Abstract:Objective To study the impact of single nucleotide polymorphism 43 (UCSNP43) of calpain 10 gene (CAPN 10) at NIDDM1 locus, chromosome 2q37.3 on glucose intolerance status and its intermediate traits. Methods The studied population consisted of 320 Chinese subjects in Shanghai 〔normal glucose tolerance (NGT) 148, impaired glucose tolerance (IGT) 44 and type 2 diabetes (T2DM) 128〕. Plasma glucose, insulin, C peptide and free fatty acid levels were measured during fasting and 30, 60, 120 and 180 min after oral 75 g glucose load. The tissue insulin sensitivity and islet beta cell insulin secretion were assessed by HOMA formulae and the ratio of the increment of insulin and glucose levels at 30 min. The CAPN 10 UCSNP43 was genotyped by DNA direct sequencing. Results (1) The frequencies of GG, GA and AA genotypes of CAPN 10 UCSNP 43 were 0.80, 0.18 and 0.02 respectively. The G allele frequency was 0.89, which was statistically significant higher than those in Caucasians, Mexican Americans and Pima Indians (all P0.0001) and similar to that in Japanese reported in the literatures. (2) CAPN 10 UCSNP43 was not related to glucose intolerance status (IGT and/or T2DM). (3) Among the NGT subjects, the tissue insulin sensitivity was lower in CAPN 10 UCSNP GA+AA subgroup than that in GG subgroup 〔HOMA IR P=0.006, afterbeingadjustedwithage, sex, bodymass index (BMI) and waist circumference, P=0.017〕. Moreover, the insulin levels at 0, 60, 120, 180 min and area under curve after glucose load weresignificantlyhigherinGA+AAsubgroupthan those in GG subgroup (P=0.019, 0.016, 0.007, 0.039 and 0.009 respectively, after being adjusted with age, sex, BMI, waist circumference and related timing glucose level, P=0.026, 0.024, 0.020, 0.112 and 0.011 respectively). Conclusion CAPN 10 UCSNP43 is associated with tissue insulin sensitivity and insulin levels during glucose load.
