Abstract:Objective To investigate the possibility of reinnervation of denervated skeletal muscle by transplantation of neural stem cells. Methods One hundred and eight SD rats were randomly divided into 3 groups: with 36 in each. The model of transection of tibial nerve at the right side was set up. In the experimental group, neural stem cells were injected into the distal stump of the tibial nerve. In the control group, the same dose of cell culture media was used. In the injury group, the same dose of normal saline was used. At 8, 10 and 12 weeks postoperatively, the reinnerivation of denervated skeletal muscle was determined by using the techniques of immunohistochemistry, horseradish peroxidase(HRP) retrograde tracing and image analysis. Results At 8 weeks after transplantation, the differentiated neurons and glial cells were detected. The synaptic connection between regenerated axon and targeted muscle was reestablished.
