Heat shock protein 72 inhibits cytochrome C release in ATP depletion-induced apoptosis in renal tubular epithelial cells

Abstract:Objective To clarify the effect and molecular mechanism of heat shock protein (HSP)72 on cytochrome C release in ATP depletion-induced apoptosis in renal tubular epithelial cells. Methods To induce apoptosis, cells were transiently exposed to metabolic inhibitor for 60 minutes followed by recovery. Wild-type HSP72 was over-expressed by transient heat stress or by infecting OK cells with adenovirus containing wild-type human HSP72. Cellular distribution of cytochrome C was detected with Western-blot. Caspase 3 activity was measured with fluorometric assay. Nuclear condensation and apoptotic bodies were examined by Hoechst 33342 dye. Results During ATP depletion in renal epithelial cells, cytochrome C content in cytoplasm arose. Caspase 3 activity appeared after 60 minutes exposure to metabolic inhibitor and progressively increased to seven- to eightfold after 30 minutes recovery. Cell shrinkage, chromatin condensation and/or apoptotic bodies were observed in control cells after 1 hour of ATP depletion with 2 hours of recovery. In contrast to controls, heat stress significantly decreased cytochrome C release from mitochondria and caspase 3 activity( P 0. 05, n = 3). The selective over-expression of HSP72 reproduced the inhibitory effect of heat stress on caspase 3 activity, and cell shrinkage, chromatin condensation and/or apoptotic bodies were much less marked in the over-expression of HSP72 cells. Conclusion HSP72 can inhibit ATP depletion-induced apoptosis in renal tubular epithelial cells by preventing cytochrome C release and caspase 3 activation.
