Declined Presentation Effect of Fbxw11 on the Proliferation of Lymphocytic Leukemia Cells

Abstract:F-box and WD-40 domain protein 11 (Fbxw11), a member of F-box protein family, is a constituent of the SCF (Skp1-Cul1-F box) E3 ubiquitin ligase complex. E3 ubiquitin ligases recognize specifically phosphorylated substrates and confer their ubiquitination. A given F-box protein permits these E3 ligases to ubiquitinate a wide range of different substrates. Fbxw11 play pivotal roles in many aspects of hematopoiesis and tumorigenesis by regulating various signal transduction pathways. We found two transcript variants (Fbxw11c and Fbxw11d) in mouse bone marrow. However the role of Fbxw11 variants in the development of leukemia and the underlying mechanisms remain largely unknown. In this study, we established L1210 cell lines over-expressing Fbxw11a, Fbxw11c or Fbxw11d, respectively. The in vitro effect of these variants on the proliferation of L1210 cells was first studied. Growth curve and the MTT experiment suggested that over-expression of Fbxw11 variants stimulated the proliferation of L1210 cells in vitro. Xenograft model with L1210 cells over-expressing Fbxw11a, Fbxw11c or Fbxw11d in DBA/2 mice was established. The tumor growth curves and volume of the xenograft tumors showed that over-expression of Fbxw11 variants could increase growth of tumor in vivo. Cell cycle and apoptosis experiments were carried out, demonstrating that increased cell proliferation in L1210 cells over-expressing Fbxw11 variants was not due to decrease in cell apoptosis but due to increase in cell cycle. In addition, we tested the effect of Fbxw11 variants on the signal transduction by dual-luciferase reporter gene system. The results showed that over-expression of Fbxw11 variants caused the activation of NF-κB signaling pathway. These findings suggested the important role of Fbxw11 on the regulation of leukemia development. Fbxw11 might be a potential therapeutic target against lymphocytic leukemia.
