Abstract:AIM: To observe the combined cultivation of pig iliac endothelial cell (PIEC) and small intestinal submucosa (SIS), and verify whether the small intestinal submucosa could serve as a good support material for tissue engineered vessels.METHODS: From September 2005 to January 2006, the preparation and disposal of SIS and combined cultivation of PIEC and SIS were conducted in the Institute of Biomedical Engineering, School of Life Science and Technology, Tongji University; meanwhile, the radial irradiation and disinfection for SIS was conducted at the Cancer Center of Zhongshan Hospital Fudan University. Materials included the SIS, which was from the fresh pig jejunal harvested from the slaughter house; PIEC, which was provided by the cell bank of Shanghai Institute of Cell Biology, Chinese Academy of Sciences. There were two groups in the experiment: PIEC group and PIEC-SIS group. ①The SIS was processed by physical and chemical methods; ②PIECs were cultured alone; ③SIS and endothelial cells were co-cultured in order to observe the cell attachment and growth, and the structure of SIS with inverted phase contrast microscope. The cell growth curves of PIEC and PIEC-SIS were determined at the same time; three-well cultured cells were counted after digestion everyday for 8 days to evaluate the biological compatibility of PIEC and SIS.RESULTS: ①The superficial shape of SIS: The submucosa was a white semitransparent film, about 80 μm. Under the inverted microscope, the structures of two sides were different after the disposal: one side was rough and big pore diameter, just could be used to plant cells, while the other side was smooth. There were a lot of cell fragments on the submucosa after the physical disposal, then the cell fragments disappeared after the chemical disposal, and the collagen fibers were not damaged. ②Comparison of the cell shape between simple culture and co-culture with SIS: The inverted microscope showed that a majority of PIECs adhered to SIS in 24 hours, and the proliferation of cell was not obvious from the first day to the fourth day. A lot of cells glued to the margin of the material on the fifth day directly. The cells distributed in the SIS evenly and grew well in the material with the time. The PIECs co-cultured with SIS were polygon or fusiform. Compared with the cells cultured solely, the pseudopods of co-cultured cells were not obvious and the shape of cells was regular relatively. ③Cell growth curve: The cell count of combined cultivation group was less than the sole cultivation group in the early four days, but more from the fifth day. The number of cells in the combine cultivation group was the least in the next day after inoculation, and cell counts of two groups reached the peak values at the last day.CONCLUSION: PIEC could attach to the SIS and grow well; SIS accelerates the proliferation of the cells. SIS shows a good compatibility and its pore diameter and structure are beneficial to the attachment and growth of endothelial cells, therefore, SIS is a good bio-derived material in tissue engineering.
