Incidence of Aneuploid of Chromosomes in Spermatozoa among Infertile Men Detected by Multi-Color Fluorescence in Situ Hybridization

Abstract:Objective: To study the difference in frequency of aneuploid of chromosomes in sperms between infertile men and normal control in order to ascertain whether it serves as a risk factor for male infertility and provide the theory guidance in IUI. Methods: Twelve infertile men were divided into two groups: 10 with oligo-astheno-teratospermia (OAT) (group A) and two with a normal semen analysis (group B). Two normal healthy semen donors were used as control (group C). Fluorescence in situ hybridization (FISH) and probes for chromosomes including X,Y and 18 were used to determine the frequency of aneuploid of chromosomes in 1 000 spermatozoa per individual. Results: In group B the frequencies of spermatozoon disomy for chromosomes including X,Y and 19 were 0.30% and 0.30% respectively. The percentages were not significantly different from those of group C (0.15%,0%). The frequencies of nullisomy ofr chromosome X,Y and 18 in group B were 0.15% and 0%, there was no significant difference from group C (0%, 0.15%). In group A the incidences of disomy for chromosomes X,Y and 18 were 1.13% and 0.96% respectively, nullisomy frequencies were l.13% and 1.6%. In three groups the incidences of diploid were 0.64%,1.0% and 0.35% respectively. In the group A both the frequencies of disomic and nullisomic cells for chromosome X,Y,chromosome 18 and of diploid spermatozoa were significantly higher as compared with group B and C ( P0.05 ). The estimated total aneuploid in the sperms of group A,B and C were 42.44%,6.05% and 2 59%, the percentage in group A was significantly higher than that in group B and C( P0.05 ). Conclusion: The results indicate that chromosomal aneuploid in sperms may be a risk factor for male infertility.
