Abstract:Objective: To investigate the effect of T cell vaccination in prevention of allograft rejection in clinical renal transplantation. Methods: Donor antigen (peripheral white blood cells)-primed renal recipients` lymphocytes were obtained by the method of density gradient centrifugation. The antigen primed-lymphocytes were activated, proliferated and attenuated in vitro for the preparation of T cell vaccine (TCV). TCV was intravenously into the renal recipients before transplantation. Results: Of the 6 cases of renal recipients who received T cell vaccination, 1 case had a fever during the injection of TCV, no acute rejection was observed. T lymphocyte proliferation induced by PHA was increased in the vaccinated patients. The mixed lymphocyte reaction and the antibody-mediated cytotoxicity were not affected by TCV. Conclusions: It is feasible to applicate T cell vaccine to clinical renal transplantation as a preventive for allograft rejection.
