The effect of atypical antipsychotics on glutamte-deficiency-induced behaviors in Apella Cebus monkeys

Abstract:Objective To study the efficacy of atypical antipsychotics in treating MK 801 induced hypoglutamatergic behavioral symptoms in six Cebus Apella monkeys Methods Animals were injected subcutaneously MK 801(0 06 mg/kg), clozapine (1, 3 and 5 mg/kg), olanzapine (0 02, 0 04 and 0 08 mg/kg), quitapine (1, 3 and 5 mg/kg), and coadministration of the three dosages of the three different atypical antipsychotics with the same dosage of MK 801 by turns Animal′s behaviors were evaluated with a 7 point scale Results High dose of olanzapine ameliorated decreased locomotion and bradyskinesia, reversed salivation without the dystonia Three doses of quetiapine antagonised significantly the decreased locomotion and salivation, high dose of quetiapine reversed the dyskinesia also, while induced the tardive dystonia Clozapine′s effects was complicated, low dose of clozapine minimized the decreased locomotion, though any effects of high dose were possibly masked by increased ataxia and salivation Conclusion Three atypical antipsychotics have varying efficacy on the behaviors induced by glutamatergic hypofunction, based upon their unique pharmacology in the brain
