Effects of Developmental Exposure to BDE - 47 on the Thyroid Hormone Homeostasis and Antioxidant System of Glutathione

Abstract:The present paper is aimed to introduce its authors' experimental findings of the effects of developmental exposure to BDE-47 on the thyroid hormone homeostasis and antioxidant system of glutathione. In conducting our experiments, new-born three-day old baby Sprague-Dawley rats were let to be exposed to different doses for 21 days to test the thyroid hormone disruptive effects and the glutathione related oxidative stress. The results of our experiments show that compared with the control group, no significant changes were found of the total thyroxine and free thyroxine (FT4) in the rat serum. Thus, a decrease trend of the total 3,3' ,5-triiodothyronine was noted after the big dose exposure though not statistically significant. However, free 3,3' ,5-triiodothyronine (FT3) in rat serum of high-dose exposures tends to decrease significantly (p 0.01) when compared with the control group, thus suggesting that FT3 is likely to be the most sensitive subject to the thyroid disruption of BDE-47. The experimental results of induced oxidative stress also indicate that 21-day exposure, though statistically insignificant, reveals an overall downward tendency of the activity of glutamate-cysteine ligase (GCL), hence proving the inhibition on the GCL as a result of different doses exposure. As for the index of reduced glutathione (GSH) , no significant changes were detected in any of the exposure groups when compared with the control group. Nevertheless, the variation trend of glutathione peroxidase (GSH-Px) remains similar as GSSG, where a significant increase was found after the exposure of 100 μg/kg BDE-47 (p 0.01) but drop to the level of controls after the high dose exposure. Therefore, this study claims to find that the developmental exposure of BDE-47 has resulted in the imbalance of thyroid homeostasis, especially for the index of FT3, which may also lead to a disorder of GSH red-ox system.
