Tissue distribution and cell location of CA11:a novel gene down-regulated in gastric cancer

Abstract:Aim To screen down-regulated genes in gastric cancer, and demonstrate their tissue distribution and cell location. Methods cDNA suppression subtractive library that established by counterpart of normal gastric mucous membrane mRNA(Tester) and subtracting gastric cancer cell mRNA(Driver) of five patients was screened by random sequencing. Screened down-regulated gene CA11 in gastric cancer was confirmed by RT-PCR.CA11 mRNA was located by in situ mRNA hybridization, and was amplified in other organs and tissues. CA11 was cloned into vector pcDNA3.1/Myc-His(-)A and was transfected into COS-7 cells by lipofectamin. The loading sample of SDS electrophoresis came from culture medium of transfected COS-7 cells that stay in Log growth period when Western blot was undergone. COS-7 cells were transfected by CA11, and anti-myc tag antibody was added which bound to the FITC-labelled second antibody. Then CA11 the expressed in sub-cell structure was observed unter fluorescent microscope. Results Down-regulated CA11 in gastric cancer was screened, and confirmed by RT-PCR.CA11 was located in gastric mucous epithelial cells. In other organs and tissues, CA11 mRNA could not be found. Western blot could not prove the existence of CA11-Myc fusion protein in culture medium of COS-7 cells. Its expressed protein was located in cytoplasma. Conclusion CA11 is a down-regulated gene in gastric cancer. Its mRNA is located in gastric mucous epithelial cells, and the protein is expressed in cytoplasma.
