Abstract:Objective: To investigate the effect of ALA-PDT under the same experimental conditions on the survival rate of various kinds of leukemia cells and the difference in death model. Methods: We selected five kinds of leukemia cell lines to research and the survival rate of the cell was assessed by MTT method, the death model was inspected by Annexin V-FITC/PI. Results: Different kinds of cells have different sensitivities under the same experimental conditions of ALA-PDT. The sensitivity decreased in the following order U937MOLT-46T-CEMK562HL60. Cells treated by ALA combined with irradiation had significant lower survival rate than by single factor. Different kind of cell has different death model, early apoptosis is the main death model in U937 and MOLT-4 cells, late apoptosis is the main death model in HL60 and K562 cells, but most 6T-CEM cell presented necrosis. Conclusion: ALA-PDT can kill leukemia cells and the different kinds of cells have different sensitivity to it.
OPTIMIZATIONS FOR 5-AMINOLEVULINIC ACID BASED PHOTODYNAMIC THERAPY IN PURGING LEUKEMIA CELL HL60

Abstract:Objective To optimize experimental parameters for the photosensitization of 5-aminolevulinic acid (ALA) in promyelocytic leukemia cell HL60 and compare them with normal human peripheral blood mononuclear cell (PBMC). Methods ALA incubation time, wavelength applied to irradiate, concentration of ALA incubated, irradiation fluence may modulate the effect of 5-aminolevulinic acid based Photodynamic Therapy (ALA-PDT).The high-pressure mercury lamps of 400W served as light source, the interference filter of 410nm, 432nm, 545nm, 577nm were used to select the specific wavelength. Fluorescence microscope was used to detect the fluorescence intensity and location of protoporphyrin IX (PpIX) endogenously produced by ALA. MTT assay was used to measure the survival of cell. Flow cytometry with ANNEXIN V FITC kit (contains annexin V FITC, binding buffer and PI) was used to detect the mode of cell death. Results ① 1mmol/L ALA incubated 1×105/mL HL60 cell line for 4 hours, the maximum fluorescence of ALA induced PpIX was detected in cytomembrane. ② Irradiated with 410nm for 14.4J/cm2 can result in the minimum survivability of HL60 cell. ③ The main mode of HL60 cell death caused by ALA-PDT is necrosis. Conclusion ALA for 1mmol/L, 4 hours for dark incubation time, 410nm for irradiation wavelength, 14.4J/cm2 for irradiation fluence were the optimal parameters to selectively eliminate promyelocytic leukemia cell HL60 by ALA based PDT. The photosensitization of ALA based PDT caused the necrosis of HL60 cell, so it could be used for inactivation of certain leukemia cells.
Experiment Research of the Vivo Destruction of HL60 Cells by the Use of 5-Aminolevulinic Acid-based Photodynamic Therapy

Abstract:The leukaemia cells HL60 was carried an experimental research in photodynamic therapy(PDT) based on 5-aminolevulinic acid(ALA) by the use of our PDT reaction room.We got the best optics parameter factor to kill HL60 cell:The desnity of end that ALA trains is 1.0 mM/ml;the cultivation time of ALA is 4 h,it is 412 nm that the irradiation light wave is efficient;the mere dosage of the irradiation is 18 J/cm2;the more power of the irradiation is 5 mW/cm2;FCS density of training the cell is 8%～10%.Can be up to89.9% to the kill of HL60 under the optics parameter,and the damage of PBMC is only 15%,can reach the alternative ground kill tumor cell HL60 basically.

Abstract:Objective:To investigate whether 5-Aminolevulinic acid-baced photodynamic therapy(ALA-PDT) can induce resistant leukemia HL-60/ADR cell line apoptosis.Methods:Using HL-60/ADR cell line as a model,the cells were divided into 4 groups,cell survival rate was determined by MTT assay and cell morphological change was observed by Wright's staining.Apoptotic cell was studied by Annexin V-FITC/PI through flow cytometry assay and character of apoptotic cell was inspected by laser scanning confocal microscopy(LSCM).Result:Wright's staining showed apoptotic change of HL-60/ADR cell after ALA combined with irradiation.The survival rate of the cells of ALA+PDT group decreased more significantly after 48hrs(22.23±6.07)% than 24hrs(48.67±8.14)%.The apoptotic ratio of HL-60/ADR cells was(6.35±1.40)%,(8.07±1.87)% and(57.66±7.55)% respectively 3hrs,4hrs and 24hrs after ALA combined with irradiation.It is markedly higher than the control group.LSCM showed the typical apoptotic character.Conclusion::ALA-PDT can kill resistant leukemia HL-60/ADR cell.Apoptosis is the main death model with time-dependent.
The Experiment Conditions for Destruction of HL 60 and K562 Leuk Emia Cells By5-Aminolaevulinic Acid-Based Photodynamic Therapy

Abstract:Objective: To investigate the influence of concentration of 5-aminolaevulinic acid (ALA),time of incubation with ALA,wavelength of light and post-PDT incubation time on the survival rate of HL60 and K562 leukemia cells killed by 5-aminolaevulinic acid-based photodynamic therapy(ALA-PDT).Methods:Different experimental conditions were used to kill HL60 and K562 leukemia cells.The survival rate of the cells was assessed by MTT method.Results:ALA combined with irradiation could induce death of leukemia cells effectively.The survival rate of the cells decreased with increased concentration of ALA.When the cells were incubated for 4 hours in darkness with 2×105/ml cell density and 2mmol/L ALA concentration,the survival rate was the lowest.The cells treated by irradiation with 410nm wavelength light source had lower survival rate than with 570nm.After ALA-PDT,if cells were incubated for another 24 hours,the survival rate of K562 cells became higher than immediate survival rate,but the survival rate of HL60 cells decreased further.ALA-PDT had slight effect on the survival rate of PBMC and normal peripheral hematopoietic stem cells under the same conditions.Conclusion:ALA-PDT can kill HL60 and K562 leukemia cells.With concentration 2mmol/L of ALA,4 hours incubation in darkness,410nm of wavelength of light source,the effects of ALA-PDT on the survival rate of leukemia cells are significant.Under this condition,HL60 cells are more sensitive than K562,while PBMC and peripheral blood stem cells are affected slightly.

Abstract:A study to explore the optimal experimental parameters and the photosensitization of 5-aminolevulinic acid (ALA)-based photodynamic therapy (PDT) in promyelocytic leukemia cell HL60 has been conducted, in which HL60 cells and their control groups, peripheral blood mononuclear cell (PBMC), first are incubated with different concentrations of ALA in dark for different periods of time and then followed by irradiating with different wavebands for different fluences. Fluorescence microscope and spectrofluorometer have been used to detect the fluorescence of protoporphyrin IX (PpIX) endogenously produced by ALA. The response of the cells to ALA-PDT was evaluated by the 3-(4,5-dimethyl-2-thiazolyl)-2-5-diphenyl-2H-tetrazolium bromide (MTT) assay (interval between irradiation and the MTT assay is 24 h) and by flow cytometry (the length of time between irradiation and the flow assay is 30 min). MTT results will reflect the relative number of metabolically active mitochondria in the population. Propidium iodide uptake in flow cytometry will test for membrane damage. The results of parameter experiments were obtained: 1 x 10(5)/mL HL60 cell was first incubated with 1 mmol/L ALA in dark for 4 h and the maximum fluorescence of PpIX level appeared; then irradiated with 410 nm (4 mW/cm2) for 14.4 J/cm2 and maximum photodamage to membrane and mitochondrial function of HL60 cell resulted. With the normal granulocytes, such response was not detected. Therefore a hypothetical idea can be brought forward that ALA-based PDT can be used for inactivation of leukemia cell HL60 and these optimal parameters may be useful for clinical application.

Abstract:In order to study the effects of nanoparticle TiO2-xNxon the inactivation of Leukemic HL60 cells in vitro by use of 5-aminolevulinic acid(ALA) based on photodynamic therapy(PDT),the 3-(4,5-dimethyl1-2-thiazolyl)-2-5diphenyl-2H-tetrazolium bromide(MTT) assay was used to evaluate the effects of nanoparticle TiO2-xNx on the growth of Leukemic HL60,the effects of nanoparticle TiO2-xNx with different concentrations on the relative survival rate of Leukemic HL60 after PDT and the relative survival rate of Leukemic HL60 based on ALA and PDT(ALA-PDT) by adding nanoparticle TiO2-xNx.The results indicated that the nanoparticle TiO2-xNx had an obvious inactivation to Leukemic HL60 cells when its final concentration was between 50 μg/mL and 250 μg/mL;and the 150 μg/mL was the best concentration especially among them.ALA-PDT based on a suitable concentration of nanoparticle TiO2-xNx could improve the efficiency of inactivation of Leukemic HL60 cells;the relative survival rate of Leukemic HL60 cells was the least after PDT based on both ALA and TiO2-xNx.
The mechanism and influencing factors on photodynamic purging of leukemia cells

Abstract:The paper compared the response of HL60 cells with peripheral blood mononuclear cell (PBMC) after 5-ALA PDT for different periods of time and then followed by irradiating with different wavebands for different fluences. The results of parameter experiments were obtained: 1 x 10(5)/mL HL60 cell was incubated with 1 mmol/L ALA in dark for 4 h and the maximum fluorescence of PpIX level appeared; then irradiated with 410 nm (4 mW/cm(2)) for 14.4 J/cm(2) and maximum photodamage to membrane and mitochondrial function of HL60 cell resulted. Analyzed the experimental results of references, we got that the effect of photodynamic purging of leukemia cells may be modulated by the leukemia cell type; the photosensitizer's type, dose, and its location point; the incubation time and condition; the light wavelength and its dosage. If the light is delivered at a high rate, significant heating of a molecule and its surrounding may take place. A too low dosage has no effect at all or will result in high relapse rate due to the presence of cancer cells. The wavelength and intensity of light required to carry out photopurging is determined by the absorption properties of the photosensitizer. The light dose actually received by the photosensitizer and tissue may be higher or lower depending on the manner of irradiation.
[An Experimental Study on Destruction of K562 and HL60 Induced by 5-Aminolaevulinic Acid-Based Photodynamic Therapy].

Abstract:This experiment was designed to explore the pattern of K562 and HL60 leukemia cells death, the effects on their cell cycle and the cytoplasmic free calcium concentration ([Ca2+]i) induced by 5-aminolaevulinic acid-based photodynamic therapy (ALA-PDT). Under the transmission electron microscope (TEM), two kinds of leukemia cells' ultrastructure were observed. Flow cytometry combined with Annexin V-FITC/PI labeling was used to detect the pattern of K562 and HL60 cells' death induced by ALA-PDT. Flow cytometry combined with PI labeling was used to analyze the change in the cell cycle induced by ALA-PDT, and confocal laser scanning microscopy (CLSM) combining with calcium fluorescence probe was used to detect the change in the cytoplasmic free calcium concentration ([Ca2+]i). Immediately after irradiation, many typical apoptotic bodies were seen in the cells treated. Most of the cells treated were necrotic at 24 hours following irradiation. Flow cytometry analysis suggested that the main patterns of the cells' death were apoptosis immediately after irradiation and necrosis post-apoptosis at 24 hours post irradiation. Immediately and 24 hours after irradiation, the proportion of S phase of K562 was 57. 67% +/- 1.13% and 84.77% +/- 6.20% respectively, and the proportion of S phase of HL60 was 74.60% +/- 7.27% and 84.60% 1.74% respectively. Both [Ca+]i of the treated K562 and HL60 were increased obviously. In the best experiment condition, the initial pattern of the K562 and HL60 leukemia cells' death induced by PDT was apoptosis and the main pattern was necrosis post apoptosis. The two kinds of cells were arrested at S phase by ALA-PDT. During the death of the leukemia cells, the increase in intracellular free calcium concentration could be responsible for the ALA photodynamically induced damage to K562 and HL60 cells.

Abstract:The leukaemia cells HL60, incubated in 10 mM/ml ALA (5-aminolevulinic) for 4 hours, were carried an experimental research with fluorescent probes in photodynamic therapy (PDT) based on ALA by using PDT reaction room (The average fluence rate of the 412 nm source was 5 mW/cm 2 ).Cells viability were determined using a Cell Counting Kit-8 (CCK-8) assay, and PpIX Photobleaching of subcellular distributed sites of HL60 cells in vitro were investigated by fluorescence spectra acquired during treatment.The results showed that the fluorescence intensity of mitochondria, lysosomes, endoplasmic reticulum had decreased by 81.5%, 52.3% and 21.0%, respectively, compared with their initial values after a 45-minute light treatment.The rate of PpIX photobleaching in mitochondria was significantly higher than others.Additionally, the change of the activity of HL60 cells was basically characterized by the change fluorescence intensity in mitochondria, which suggest that mitochondria is one of main therapeutic targets of photodynamic therapy.

Abstract:Background and purpose:Because of a preferable uptake of endogenous photosensitizer(PpIX) derived from ALA in neoplastic cells,there is a selective destruction of tumor.Up to now,the specific mechanism of ALA-PDT-mediated cytotoxicity has not been defined.In this study,we investigated the apoptotic effect on HL-60 cell line induced by 5-aminolevulinic acid-based photodynamic therapy.Methods:HL-60 cells were treated and divided into four groups,only ALA group,only PDT group,ALA+PDT group and control group.Cell survival rate was determined by MTT assay and cell morphological change was observed by Wright's staining.Apoptotic cell was studied by Annexin V-FITC/PI through flow cytometry assay and character of apoptotic cell was inspected by laser scanning confocal microscopy(LSCM).Results:Wright's staining showed apoptotic change of HL-60 cell in ALA+PDT group.The survival rate of the cells of ALA+PDT group decreased more significantly after 48 hrs(46±9)% than after 24 hrs(26±8)%(P0.05).The apoptosis rates of HL-60 cells at 4,5 and 24 hrs after ALA combined with irradiation were(26±9)%,(29±11)% and(51±6)%,respectively.It is markedly higher than that in the control group(P0.05).LSCM showed the typical apoptotic character.Conclusions:ALA-PDT exerts profound cytotoxic effects on HL-60 cell line.Apoptosis is the main death model with time-dependent.
An Experimental Research on Destructive Effect of ALA-PDT on K562 Cells by Different Light Source

Abstract:Objective:To investigate the effect of ALA-PDT on K562 cells by 532 nm pulse laser,532 nm continuous wave laser and Xe lamp,and to choose the optimal light source and optimal irradiation dose for exogenesis' experiment of ALA-PDT.Methods:The light source and the irradiation dose of different ALA-PDT groups were different,while the other experimental conditions were same.The survival rate and the death model of cells was assessed by inversion microscope and trypan blue dye exclusion test.Results:The suppressive rate of ALA-PDT is low by 532 nm laser and it's useless to improve the rate by enhancing the laser power.Five minutes is the optimal irradiation time for ALA-PDT by Xe lamp under the same irradiation conditions.Irradiation injury caused by the light alone was slight,but the destruction rate of ALA-PDT was very high.Conclusion:Xe lamp's action spectrum is wider and it's power is higher,so it's better than 532 nm laser for ALA-PDT.Xe lamp is suitable for ectogenesis experiment of ALA-PDT.

Abstract:5-Aminolevulinic acid-based photodynamie therapy (ALA-PDT) is a relatively new modality to cancer treatment.Be- cause of a preferable uptake of endogenous photosensitizer(PpIX) derived from ALA in neoplastic cells,there is a selective destruction of tumor.The studies on leukemia are limited in basic research today.We will review the mechanism of the cytotoxic effects on leukemic cell lines,studies on multidrug resistance,bone marrow purging in autologous transplants and application prospect of ALA-PDT.
The Comparative PDT Experiment of the Inactivation of HL60 on Modified TiO2Nanoparticles

Abstract:Four samples of modified titanium dioxide (TiO2), Fe/TiO2(2 wt%), Fe/TiO2(5 wt%), and 5-ALA/TiO2, were experimented in photodynamic therapy (PDT) on leukemia cells HL60, performing promising photocatalytic inactivation effect. Fe/TiO2and 5-ALA/TiO2were synthesized in methods of precipitation and ultrasonic methods, respectively. X-ray diffraction spectra and UV-Vis spectra were studied for the samples’ crystalline phase and redshift of absorption peak. Further, FTIR spectra and Raman spectra were obtained to examine the combination of 5-aminolevulinic (5-ALA) and TiO2nanoparticles. The toxicity of these four kinds of nanoparticles was studied through darkroom experiments. And based on the concentration which caused the same toxic effect (90%) on HL60, PDT experiments of TiO2, Fe/TiO2(2%), Fe/TiO2(5%), and ALA/TiO2were done, resulting in the fact that the photokilling efficiency was 69.7%, 71.6%, 72%, and 80.6%, respectively. Scanning electron microscope (SEM) images of the samples were also taken to study the morphology of HL60 cells before and after PDT, resulting in the fact the activation of the modified TiO2from PDT was the main cause of cell apoptosis.

Abstract:Photodynamic therapy is a kind of new method of treatment for leukemia. In this thesis, we use the quantum dots CdTe as photosensitive drugs on photodynamic therapy study of leukemia with chemotherapy, in order to explore new methods. This thesis studied the Qds-PDT treatment based on CdTe on HL60 in vitro.And it also studied the effect of the inactivated leukemia HL60 tumor cells by the three antibiotics in vitro.What’s more, based on the selected quantum dot and the PDT reaction chamber, photodynamic therapy of inactivation tumor cells HL60 is carried out by using the new photosensitizer with chemotherapy in vitro.

Abstract:The 5-aminolevulinic-adsorbed TiO2( 5-ALA/TiO2) were synthesized by a surface modification method.ATR-FTIR,Raman imagine and ultraviolet-visible absorption spectrocopy( UV-Vis) were employed to study the features of 5-ALA /TiO2. Then the inactivation of 5-ALA /TiO2on leukemia tumors was investigated by using Cell Counting Kit-8( CCK-8) assay. Furthermore,the morphology and apoptotic percentage of treated cells by 5-ALA/TiO2were studied.The inactivation efficiency of 5-ALA /TiO2was higher than 5-ALA and TiO2,indicating that the inactivation efficiency could be effectively enhanced by synthesizing 5-ALA and TiO2. The inactivation efficiency of HL60 cells was 77. 9%.The study on the morphology of treated cells by phase contrast microscope showed that cells apoptosis start from the burst of film of cell. The micro Ramans spectroscopy test showed that TiO2nanoparticles can get inside and interact with the HL60 cell.

Abstract:With different Cultured time and different dosages,the TiO2, iron-doped TiO2 nanoparticles and the HL60 cells are cultured in the Same Container.The inactivated effect ot TiO2,iron-doped TiO2 nanoparticles on HL60 leukemia tumor cells was studies.Then results of the cells activities under the condition of the visible radiation and under the condition of without radiation are compared.The possibilities of the TiO2 and iron-doped TiO2 nanoparticles as photo sensitizer PDT inactivated tumor cells are also explored.The experiment results showed that nanoparticies have inhibited toxicity effect on the growth of cells,the longer the culture time and the thicker the nanoparticles density are the better the effect of toxicity will be the rate of the PDT extinguishment under addition of the nanoparticies is higher than under no nanoparticies,but the interaction is not obvious.

Abstract:A LED array is used as PDT light source in a photo-reaction chamber. The LED array is alterable in wavelength by replacing different LEDs, stable in output power, adjustable in power value and spot size; the geometry of the LED array is flat shape. Based on the experiment of ALA-PDT, we measured and analyzed the spectrum characteristics and output power of some different purple LEDs, selected one as PDT light source whose emission peak is near the absorption peak of the ALA. The experiment on HL60 tumor cells in vitro demonstrates our photo-reaction chamber has visible photodynamic effect on ALA-PDT.
[Mitochondria-Mediated Apoptosis Induced by 5-Aminolevulinic Acid-Based Photodynamic Therapy in HL-60 Cells].

Abstract:This study was purposed to investigate the changes of mitochondrial membrane potential (MMP) and apoptosis-related gene Bcl-2 expression of HL-60 cells treated with 5-aminolevulinic acid-based photodynamic therapy (ALA-PDT). HL-60 cell line was used as a model and divided into 4 groups: ALA group, PDT group, ALA+PDT group and control group. The change of MMP was detected by flow cytometry with JC-1 (lipophilic cation 5, 5', 6, 6'-tetrachloro-1, 1', 3, 3'-tetraethyl-benzimidazol-carbocyanine iodide); the mRNA expression of Bcl-2 was determined by semi-quantitative RT-PCR and real-time PCR. The results demonstrated that MMP significantly decreased after treatment with ALA-PDT and the ratio of cells with disrupted MMP obviously increased in ALA+PDT group in time-dependence manner, as compared with control, ALA and PDT groups (P < 0.05), while no difference between ALA and PDT groups was found. The semi-quantitative RT-PCR and real-time PCR showed that the expression level of Bcl-2 was obviously down regulated at 2 h after ALA-PCT, further down-regulated at 4 h, and lasted in low level at 24 h. It is concluded that ALA-PDT-induced apoptosis of HL-60 cells is associated with its effect on MMP, that is ALA-PDT promotes cell apoptosis through effect on mitochondrial function.
