Abstract:This article reviews studies on the heredity of personality disorders and family environment factors from the population-based studies, and summarizes the interaction between genetic factors and environmental factors. Studies have shown that there is a genetic basis for personality disorders, including family studies, twin studies, and neurophysiological studies. In addition, environmental factors, including social psychological factors, also play an important role in the development of personality disorder. In sum, the occurrence of personality disorder is the result of interactions between genetic factors and environmental factors. This paper snmmarizes sresearch on personality disorders, and investigates the role of interaction between genetic and environmental factors in the development of personality disorders. Such evidence has implications for effective prevention and intervention of personality disorders.
Correlation analysis between schizoid traits and some genes

Abstract:Objective:In order to explore the etiology of schizophrenia,to regard schizoid trait as endophenotype to do genetics analysis.Correlation between schizoid trait and gene of catechol-o-methyltransferase(COMT),proline dehydrogenase(PRODH)and brain-derived neurotrophic factor(BDNF)were studied to provide related data for searching candidate genes of schizophrenia.Method:Using gender,age and education years as limitation factors,465 healthy adults were randomly selected from 5 city zones in Chengdu and enrolled in the study.Schizotypal personality questionnaire(SPQ)was used to evaluate the personality.Regression analysis was conducted between patients' gender,age,education years and the related scores of SPQ by SPSS 11.0 software.Quantitative analysis of allele and haplotype was done between gene of COMT(rs4680G/A、rs165599G/A),PRODH(1852G/A、1945G/A)and BDNF(rs6265)and the related scores of SPQ respectively by QTPHASE software.Correlated analysis was conducted between genotype and schizoid trait by analysis of variance.Results:There was a correlation between education years and seven subscales,three factor scores and the total score of SPQ respectively.The scores above decreased along with the increase of education years.Before correction for multiple testing,there was a relationship between COMT rs4680G/A and weird speech(allele-wise,P=0.04;genotype-wise,P=0.049)and suspiciousness subscale(genotype-wise,P=0.024).And BDNF rs6265 was related to factor 2 interpersonal measure(genotype-wise,P=0.027).The correlation above would disappear after correction for multiple testing.It was found only in male COMT rs4680G/A correlated with the total score,disorganization factor and subscale of constricted emotion of SPQ.Conclusion:COMT rs4680G/A may be associated with male schizoid trait.

Abstract:Personality is influenced by both genetic and environmental factors and is associated with other psychiatric traits such as anxiety and depression. The “Big Five” personality traits, which include neuroticism, extraversion, agreeableness, conscientiousness, and openness, are a widely accepted and influential framework for understanding and describing human personality. Of the big five personality traits, neuroticism has most often been the focus of genetic studies and is linked to various mental illnesses including depression, anxiety, and schizophrenia. Our knowledge of the genetic architecture of the other four personality traits is more limited. Utilizing the Million Veteran Program (MVP) cohort we conducted a genome-wide association study (GWAS) in individuals of European and African ancestry. Adding other published data, we performed GWAS meta-analysis for each of the five personality traits with sample sizes ranging from 237,390 to 682,688. We identified 158, 14, 3, 2, and 7 independent genome-wide significant (GWS) loci associated with neuroticism, extraversion, agreeableness, conscientiousness, and openness, respectively. These findings represent 55 novel loci for neuroticism, as well as the first GWS loci discovered for extraversion and agreeableness. Gene-based association testing revealed 254 genes showing significant association with at least one of the five personality traits. Transcriptome-wide and proteome-wide analysis identified altered expression of genes and proteins such as , and . Pathway enrichment and drug perturbation analyses identified complex biology underlying human personality traits. We also studied the inter-relationship of personality traits with 1,437 other traits in a phenome-wide genetic correlation analysis, identifying new associations. Mendelian randomization showed positive bidirectional effects between neuroticism and depression and anxiety while a negative bidirectional effect was observed for agreeableness and these psychiatric traits. This study improves our comprehensive understanding of the genetic architecture underlying personality traits and their relationship to other complex human traits.

Personality traits and polymorphisms of genes coding neurotransmitter receptors or transporters: review of single gene and genome-wide association studies

Abstract:Background: The most popular tool used for measuring personality traits is the Five-Factor Model (FFM). It includes neuroticism, extraversion, openness, agreeableness and conscientiousness. Many studies indicated the association of genes encoding neurotransmitter receptors/transporters with personality traits. The relationship connecting polymorphic DNA sequences and FFM features has been described in the case of genes encoding receptors of cannabinoid and dopaminergic systems. Moreover, dopaminergic system receives inputs from other neurotransmitters, like GABAergic or serotoninergic systems. Methods: We searched PubMed Central (PMC), Science Direct, Scopus, Cochrane Library, Web of Science and EBSCO databases from their inception to November 19, 2020, to identify original studies, as well as peer-reviewed studies examining the FFM and its association with gene polymorphisms affecting the neurotransmitter functions in central nervous system. Results: Serotonin neurons modulate dopamine function. In gene encoding serotonin transporter protein, SLC6A4, was found polymorphism, which was correlated with openness to experience (in Sweden population), and high scores of neuroticism and low levels of agreeableness (in Caucasian population). The genome-wide association studies (GWASs) found an association of 5q34-q35, 3p24, 3q13 regions with higher scores of neuroticism, extraversion and agreeableness. However, the results for chromosome 3 regions are inconsistent, which was shown in our review paper. Conclusions: GWASs on polymorphisms are being continued in order to determine and further understand the relationship between the changes in DNA and personality traits.
Functional MRI findings in personality disorders: A review

Abstract:Personality disorders (PDs) have a prevalence of approximately 10% in the United States, translating to over 30 million people affected in just one country. The true prevalence of these disorders may be even higher, as the paucity of objective diagnostic criteria could be leading to underdiagnosis. Because little is known about the underlying neuropathologies of these disorders, patients are diagnosed using subjective criteria and treated nonspecifically. To better understand the neural aberrancies responsible for these patients' symptoms, a review of functional MRI literature was performed. The findings reveal that each PD is characterized by a unique set of activation changes corresponding to individual structures or specific neural networks. While unique patterns of neural activity are distinguishable within each PD, aberrations of the limbic/paralimbic structures and default mode network are noted across several of them. In addition to identifying valuable activation patterns, this review reveals a void in research pertaining to paranoid, schizoid, histrionic, narcissistic, and dependent PDs. By delineating patterns in PD neuropathology, we can more effectively direct future research efforts toward enhancing objective diagnostic techniques and developing targeted treatment modalities. Furthermore, understanding why patients are manifesting certain symptoms can advance clinical awareness and improve patient outcomes.
Association Study of Serotonin Transporter and Monoamine Oxidase A Genes Polymorphisms with Antisocial Personality Disorder in Han Chinese Male

Abstract:Objective To investigate whether the 5-HTT-VNTR, the EcoRV polymorphism of MAOA gene was associated with antisocial personality disorder (APD) in Han Chinese male criminal offenders. Method In a prison in Jiangsu, the male criminals were evaluated with the Personality Diagnostic Questionnaire 4, and the diagnosis of antisocial personality disorder was made based on the DSM-Ⅳ criteria. Altogether 118 criminal people with antisocial disorder were evaluated with the Barratt-11 scale, in which 60 cases of total score ≥ the median were as the high impulsion group. Another 250 male criminals without personality disorder and 300 healthy people were recruited as controls. The DNA fragments of 5-HTT-VNTR and MAOA-EcoRV were measured using PCR assay. Both allele frequencies and genotype frequencies among the groups were compared. Results ( 1 ) There were significant differences of alleles of the 5-HTT-VNTR and MAOA-EcoRV ( + ) frequency between 60 APD with high impulsiveness (12/12: 70.0%, 12/10:21.7%, 10/10: 8.3%, 71.7%) and normal controls ( 12/12: 84.7%, 12/10: 10.7%, 10/10: 4.7%,55.3%; x2 = 7.422, P = 0.024, x2 = 5.478, P = 0.019 ), while no significant differences between 118 APD ( 12/12:78.0%, 12/10: 14.4%, 10/10: 7.6%, 63.6% ) and normal controls ( x2 = 2.819, P = 0.244;x2 =2.347,P=0.126). (2)When two genetic locus were combined, the polyploidy of 12/12/- frequency was significantly increased in normal controls compared to 118 APD (x2 =7.164, P=0.007) and APD with high impulsiveness (x2 =9.590, P =0.002), respectively. There was significant difference of 12/12/+frequency only between APD with high impulsiveness and normal controls (x2 = 5.378, P = 0.020 ).Conclusion There may be an association between the 5-HTT-VNTR, MAOA-EcoRV polymorphisms and susceptibility to impulsiveness in male Han Chinese criminal offenders with antisocial personality disorder,with the polyploidy type of 12/12/- may play a role in the development of antisocial personality disorder.
Molecular genetics in psychology and personality neuroscience: On candidate genes, genome wide scans, and new research strategies

Abstract:Despite the substantial heritability estimates for psychological traits, their precise genetic foundation from a molecular perspective remains elusive. We summarize findings and advances from twenty years of research into the molecular genetics of personality and other psychological traits. We describe how the candidate gene approach has – despite its appealing theoretical foundations – often (but not always) failed to point towards replicable associations between genetic polymorphism and behavioral traits. The genome wide analysis (GWAS) approach on the contrary has become more fruitful in recent years and pointed towards reliable genetic associations. GWAS results are currently leveraged to explore gene-behavior associations through genetic correlation and polygenic score prediction which are important steps towards a precision medicine where treatment options are tailored to a patient’s individual biology. The GWAS approach, however, faces its own problems when it comes to linking its extensive findings to biopsychological theory. We argue that research strategies from the candidate gene approach can be used to address these issues – given that necessary precautions are taken to avoid the problem of false-positive associations.

Abstract:Background: The real impact of genetic factors on personality is still unknown, even if in literature about 50% of variance in personality traits are considered genetically determined. The determination of the genetic variance in personality traits could promote psychological well-being and the prevention of psychopathologies, because there are many experimental evidences showing that mental illness is associated to personality. Numerous studies have showed that Catechol-O-methyltransferase (COMT), brain derived neurotrophic factor (BDNF) and serotonin transporter (5-HTT) are genes whose variants are associated with personality traits. This aim of this study is the investigation of the association between personality traits and 5-HTTLPR/rs255315-HTT promoter variant, COMT Val158Met and BDNF Val66Met gene polymorphisms. Methods: The sample was composed by 132 healthy female students. Genomic DNA was extracted from buccal swab, while personality was assessed with Cloninger's Temperament and Character Inventory-Revised (TCI-R). Linear discriminant analysis was used to analyze how personality characteristics can differentiate individuals in relation to their genetic polymorphisms. Results: Data showed that the temperament trait Reward Dependence discriminated individuals with different BDNF variants; Novelty Seeking and Harm Avoidance discriminated individuals with different 5HTTLPR variants; Persistence discriminated individuals with different COMT variants. Conclusions: Since these traits are connected to psychological diseases as depression, social anxiety, anorexia and obsessive-compulsive disorders of personality, the study of their genetic component can be used as intermediary issue to better define the connection between genes and predisposition toward maladaptive behavior and mental illness.
[The influence of genes on the development of personality].

Abstract:Antisocial behavior and personality disorders are both heterogeneous and the product of interacting genetic and environmental factors acting at different levels of causation. Heritability studies show that individual differences in predisposition to antisocial behavior are transmitted by genetic mechanisms in families. Direct gene analysis and genetic linkage analysis have identified structural variants in genes involved in neurotransmitter function, and some progress has been made towards relating these genetic variants to antisocial personality and other behaviors. The monoamine oxidase-A variant leads to aggressive behavior in one family. Direct gene analyses have revealed amino acid substitutions and structural variants at DRD2, DRD3 and DRD4 dopamine receptors and 5-HT2A, 5-HT2C serotonin receptors, serotonin transporter gene, and genes for enzymes, metabolizing biogenic amines MAO-A, MAO-B. The stage is set to identify the phenotypic significance of these as well as genetic variants at other loci which may be relevant as candidate genes for antisocial behavior and related behavioral differences.

Abstract:Personality traits are the relatively enduring patterns of thoughts, feelings and behaviors that reflect the tendency to respond in certain ways under certain circumstances. Twin and family studies have showed that personality traits are moderately heritable, and can predict various lifetime outcomes, including psychopathology. The Research Domain Criteria characterizes psychiatric diseases as extremes of normal tendencies, including specific personality traits. This implies that heritable variation in personality traits, such as neuroticism, would share a common genetic basis with psychiatric diseases, such as major depressive disorder. Despite considerable efforts over the past several decades, the genetic variants that influence personality are only beginning to be identified. We review these recent and increasingly rapid developments, which focus on the assessment of personality via several commonly used personality questionnaires in healthy human subjects. Study designs covered include twin, linkage, candidate gene association studies, genome-wide association studies and polygenic analyses. Findings from genetic studies of personality have furthered our understanding about the genetic etiology of personality, which, like neuropsychiatric diseases themselves, is highly polygenic. Polygenic analyses have showed genetic correlations between personality and psychopathology, confirming that genetic studies of personality can help to elucidate the etiology of several neuropsychiatric diseases.

Abstract:Updates regarding the dimensional model of personality disorder in the DSM-5 and ICD-11 have stimulated interest in the concept of personality functioning (PF). A growing number of literature has demonstrated the extensive empirical basis, validated evaluation tools, and clinical utility of the concept of PF. The concept of PF provides a construct for the diagnosis and evaluation of personality disorders. As a trans-diagnostic factor, PF is of great significance in the etiology and development of many mental and physical conditions. PF can be improved per se, primarily through psychotherapy. The evaluation and treatment of low PF should be considered in all relevant cases. The current study aims to provide a comprehensive review of the concept, pathogenesis, measurement, prevalence, psychopathological significance, as well as intervention for disordered PF.

Abstract:Personality is influenced by genetic and environmental factors and associated with mental health. However, the underlying genetic determinants are largely unknown. We identified six genetic loci, including five novel loci, significantly associated with personality traits in a meta-analysis of genome-wide association studies (N = 123,132-260,861). Of these genome-wide significant loci, extraversion was associated with variants in WSCD2 and near PCDH15, and neuroticism with variants on chromosome 8p23.1 and in L3MBTL2. We performed a principal component analysis to extract major dimensions underlying genetic variations among five personality traits and six psychiatric disorders (N = 5,422-18,759). The first genetic dimension separated personality traits and psychiatric disorders, except that neuroticism and openness to experience were clustered with the disorders. High genetic correlations were found between extraversion and attention-deficit-hyperactivity disorder (ADHD) and between openness and schizophrenia and bipolar disorder. The second genetic dimension was closely aligned with extraversion-introversion and grouped neuroticism with internalizing psychopathology (e.g., depression or anxiety).
Molecular imaging in patients with mood disorders: a review of PET findings

Abstract:Mood disorders are chronic, recurrent psychiatric disorders with high morbidity rates that cause severe disability. Researchers have used molecular imaging extensively in studies of mood disorders. In this article, we concisely and selectively review the major findings of positron emission tomography studies of patients with mood disorders. Specifically, we describe findings from cerebral blood flow, cerebral glucose/oxygen metabolism, and radioligand studies in both cross-sectional and longitudinal investigations. Patients with mood disorders have mood-correlated regional metabolism changes and molecular abnormalities in several neurotransmitter systems. Although the findings of these studies are not completely consistent and confounding factors, including drug effects and specific methodology, should be strictly controlled, these results reveal the pathophysiology of mood disorders and aid the development of novel treatment approaches for mood disorders. Future positron emission tomography research will benefit greatly from the development of better radioligands to simultaneously identify multiple neurotransmitter systems in the specific brain region and the integration of more detecting methods in specifying the neurobiological predictors of treatment response in patients with mood disorders. Understanding the molecular mechanisms in underlying mood disorders will result in aetiological diagnosis and individualization of treatment of these disorders.
Advances in Molecular Genetics of Schizophrenia

Abstract:Schizophrenia (MIM 181500) is a complex disorder affecting approximately 1% of the population worldwide. Epidemiologic evidences, together with recent linkage and association studies, have clearly demonstrated the high heritability of schizophrenia (up to 80%). Uncovering the genetic mechanism of schizophrenia has became one of the greatest challenges for both psychiatry and genetics. In recent years, remarkable advances in the genetics of this disorder has been achieved with the rapid growth of human genome information and experiment technologies. Several candidate genes within some of the best-supported linkage regions have been reported and, more importantly, replicated. Moreover, these genes present a significant connection in the signaling pathways implicated in the development of schizophrenia, especially NMDA receptor-mediated glutamate transmission. In this review, we summarize the recent advances in the genetics of schizophrenia, focusing particularly on linkage disequilibrium analysis and the latest understanding of the neurobiology of the disorder.
Contributions of Dopamine-Related Genes and Environmental Factors to Highly Sensitive Personality: A Multi-Step Neuronal System-Level Approach

Abstract:Traditional behavioral genetic studies (e.g., twin, adoption studies) have shown that human personality has moderate to high heritability, but recent molecular behavioral genetic studies have failed to identify quantitative trait loci (QTL) with consistent effects. The current study adopted a multi-step approach (ANOVA followed by multiple regression and permutation) to assess the cumulative effects of multiple QTLs. Using a system-level (dopamine system) genetic approach, we investigated a personality trait deeply rooted in the nervous system (the Highly Sensitive Personality, HSP). 480 healthy Chinese college students were given the HSP scale and genotyped for 98 representative polymorphisms in all major dopamine neurotransmitter genes. In addition, two environment factors (stressful life events and parental warmth) that have been implicated for their contributions to personality development were included to investigate their relative contributions as compared to genetic factors. In Step 1, using ANOVA, we identified 10 polymorphisms that made statistically significant contributions to HSP. In Step 2, these polymorphism's main effects and interactions were assessed using multiple regression. This model accounted for 15% of the variance of HSP (p<0.001). Recent stressful life events accounted for an additional 2% of the variance. Finally, permutation analyses ascertained the probability of obtaining these findings by chance to be very low, p ranging from 0.001 to 0.006. Dividing these loci by the subsystems of dopamine synthesis, degradation/transport, receptor and modulation, we found that the modulation and receptor subsystems made the most significant contribution to HSP. The results of this study demonstrate the utility of a multi-step neuronal system-level approach in assessing genetic contributions to individual differences in human behavior. It can potentially bridge the gap between the high heritability estimates based on traditional behavioral genetics and the lack of reproducible genetic effects observed currently from molecular genetic studies.

Abstract:Objective:To explore the genetic and family environmental effects on children personality.Methods:Using cross-sectional design,totally 56 pairs of twins aged 6-16 years were recruited according to the school registration information.DNA was extracted from buccal mucosa samples to identify zygosity.The Eysenck Personality Questionnaire(EPQ) was applied to the twins for assessing their personality characteristics,and the General Health Questionaire-12(GHQ-12),Parenting Styles And Dimensions Questionnaire(PSDQ),The General Functioning Scale of the MacMaster Family Activity Device(FAD-GFS),and Stressful Life Events(SLE) were applied to their parents to collect information of family environment.Results:(1) Fifty twin pairs' DNA were picked up successfully;zygosity identification showed that there were 24 monozygotic(MZ) twin pairs and 26 dizygotic(DZ) twin pairs among them.(2) There were significantly positive correlations between the two individuals of MZ twins pair in Neuroticism(N),Extraversion(E) and Lie(L) scores of EPQ(r=0.75,0.73 and 0.56 respectively,P<0.05),and compared with MZ twins pairs,DZ twin pairs only had a weaker correlation between the two individuals of twins pair in N score of EPQ(r=0.47,P<0.05;MZ group:confidence interval of Z:0.94±0.44;DZ group:Z=0.51).(3) E score of EPQ was negatively correlated with score of FAD-GFS(r=-0.20,P<0.05).P score was positively correlated with score of mother's GHQ-12(r=0.22,P<0.05).N score was positively correlated with father's autocratic-parenting-style score.(r=0.24,P<0.05).Conclusion:Gene plays an important role in children personality.However,it should not be ignored that the family environmental,such as mother's mental health,father's rearing style,and general function of family also play a role in children personality.
Molecular Genetic Studies of ADHD and Its Candidate Genes: a Review.

Abstract:Attention-deficit/hyperactivity disorder (ADHD) is a common childhood-onset psychiatric disorder with high heritability. In recent years, numerous molecular genetic studies have been published to investigate susceptibility loci for ADHD. These results brought valuable candidates for further research, but they also presented great challenge for profound understanding of genetic data and general patterns of current molecular genetic studies of ADHD since they are scattered and heterogeneous. In this review, we presented a retrospective review of more than 300 molecular genetic studies for ADHD from two aspects: (1) the main achievements of various studies were summarized, including linkage studies, candidate-gene association studies, genome-wide association studies and genome-wide copy number variation studies, with a special focus on general patterns of study design and common sample features; (2) candidate genes for ADHD have been systematically evaluated in three ways for better utilization. The thorough summary of the achievements from various studies will provide an overview of the research status of molecular genetics studies for ADHD. Meanwhile, the analysis of general patterns and sample characteristics on the basis of these studies, as well as the integrative review of candidate ADHD genes, will propose new clues and directions for future experiment design.
The Correlation Between Peripheral Blood Micro-Ribonucleic Acid Expression Level and Personality Disorder in Patients with Schizophrenia

Abstract:Objective: Schizophrenia patients often have personality disorders; schizophrenia patients with personality disorders are more difficult to treat and have a worse prognosis. Early identification of this group of patients and early intervention can achieve better prognosis. Therefore, it is very important to explore effective biomarkers and early diagnosis for the prognosis of schizophrenia. The primary purpose of this paper is to explore the relationship between plasma miRNA expression level and personality disorder with schizophrenia. Methods: Gene microarrays in miRNA files were employed, and the plasma of peripheral blood of 82 schizophrenic patients and 43 healthy control subjects were examined. Real-time reverse transcription polymerase chain reaction detection were performed to explore the results. Spearman correlation analysis was used to analyze the correlation between expression level of miRNAs and Personality Diagnosis Questionnaire-4 score. Results: The results showed that miR-1273d, miR-1303, miR-3064-5p, miR-3131, miR-3687, miR-4428, miR-4725-3p, and miR-5096 were up-regulated in schizophrenic patients. Compared to healthy control subjects, the difference was statistically significant (P < .05). Schizophrenic patients with schizoid, paranoid, schizotypal, and obsessive compulsive traits had negative correlation with miR-1303, miR-3131, miR-4428, and miR-5096 expression level (r = -0.40 to -0.62, P < .05); there were no significant differences in the other miRNAs. Correlation with other personality traits was not significant (P > .05). The stepwise regression analysis indicated that miR-5096, miR-3131, and miR-1273d have a significant predictive effect on the schizoid trait (P < .01). MiR-4428 and miR-1303 had a significant predictive effect on the schizotypal trait (P < .01). MiR-5096, miR-4428, and miR-4725-3P had a significant predictive effect on the paranoid trait (P < .05). MiR-4428, miR-1303, and miR-5096 had a significant predictive effect on the obsessive compulsive trait (P < .05). Conclusion: The expression levels of miR-1273d, miR-1303, miR-3064-5p, miR-3131, miR-3687, miR-4428, miR-4725-3p, and miR-5096 were up-regulated in the peripheral blood of patients with schizophrenia, and these miRNAs are expected to be diagnostic biomarkers for accurate diagnosis of schizophrenia. The expression levels of miR-1303, miR-3131, miR-1273d, miR-4428, miR-4725-3p, and miR-5096 have significant predictive effects on personality disorder in schizophrenia.

Abstract:Objects Polymorphism of genes for dopamine D 2 receptor promoter (DRD2P), 5 HT 2A receptor (5 HT 2A ), catechol o methyltransferase (COMT), and monoamine oxidase A type (MAO A) were examined synchronously for exploring the genetic factors related to schizophrenia Methods Forty three pedigrees with at least two siblings affected schizophrenia according to ICD 10 criteria and biological parents alive were recruited The Chinese versions of the Diagnosis interview for Genetics Study (DIGS) and the Family Interview for Genetics Study (FIGS) were fulfilled Genomic DNA was isolated from lymphocytes by a standard procedure After polymerase chain reaction (PCR), the cleavage was done Then the genotypes were read by 1% agarose gel electrophoresis with 100 bp DNA ladder The chi square test, correlation analyses, and the transmission disequilibrium test (TDT) were employed Results (1) No significantly different distribution between groups of parents, non affected siblings and affected siblings was found, when polymorphism of DRD2P, 5 HT 2A , and COMT were analyzed But significantly different distribution of genotype or allele was found, when comparing with 941A/941G and 941G/941G genotypes of MAO A between groups of parents and non affected siblings, or parents and affected siblings No significant difference, however, was found between the groups of non affected siblings and affected siblings (2) 941G allele of MAO A was transmitted significantly more to affected siblings from parents (χ 2= 5 33, P 0 05, OR =3 32,95% CI : 1 58 6 99) And 941A allele of MAO A and -241A allele of gene for DRD2 promoter were transmitted significantly more to non affected siblings from parents (χ 2=4 84, P 0 05, OR =6 04, 95% CI : 2 01 18 69) But both T102G polymorphism of 5 HT 2A and Val108Met polymorphism of COMT were not found to have significant transmission disequilibrium ( P 0 05) (3) Significant relationship was found between genotype of gene for DRD2P promoter, or COMT, or MAO A and schizophrenic clinical features, but not genotype of T102C polymorphism of 5 HT 2A Conclusions (1) Our results support hypothesis of DA dysfunction in the etiology of schizophrenia (2) From this study, we infer that there are relationships between these candidate genes and schizophrenia at different degree That is MAO ADRD2PCOMT5 HT 2A .
