Improved Recyclability of Spirulina Culture Medium Through Ultrafiltration

Abstract:Spirulina, hailed as "the food of the future for humanity", would release algal organic matter (AOM) that has an inhibitory effect on its growth. In this study, ultrafiltration (UF) was applied to control AOM under different molecular weight cutoffs (MWCO: 1, 5, 10, 30, 100 kDa) in order to investigate the improvement in recyclability of Spirulina cyclic culture medium. During the regenerative processes, AOM was entrapped while the nutrient solution penetrated through the UF. Changes in the contents of the main intracellular metabolites, including protein, lipid and polysaccharide, were analyzed. The chemical compositions of the treated medium were detected. Furthermore, the mechanism of UF in enhancing the recyclability of Spirulina culture medium was explored. The results showed that the accumulation of large molecular weight AOM inhibited the growth of Spirulina, with its maximum biomass concentration decreasing to 83.6% only after only two cycles. Compared to microfiltration (MF), UF alleviated the growth suppression and improved the recyclability of Spirulina culture medium. Especially with MWCO of 5-10 kDa, nearly 9.0 % increased Spirulina biomass was obtained even after five long-term cyclic cultivation of 53 days. After treatment by UF, the main intracellular metabolite contents of Spirulina (including protein, lipid and polysaccharide) were respectively 54.6 %, 12.1 % and 6.7 %, which gradually recovered to the level of the control group. The mechanism study showed that UF could remove components of AOM, such as polysaccharide, protein and the fluorescent substances. In addition, UF could also restore superoxide dismutase (SOD) activity in Spirulina cells and reduce the oxidative damage.
