Comparative Studies on Rdna-Its Region of Intrapopulations of Heterodera Glycines

Abstract:The rDNA-ITS region of ten soybean cyst nematode (SCN), Heterodera glycines, populations from different soybean production areas of China and one population from USA were amplified. All populations gets the same length fragment, i.e. approximately 1000bp. The fragments were digested with five restriction enzymes, AluⅠ,CfoⅠ,MvaⅠ,RsaⅠ and AvaⅠ, of which, AluⅠ,CfoI, MvaI and RsaⅠ, produced restriction profiles identical among all populations, while AvaⅠyielded two phenotypes in the population from Taigu, Shanxi. The sequence alignment showed that the ITS1 region and 5.8S RNA gene are all identical among all the populations, while only three base pare differences were found in ITS2 region. The results indicated that rDNA-ITS region is relatively stable among the intrapopulations of Heterodera glycines.
