Abstract:Chymosin(rennin;EC 3.4.23.4) belongs to a aspartic proteinase family which was found in unweaned bovine stomachs at first.It can specifically cleave the κ-casein at the Phe10 5-Met10 6 peptide bond to broke the casein micelles causing milk-clotting.In cheese produc-tion,chymosin plays an important role as the key enzyme in milk-clotting and processing of texture and flavour.The structure,physicochemi-cal characteristics and milk coagulation mechanism of bovine chymosin were reviewed in this paper.Otherwise,the source of chymosin and the diversity from different sources were discussed.

Abstract:Caseins with known sequences were simulated peptide-cut into small peptides by 39 kinds of proteases using the software of peptidecutter,and the suitable effective proteases were chosen in terms of the amount of oligo-peptides and enzymatic peptide library was established.The most effective protease was determined in the confirmatory experiment.Angiotensin Ⅰ-converting enzyme inhibitory peptides(ACEIPs) and CPPs were obtained after ultra-filtration,and the inhibitory activity of ACEIP and the N/P ratio of CPP were used to assess the activity of ACEIP and the purity of CPP.The results implied that:the trypsin was chosen as the most suitable protease.Compared with the mixture before ultra-filtration,the activity of ACEIP and the purity of CPP were extremely significant improved(p<0.01).In this way,higher activity of ACEIPs and higher purity of CPPs were obtained.
Advances in research on calf rennet substitutes and their effects on cheese quality

Abstract:Milk coagulation is an important step in cheese production, and milk-clotting enzymes (MCEs) play a major role in this process. Calf rennet is the most widely used MCE in the cheese industry. The use of calf rennet substitutes is becoming necessary due to the limited availability of calf rennet and the increase in cheese consumption. The objective of this review is to summarize the latest findings on calf rennet substitutes (animal MCEs, plant-derived MCEs, recombinant MCEs and microbial MCEs) and their application in cheese production. Special emphasis has been placed on aspects of the effects of these substitutes on hydrolysis, functional peptides, cheese variety and cheese yield. The advantages and disadvantages of different calf rennet substitutes are discussed, in which microbial MCEs have the advantages of less expensive production, greater biochemical diversity, easier genetic modification, etc. In particular, some of these MCEs have suitable characteristics for cheese production and are considered to be the most potential calf rennet substitutes. Moreover, challenges and future perspectives are presented to provide inspiration for the development of excellent calf rennet substitutes.
Production and evaluation of enzyme-modified cheese adding protease or lipase to improve quality properties

Abstract:Enzyme-modified cheese (EMC) produced by enzyme hydrolysis is a natural, cost-effective, and flexible alternative to using natural cheese in industrial applications. The modification of cheese by enzymes can increase their benefits for consumer acceptance and health, and intensify the specific cheese flavor. We evaluated the properties of cheese with added protease (Ep) or lipase (El), including texture, sensory, organic acids, volatile compounds, and free amino acids. As results, the hardness and gumminess of the cheese reached their maximum values when the concentration of protease and lipase was 0.1% and 0.6%, respectively. Interestingly, the bitterness and astringency of the cheese was reduced. The highest scores for odor, taste, and overall acceptability were observed on 0.08% protease in Ep and 0.8% lipase in El. Compared with the anchor cheese, eight new compounds were produced after the addition of protease and nine new compounds were produced after the addition of lipase. Irrespective of the type of enzyme, the content of free amino acids decreased slightly with the increase in enzyme content. From the point of view of adding enzyme species, the free amino acids content of Ep was generally higher than that of El, and glutamic acid and proline contents were high. Acetic acid concentrations (aroma-active compounds) of enzyme-modified cheese using protease and lipase were 482-931 mg/100 g and 30-36 mg/100 g, respectively, which were significantly increased. According to the results obtained in this study, a cheese with higher sensorial and textural acceptability was obtained by adding the appropriate protease or lipase.
Isolation and identification of milk-clotting proteases from Prinsepia utilis Royle and its application in cheese processing

Abstract:The aim of this study was to isolate and identify the main milk-clotting proteases from Prinsepia utilis Royle. Protein isolates obtained using precipitation with 20 %-50 % ammonium sulfate (AS) showed higher milk-clotting activity (MCA) at 154.34 + 0.35 SU. Two milk-clotting proteases, namely P191 and P1831, with molecular weight of 49.665 kDa and 68.737 kDa, respectively, were isolated and identified using liquid chromatography-mass spectrometry (LC-MS/MS). Bioinformatic analysis showed that the two identified milk-clotting proteases were primarily involved in hydrolase activity and catabolic processes. Moreover, secondary structure analysis showed that P191 structurally consisted of 40.85 % of alpha-helices, 15.96 % of beta-strands, and 43.19 % of coiled coil motifs, whereas P1831 consisted of 70 % of alpha-helices, 7.5 % of beta-strands, and 22.5 % of coiled coil motifs. P191 and P1831 were shown to belong to the aspartic protease and metalloproteinase types, and exhibited stability within the pH range of 4-6 and good thermal stability at 30-80 °C. The addition of CaCl2 (<200 mg/L) increased the MCA of P191 and P1831, while the addition of NaCl (>3 mg/mL) inhibited their MCA. Moreover, P191 and P1831 preferably hydrolyzed kappa-casein, followed by alpha-casein, and to a lesser extent beta-casein. Additionally, cheese processed with the simultaneous use of the two proteases isolated in the present study exhibited good sensory properties, higher protein content, and denser microstructure compared with cheese processed using papaya rennet or calf rennet. These findings unveil the characteristics of two proteases isolated from P. utilis, their milk-clotting properties, and potential application in the cheese-making industry.

Preparation of pH-responsive chitosan microspheres containing aminopeptidase and their application in accelerating cheese ripening

Abstract:Microencapsulated enzymes have been found to effectively accelerate cheese ripening. However, microencapsulated enzyme release is difficult to control, often resulting in enzyme release during cheese processing and causing texture and flavor defects. This study aims to address this issue by developing aminopeptidase-loaded pH-responsive chitosan microspheres (A-CM) for precise enzyme release during cheese ripening. An aminopeptidase with an isoelectric point (pH 5.4) close to the pH value of cheese ripening was loaded on chitosan microspheres through electrostatic interaction. Turbidity titration measurements revealed that pH 6.5 was optimal for binding aminopeptidase and microspheres, affording the highest loading efficiency of 58.16%. Various characterization techniques, including scanning electron microscopy, energy-dispersive X-ray spectroscopy, and Fourier-transform infrared spectroscopy confirmed the successful loading of aminopeptidase molecules on the chitosan microspheres. In vitro release experiments conducted during simulated cheese production demonstrated that aminopeptidase release from A-CM was pH responsive. The microspheres retained the enzyme during the coagulation and cheddaring processes (pH 5.5-6.5) and only released it after entering the cheese-ripening stage (pH 5.0-5.5). By loading aminopeptidase on chitosan microspheres, the loss rate of the enzyme in cheese whey was reduced by approximately 79%. Furthermore, compared with cheese without aminopeptidase and cheese with aminopeptidase added directly, the cheeses made with A-CM exhibited the highest proteolysis level and received superior sensory ratings for taste and smell. The content of key aroma substances, such as 2/3-methylbutanal and ethyl butyrate, in cheese with A-CM was more than 15 times higher than the others. This study provides an approach for accelerating cheese ripening through the use of microencapsulated enzymes.

Abstract:The processing technology of Chinese cheese sauce was studied and the effects of concentrations of cheese powder, skimmed milk powder, whey powder, compound phosphate, modified starch and salt on the sensory and texture qualities of the sauce were investigated by single factor experiment. The optimum formula was determined as follows: the amount of cheese powder of 14 %, the amount phosphate compound of 2.5 %, the amount of skimmed milk powder and whey powder of 20 %, the amount of modified starch of 1.75%, and the amount of salt of 1.8 %.

Abstract:Plant coagulant, animal rennet and microbial coagulant are used as milk coagulating agents resulting in different cheese products. However, emphasis has been on the use of plant coagulant as opposed to that from animal source, rennet. Alternative milk coagulants are therefore investigated instead of animal enzymes. The use of vegetable extracts as milk coagulants has evolved in soft cheese processing. This study explored the potential of plant coagulants as an ideal choice in the production of cheese. Hence, the objective of this study was to evaluate the effects of the coagulants on the yield and proximate composition of cheese. To compare them with cheese coagulated with supernatants from fermented white sorghums (WSS) and wheat (WS) in the laboratory. The results were also compared with cheese coagulated with Calotropis procera leaf extract (CPE) in terms of yield, the percentage composition of total solids (TS), fat, protein, lactose ash and the pH of the cheeses. The same parameters were evaluated in the whey (by-product). The minerals, pH and the proximate components in WS and WSS were also compared. Maximum cheese yield was recorded at equal volumes of milk and coagulants. CPE was superior to these other two considering the constituents in all three cheeses. The chemical composition of CPE coagulant was superior to WS and WSS in terms of TS, fat, protein and ash but lower in moisture content compared with the other two. Protein content was higher in cheese prepared with CPE (22%) about six times the amount in raw milk and the least was WSS (12%). Moreover, the cheese made with CPE had the highest yield (31.76g) and differed significantly (P< 0.05) from WSS (28.87g) and WS (29.70g). Also, cheese made with WS (30.57g/ml) had the highest whey volume compared with WSS (28.11g/ml). However, increasing levels of each supernatant with a constant volume of raw milk (100ml) produced higher cheese yield at 100ml of the supernatant and 100ml of milk. Whey from WSS was the most acidic (5.85) of all the three produced but with increased acidity as the storage days increased. The study revealed that cheese with high nutritional quality can be produced with CPE, WSS and WS. The use of equal volume of coagulant (WSS or WS) to raw milk would give optimal cheese yield. This will promote the utilization of WSS and WS, which is naturally left as the waste product.

Abstract:A new rennase which was gained from fermentation of Blue Phycomycetes was applied.The effects of the ferment amount,the quantity of CaCl2,the rennase concentration and the pH to the coagulating property of cheese were researched during the production.The results showed that when the adding ferment amount 0.2 g/L,the quantity of CaCl2 0.02%,the rennase concentration 2% and the pH 5.9.The cheese had soft flavor and exquisite tissue.

Abstract:Cheese has a long history and this naturally fermented dairy product contains a range of distinctive flavors. Microorganisms in variety cheeses are an essential component and play important roles during both cheese production and ripening. However, cheeses from different countries are still handmade, the processing technology is diverse, the microbial community structure is complex and the cheese flavor fluctuates greatly. Therefore, studying the general processing technology and relationship between microbial structure and flavor formation in cheese is the key to solving the unstable quality and standardized production of cheese flavor on basis of maintaining the flavor of cheese. This paper reviews the research progress on the general processing technology and key control points of natural cheese, the biochemical pathways for production of flavor compounds in cheeses, the diversity and the role of yeasts in cheese. Combined with the development of modern detection technology, the evolution of microbial structure, population evolution and flavor correlation in cheese from different countries was analyzed, which is of great significance for the search for core functional yeast microorganisms and the industrialization prospect of traditional fermented cheese.

Abstract:This study focused on the antioxidant property of protein-hydrolyzed sweet cheese whey, that is the whey hydrolyzed by papain or Subtilisins, and measured the activities of SOD (superoxide dismutase) and GSH-Px (glutathione peroxidase) and the content of MDA (malondialdehyde) in the serum and liver of mice after they were fed with Cheese Whey Enzymatic Hydrolysates for 30 days. Significant improvements of the activities of SOD in the serum and liver of aged mice were proved. In addition, the activity of GSH-Px in the serum and liver of mice were enhanced after the diet of cheese whey hydrolyzed by Subtilisins. Finally, there was no significant difference of the content of MDA between organs of different groups of mice in this assay.

Abstract:Probiotics are used in cheese fermentation to endow the product with unique functional properties, such as enhanced flavor and aroma development through proteolysis and lipolysis. In this study, two probiotic Lactobacillus strains, Lactobacillus plantarum A3 and Lactobacillus reuteri WQY-1, were selected to develop new probiotic cheeses in the form of single- and mixed-strain starters. The results demonstrated that the L. plantarum A3 single-strain group and the L. plantarum A3/L. reuteri WQY-1 mixed fermentation group exhibited superior product performance, particularly the release of functional hydrolysates during cheese ripening. Furthermore, Label-free quantitative proteomic analysis revealed 26 unique antioxidant peptides in the L. plantarum A3 single-strain group and 53 in the L. plantarum A3/L. reuteri WQY-1 mixed fermentation group. Among these, CMENSAEPEQSLACQCL (β-lactoglobulin), CMENSAEPEQSLVCQCL (β-lactoglobulin), and IQYVLSR (κ-casein) have been found to possess potential antioxidant properties both in vitro and in vivo. This confirmed that milk-derived protein peptides in cheese products exhibit potential antioxidant functions through the hydrolysis of probiotic strains.

Abstract:The influence of substitutions of amino acids involved in the formation of salt bridges on the cow chymosin technological properties has been studied. It was shown that point amino acid mutations Asp198→Lys, Asp156→Val and their combination decrease the sensitivity of coagulation activity of engineering recombinant chymosins to change in the concentration of calcium chloride in milk in the range of 1-5 mM. The found effect is a positive technological evidence in terms of cheese making, since it allows varying the content of calcium chloride added to the milk mixture without a threat of significant changes in the undesired proteolytic activity of used milk-clotting enzyme. recombinant chymosin, technological properties, amino acid substitutions, milk-clotting activity, concentration of calcium chloride. Abstract:Human and global health would be greatly improved by replacing animal-based foods with plant-based alternatives. This transition would be facilitated when more affordable, convenient, sustainable, nutritious, and tasty plant-based foods are available on the market. Interest in related research has surged these years, and dozens of plant-based cheese (PBC) products have been introduced to the market. However, studies found that most PBCs are far from comparable with dairy cheese in nutritional value, and the texture and sensory properties are still hurdles to the widespread consumption of PBCs. This paper focused on the "tissue disruption route" and adopted chickpeas and nuts as the main ingredients to form a cheese snack. Test results showed that our samples were high of nourishment value compared to the cheese sample and even healthier than the cheese in the terms of energy (709:37 +/- 1:35 vs. 865:63 +/- 0:49 kJ/100 g, p < 0:01), K/Na ratio (142:00 +/- 0:82/13:33 +/- 0:12 mg/ 100 g vs. 121:67 +/- 2:05/44:23 +/- 0:05 mg/100 g, p < 0:01), DF (6:99 +/- 0:01 g/100 g vs. N/A, p < 0:01), and cholesterol (8:81 +/- 0:25 vs. 207:30 +/- 3:35 mg/100 g, p < 0:01). From the order of magnitude, the PBC samples are similar to the cheese sample in the aspects of cohesiveness (p < 0:05) in texture test, and the fermentation process improved the imitation of texture properties in terms of hardness and cohesiveness (both p < 0:05). The volatile compounds were similar between the two PBC samples; however, the fermentation process could increase the species of the volatile compounds and make the PBC a little more similar to the dairy cheese. Overall, the observed properties of our PBC snack using raw chickpeas and nuts make the "plant-based cheese substitutes" extremely promising because of their nutritional value, sensory, and texture properties.

A Starch-Milk Paste Enables the Incorporation of Ripened Cheese in Novel Fresh Cheese

Abstract:Research background. Fresh cheese varieties represent an important share of the whole cheese market. Although with great variability in terms of composition and method of preparation, fresh cheese varieties are bland in flavour and their production originates whey drainage. On the other hand, the cheese market is also responsible for a significant amount of food waste. These motivated the development of a novel fresh cheese incorporating ripened cheese, which can then represent a valorisation of ripened cheese surpluses. Experimental approach. A variable amount of ripened cheese was dispersed in a paste of gelatinized starch (normal corn or waxy rice) in milk, producing melted cheese bases. These cheese bases were diluted with milk, sometimes enriched with skimmed milk powder, and then renneted. Macronutrient content and physical properties of the resultant fresh cheese were characterized. Sensory analyses of samples incorporating mature Cheddar, goat’s or ewe’s cheese were carried out. Results and conclusions. Gel formation of the initial mixture was hindered above 8 % (m/m) incorporation of ripened cheese, which could be overcome by the addition of skimmed milk powder. These observations are corroborated by the hardness values from texture analysis tests. Evaluation of syneresis of different samples enabled to conclude that the addition of 2 % (m/m) starch and 2.8 % (m/m) skimmed milk powder contributes to reduction of its magnitude by half. Sensory analysis with a consumer panel indicated a preference for a more consistent texture of the fresh cheese, and for the Cheddar flavour. Novelty and scientific contribution. A novel fresh cheese variety incorporating dispersed ripened cheese was prepared. The proposed method is versatile and quite straightforward and does not use polyphosphate salts or originate whey wastage. The fresh cheese physical and sensorial properties can be manipulated by the amounts and types of added starch, ripened cheese and skimmed milk powder; such tailoring of fresh cheese properties widens product portfolio capacity for a larger number of consumer groups. The added ripened cheese can come from non-sellable pieces and unsold stocks from the retail sector, contributing to a reduction of food waste.
