The Hepatoprotective Effect of Polyphenols from Sphallerocarpus gracilis Stem Leaves against CCl4-induced Acute Hepatic Damage in Mice

Abstract:The aim of this study was to investigate the protective effect of polyphenols from Sphallerocarpus gracilis stem leaves on carbon tetrachloride(CCl4)-inducedhepatic injuries, and to identify its effective dosage. The mice in the treatment group were administrated with the polyphenols from Sphallerocarpus gracilis stem leaves at doses of 150, 300, and 600mg/kg body weight(BW)/day for 21 consecutive days. The liver injury model was established by intraperitoneal injection of 3 m L 1% CCl4 rapeseed oil(V/V). The alanine transaminase(ALT), aspartate transaminase(AST), total cholesterol(TC), total triglyceride(TG), total bilirubin(TBIL) contents in serum, and superoxide dismutase(SOD), malondialdehyde(MDA), and catalase(CAT) levels in liver were measured. Meanwhile, ahepatic pathological examination was conducted. The results of this study showed that the polyphenols from Sphallerocarpus gracilis stem leaves could, to some extent, protect mice from CCl4-induced liver injury. In addition, these polyphenols significantly prevent the increase in liver index spleen index, serum ALT, AST, TBIL, and liver MDA levels, caused by CCl4 in mice. These polyphenols could also markedly increase the activities of liver SOD and CAT. Among the tested doses, 300mg/kg Bw·d showed the best effect. Histopathological examinations further confirmed these results.
