal activity.
Studies on the Inhibitory Activity of Extracts of Chinese Herbs to Sclerotinia Sclerotiorum in Rape and Active Components in Syzygium Aromaticum L.

Abstract:[Objective] The aim of the research was to analyze the antifungal components of Chinese herb extracts and provide basis for the development of new types of plant pesticides.[Method] With the ethanol extracts from 14 kinds of Chinese herbs including Syzygium aromaticum L.as materials,the inhibitory activities of all kinds of extracts to Sclerotinia sclerotiorum in rape were determined.The chemical components in the extracts from S.aromaticum were analyzed by GC-MS method and its effects on the antifungal activity to S.sclerotiorum in rape and the mycelium dry weight of S.sclerotiorum were determined.[Result] The results showed that the inhibitory effects of the extracts from S.aromaticum on the mycelium growth of S.sclerotiorum in rape were better and the inhibitory rate was 100%.[Conclusion] Eugenol was preliminarily verified to be main antifungal component in the extracts from S.aromaticum.
Synthesis and biological activities of N-[3-cyano-1-(2,6-dichloro-4- trifluoromethylphenyl)-1H-pyrazol-5-yl]-chrysanthemumamide

Abstract:In order to find new semicarbazide lead compounds, ten N-[1-(2,6-dicholoro- 4-trifluoromethylphenyl)-1H-pyrazol-5-yl]-chrysanthemumamides were synthesized from five types of chrysanthemic acids with fipronil or 5-amino-1-(2,6-dichloro- 4-trifluoromethylphenyl)-1H-pyrazol-3-carbonitrile. The structures of all new compounds were confirmed by 1H NMR, MS spectra and elemental analysis. At the concentration of 37.5 mg/L, compound 3b shows inhibitory activity (100%) against Plutella xylostella Linnaeus. The LC 50 of this compound is 15.1 mg/L. The oral acute toxicity of this compound to big male and female rats is middle. The crystal structure of 3i is also presented. The results indicate that it is rational and feasible to find new active compounds based on the connection between the active compound and the pharmacophore.

Abstract:A series of 2′(2′,6′)-(di)halogeno-isoxazolopodophyllic acids-based esters, and oxime sulfonates of 2′(2′,6′)-(di)halogenopodophyllones were prepared by structural modifications of podophyllotoxin as insecticidal agents against Mythimna separata Walker. It was found that when 2′(2′,6′)-(di)halogenopodophyllones or 2′(2′,6′)-(di)chloropicropodophyllones reacted with hydroxylamine hydrochloride, the desired products were related with the configuration of their lactones. Three key single-crystal structures of Ie, IIe and IIIb were determined by X-ray diffraction. Especially compounds IIc and Vc showed the highest insecticidal activity. Moreover, some interesting results of structure-insecticidal activity relationships of tested compounds were also observed.

Abstract:BACKGROUNDAnthranilic diamide insecticides containing pyridylpyrazole-5-carboxamide are extremely important in modern agriculture. New structurally modified compounds with high insecticidal activity were discovered by designing a series of novel pyridylpyrazole-4-carboxamides (9I to 9IV) and pyridylpyrazole-4-carboxamides (10I to 10IV), the latter designed by the cyclisation of two amides. The structure-activity relationship (SAR) between the two series is of interest.RESULTSTwo series of novel anthranilic diamides containing pyridylpyrazole-4-carboxamide were synthesised and characterised via melting point, H-1 NMR, C-13 NMR, MS and elemental analyses. The insecticidal activities of these compounds against Plutella xylostella were evaluated. At a concentration of 100mgL(-1), the compounds with unmodified amide moieties (9I to 9IV) exhibited much better larvicidal activities than the other derivative compounds (10I to 10IV). Most of the compounds 9I to 9IV showed over 90% larvicidal activity at 100mgL(-1). Furthermore, compounds 9IIIa, 9IIIc, 9IIId and 9IVd displayed significant insecticidal activity at 10mgL(-1). Density functional theory (DFT) calculation was carried out to provide more information regarding SAR.CONCLUSIONThirty-two new anthranlic diamides containing pyridylpyrazole-4-carboxamide were designed and obtained. SAR analysis and DFT calculation results revealed that the amide moiety had a very important effect on bioactivity. Abstract:A guanidine compound was isolated and identified from Buthus martensii Karsch. Based on its characteristic structure, we used it as a lead compound to carry out structural optimization and subsequent fungicidal evaluation. Here 18 guanidine derivatives were designed and synthesized by a facile method, and their structures were characterized by 1 H, 13 C NMR, and HRMS. Biological assay indicated that most of the title compounds possess potential fungicidal activities against Alternaria altanata , and Penicillium digitatum . Especially, 5a , 5d , and 5i showed inhibition rates (IR) of 41.9, 41.9, and 44.2% respectively against P. digitatum at a concentration of 25 mg/L, whose activities surpassed that of hymexazol. This is the first report on the active derivatives from B. martensii in application of plant protection, providing scientific references for the development and utilization of active ingredients from scorpions.

Abstract:A series of anthranilic diamides analogs (5a-x) containing 1,3,4-oxadiazole rings were synthesized and characterized by H-1 NMR, C-13 NMR and mass spectrometry. The structure of N-(4-chloro-2-methyl-6-(5-(thiophen-2-yl)-1,3,4-oxadiazol-2-yl) phenyl) methacrylamide (5u) was determined by X-ray diffraction crystallography. The insecticidal activities of these new compounds against diamondback moth (Plutella xylostella) were evaluated. Preliminary bioassays indicated that some of these compounds exhibited good insecticidal activities against P. xylostella, especially 3-bromo-N-(4-bromo-2-methyl-6-(5(pyrazin-2-yl)-1,3,4-oxadiazol-2-yl) phenyl)-1-(3-chloropyridin-2-yl)-1H-pyrazole-5-carboxamide (5d), which displayed 100%, 80.95% and 57.14% activity against P. xylostella at 40 mu g mL(-1), 10 mu g mL(-1) and 4 mu g mL(-1), respectively. The relationship between structure and insecticidal activity was discussed. The density functional theory (DFT) studies could be helpful to understand the various insecticidal activities.
