Regulation function of cytochrome c oxidase on aging: research advances

Abstract:Cytochrome c oxidase(COX),located on the mitochondrial inner membrane,is the terminal enzyme of mitochondrial respiratory chain.It is the only enzyme that can directly make contact with oxygen generated from the respiratory chain complexes.About 90% of oxygen in human cells and tissues can be processed by COX,indicating its leading role and importance in oxidative metabolism and ATP synthesis.Multiple studies have shown that lowered COX activity,impaired mitochondrial function,and over-production of reactive oxygen species were closely related to aging and neurodegenerative diseases.This review summarizes the new development of COX regulatory mechanism on aging,including the research of COX subunits deficiency,COX-mediated oxidative damage and combination with biological macromolecules through the COX structural analysis.In conclusion,we demonstrate the regulative functions of COX on aging and its related diseases may provide new strategies for the treatment of aging-related diseases.
