Abstract:Mammal somatic cell nuclear transfer(SCNT) has great potential in agricultural,biotechnology,human medicine production,and protection of animals and so on.It has become an important research method of developmental biology presently.The article summarizd the types of donor cells,the age of donor cell animals,the donor cell cycles,the times of handing down from generation to generation,as well as the sex of donor animal,which influence the effect of animal cloning according to domestic and foreign data in recent years.

Abstract:Somatic cell nuclear transfer offers exciting opportunities in many areas and brings tremendous changes to agriculture, conservation and therapeutics. However, the fundamental research of nuclear transfer including nuclear reprogramming is not fully understood. Some inappropriate epigenetic modifications have been reported in the clone animals, including abnormal regulation of DNA methyltransferase expression, improper expression of imprinted genes,reactivation of the inactive X chromosome. The low efficiency of nuclear transfer hinders the further progress in some degree, we describe some recent advances in applications, methods and main problems limiting efficiency.

Abstract:Transgenic techmology has developed quickly in recent years.Thegene of interest could be integrated into the chromosome of donor-somatic cell of animal.After donor is transferred into the recipient-enucleated oocyte,the reconstructed embryo is transplanted into the female animal to establish pregency until the birth of offspring.Thus,we can get atransgenic clone animal,which is genetically modified.This paper will review the history of development,the status of research,the future of applications and the existing problems in this field.
The Effects of Different Donor Cells and Passages on Development of Reconstructed Embryos.

Abstract:In order to study the effects of different donor cells and passages on development of nuclear transfer embryos, we constructed embryos by electrofusing several kinds of donor cells into enucleated M II oocytes from Kun Ming (KM) mouse. These cells include 2-cell embryonic blastomeres, KMW embryonic stem (ES) cells, fetal fibroblast, ear fibroblast, tail tip fibroblast, sertoil cells and spermatogonia. Meanwhile, we compared the effects of passage numbers of fetal fibroblast cells on developmental competency after nuclear transfer. We found that 7.4% of reconstructed embryos from 2-cell embryonic blastomeres and 0.7% from ES cell could develop to blastocyst in vitro; embryos from fetal fibroblast could only develop to morula stage with the rate of 0.2%; embryos from spermatogonia could only develop to 8-cell stage and the rate was 0.3%; embryos respectively from ear fibroblast, sertoli cell and tail tip fibroblast could only develop to 4-cell stage. Although 2-cell development rate of embryos reconstructed from fetal fibroblast in first passage was significantly lower than those from the 2nd, the 3rd and the 4th passage, embryos from different passages could develop to 8-cell stage except the 3rd passage. The result indicated that it is more difficult for terminally differentiated cell nuclei to be reprogrammed in enucleated M II oocytes than for low differentiated cell nuclei. The reason of low development rate from ES cells maybe that most of ES cells was at S stage of the cell cycle, which out of coordination with M II oocytes. We could conclude that culture and passage of donor cells might be benefit to nucleus reprogramming.
Effect of Type of The Donor Cell on The Development of Bovine Somatic Cell Nuclear Transplantation Reconstituted Embroys

Abstract:We studied the type of donor cell on the development of bovine somatic cell nuclear transplantation reconstituted embryos. The oocyte was cultured for 22~23h to reach maturity in vitro. The nuclear of the mature oocyte was taken out by micromanipulation. The donor nuclei came from fibroblasts and granular cells. Cells were divided into two halves, which were respectively treated, by serum starvation and non- starvation. The donor nuclei were transplanted into the enucleated oocyte by the microinjection of cytoplasm to form reconstituted embryos. After being activated in Ionomycin media for 5 min, the recombinant embryos were firstly put into 6-DMAP media to be cultured for 4 h, then into the CR1 media. The cleavage rate of the reconstituted embryos was examined after 24h and 36h and the growth rate of the blastocyst was counted after 8 d. The results show that the development rate of blastocyst of the somatic cell treated by serum starvation is strikingly higher than that of the reconstituted embryos from bovine somatic cell treated by serum starvation.(fibroblasts: 3.16% vs 11.76%, p<0.05; granular cells: 7.14% vs 14.96%, p<0.05).There is not much difference in the development rate of blastocyst between the reconstituted embryos from fibroblasts and granular cells. The result implies that the reconstituted embryos from non- starvation treated somatic cells have higher development rate and the embryos from bovine granular cells have higher development rate in vitro.
Preparation of Donor Nuclei by Immunosurgery and Application in Bovine Somatic Cell Nuclear Transfer

Abstract:This study was designed to assess the effects of the donor nuclei islation which was performed by the rabbit anti-bovine-spleen serum lysising the membrane of the donor cells. The nuclei gained by this method were applicated to the bovine oocyte intraplasmic nuclear injection,which provided a new method of acquiring donor nuclei in nuclear transfer accordingly. The results indicated that the serum could successfully lyse the membrane of the donor cells. In this study,the effects of different donor cells in the development of reconstructed embryos were compared. the result showed that the cleavage rates of granular cells and ear fibroblasts were 62.62% and 67.89% ,the blastocyst rates were 23.88% and 18.92%,respectively,these data have no significant difference(P>0.05)between each other. The study also researched whether it can effect the nuclei transplant or not if the donor cell treated by low temperature(4℃)preservation. The result showed that the there was no obvious difference(P>0.05)in the cleavage rates and the blastocyst rates between the two groups which are treated with 4℃ preservation or not,no matter what the donor cell is granular cells or ear fibroblasts. The cleavage rates of granular cells with or without 4℃ preservation is 63.34% vs 62.03%,the data is 70.76% vs 66.67% on the ear fibroblasts. The blastocyst rates were 26.67% vs 24.45% on the granular cells,21.74% vs 19.56% on the ear fibroblasts. In the research of the influence of delaying activation with different time after nuclear injection on the cleavage rate and the development of reconstructed embryos,the reconstructed embryos with the delaying time of 3～ 5h has the higher cleavage rates than that of the embryos with 1～3h delaying time in the granular cells,and there had significant difference (73.91% versus 56.52%,P<0.05)between them. However,there has no significant difference between the blastocyst rates (23.53%,versus 15.38% ,P >0.05). In a word,these results demonstrated that it was feasible to prepare the donor nuclei by immunosurgery which was applicated to bovine somatic cell nuclear transfer(SCNT).
Effects of Donor Cell Categories on the Development of Buffalo Embryos Derived from Nuclear Transfer

Abstract:The development of nuclear transfer(NT)embryos derived from adult ear fibroblasts, fetal fibroblasts, fetal skin cells and ovary granulosa cells were compared. When electric fusion method was used for nuclear transfer, the fusion rate (46.0%), cleavage rate (53.9%) and blastocyte development rate (10.9%) of adult ear fibroblasts were significantly lower than those of fetal fibroblasts (64.5%, 70.1%, 21.6% respectively), fetal skin cells (71.5%, 70.8%, 22.1% respectively) and ovary granulosa cells (88.2%, 79.1%, 25.5% respectively). There was no significant difference in the cleavage and blastocyst development rate of reconstituted embryos among the later three donor cells. However, the fusion rate of ovary granulosa cells was significantly higher than that of the fetal fibroblasts and fetal skin cells (88.2% vs 64.4%, 71.5%,P0.05). When direct injection was used for nuclear transfer, the survive rate (78.2%) and cleavage rate (41.6 %) of fetal skin cells were significantly lower than those of adult ear fibroblasts (88.6%, 57.4%) and fetal fibroblasts (85.8%, 65.0%). The cleavage rate and blastocyte development rate (23.3%) of fetal fibroblasts were significantly higher than that of fetal skin cells (10.1%) and adult ear fibroblasts (12.0%). These results show that granulosa cells are suitable for nuclear transfer by electric fusion, however, the fetal fibroblasts are suitable for nuclear transfer by direct injection.
Influence of Morphology, Diameter and Passage of the Donor Cells on Porcine Somatic Cell Nuclear Transfer

Abstract:Oocytes on the surface of porcine ovaries were collected,in vitro matured and enucleated.Donor cells were dissected,cultured and passaged.The donor nucleus was transfered into the enucleated oocyte to reconstruct an embryo.The reconstructed embryos were fused and activated in order to discuss the developmental potential of the reconstructed embryos in the morphology and diameter and passage of donor cells and explore the transfer effect in the porcine somatic cells nuclear transfer.In result,the cleavage rate and blastocyst rate of reconstructed embryos using smooth granulosa cells as donor cells were higher than those of the rough granulosa cells as donor cells,and it were significantly different(P0.05).Fusion rates in two groups were no significantly different(P0.05).The fusion rate of reconstructed embryos using diameter 15 μm donor cells was lower,and it was significantly different compared with the fusion rate of the diameter 30 μm group(P0.05).The cleavage rate of the diameter of 20 to 30 μm group was significant compared with the one of the diameter 15 μm group(P0.05),but not significant compared with the diameter 30 μm group(P0.05).The cleavage rate and blastocyst rate(63.73% 21.54%)of reconstructed embryos using diameter of 20 to 30 μm as donor cells were higher than other groups,and the blastocyst rate was significantly different(P0.05).The efficiency of nuclear transfer in different culture passages of fetal fibroblast after serum starvation was examined.The fusion rate of 6 to 9 culture passages was higher than those of 3 to 5 culture passages and over 10 culture passages,but the cleavage rate and blastocyst rate among the three groups were not significantly different(P0.05).In conclusion,the smooth granulose cells as donor cells were beneficial for the development of the constructed embryos,donor cells of the diameter from 20 to 30 μm were better fit for porcine somatic cell nuclear transfer,and fetal fibroblast from 6 to 9 culture passages were better suitable for as donor cells of nuclear transfer.

Abstract:The development of the spermatogonial transplantation technique has given new impetus to reveal spermatogenesis, self-renewable and hormone control of spermatogonial stem cells proliferation. It will be a valuable technique for animal breeding, rare species preservation and transgenic animal generation. This article provides a comprehensive overview of isolation, purification, cryopreservation and transplantation of spermatogonial stem cells. Perspectives were forecasted in applications of spermatogonial stem cell transplantation in domestic animals.
Effect of Donor Cell Treatment on Development of Somatic Cell Nuclear Transfer Embryos in Buffalo

Abstract:The success of nuclear transfer is thought to be principally dependent on transfer of nuclei in G_0 or G_1 phase.In this study,the cell cycle characteristic of buffalo fetal fibroblasts and granulosa cells cultured in cycling,culture to confluency,serum-starvation and a reversible cell cycle inhibitor aphidicolin Aphidicolin(APD) 0.1 μg/mL+0.5% fetal bovine serum(FBS) treatment were investigated.This was obtained by using flow cytometry to measure cellular DNA content and calculated the percentages of cells in G_0+G_1,S,and G_2+M phase.Moreover,the developmental ability of buffalo embryos derived from fetal fibroblasts and granulosa cells that cultured to confluency,serum-starvation and 0.1 μg/mL APD+0.5% FBS treatment were examined.As a result,both buffalo fetal fibroblasts and granulosa cells,cycling culture resulted in lower percentages of cells in G_0+G_1(52.12% and 50.07%),but significantly higher percentages of cells in G_0+G_1 when cells treated with 0.1 μg/mL APD+0.5% FBS(87.89% and 89.04%)than cultured to confluency(66.03% and 63.18%) and serum-starvation(77.87% and 76.57%)(P0.05).Conversely,cycling cells contained more percentages of cells in G_2+M than other treatments,and cultured to confluency resulted in higher percentages cells existing in S phase.However,serum starvation induced more percentages of DNA fragmentation than other treatments(P0.05).The rate of cleavage and blastocysts development of embryos reconstructed with fetal fibroblasts or granulosa cells treated with 0.1 μg/mL APD+0.5%FBS were significantly higher than that of serum starvation group and culture to confluency group(fibroblasts: 70.14% vs 58.29% and 55.90%,21.33% vs 10.55% and 8.72%;granulose: 74.35% vs 60.51% and 57.61%,22.17% vs 10.26% and 9.78%,P0.05),however there was not significantly different in the cleavage rate and embryonic development of blastocysts between embryos derived from serum starvation and culture to confluency group(P0.05).When recipients received embryos derived from donor cells treated with APD,one recipient became pregnant and supported the development to term.The results showed that donor cells treated with cells cycle inhibitor 0.1 μg/mL APD+0.5%FBS was an effective cell cycle synchronization method,which could obtain higher percentage in G_0/G_1 phase and be used to produce buffalo nuclear transfer blastocyst and offspring efficiently,but serum starvation was inefficient in our system.

Abstract:In interspecies somatic cell nucleus transfer (iSCNT), somatic nuclei of one species are transplanted into enucleated oocytes of another species. This approach may be used as an effective method for cloning endangered species or establishing primate embryonic stem cells. However, very few work has been carried out so far. The incomplete nuclear reprogramming and the early development block of iSCNT embryos are still an issue. This review focused on these issues embryonic genome activation, mt/genomic DNA compatibility, epigenetics, nuclear remodeling, SCNT protocol and culture system. The challenges and possible solutions for the successful iSCNT scheme have also been discussed in this review.
Primary Study on the Nuclear Transfer of Bovine Somatic Cells

Abstract:Factors related to nuclear transfer of bovine somatic cells were examined in this study. When electronic field intensity and pulse number were fixed at 100v/mm and 2 times, 10#mu#s of pulse time resulted in significantly more cytoplasts fused with fibroblasts in comparison with 30#mu#s of pulse time, Pre-activation of fibroblasts with 7% ethanol could increase the cleavage rates of reconstructed embryos and improved the development of them. The total rates of blastocyst derived from T II enucleated recipient oocytes were significantly lower than that of recipient oocytes enucleated at M II. Recipient oocytes enucleated at M II were superior to oocytes enucleated at T II in using as recipient for bovine somatic nuclear transfer. High percentages of blastocysts obtained when reconstructed embryos were activated at 3h in comparison with 0h and 6h after injection, Interaction of donor cells with recipients cytoplasmic factors for a certain period may be important for the development of reconstructed embryos.

Abstract:Somatic cell nuclear transfer (SCNT) is the only reproductive engineering technique that can confer genomic totipotency on somatic cell. SCNT is of great significance for animal germplasm conservation, animal husbandry development, and biomedical research. Although many research advances have been made in this technology, the developmental rate of SCNT mammalian embryos is very low, which seriously limits the application of SCNT in animal husbandry and biomedicine. The primary reason for the low efficiency of cloned embryos is somatic cell reprogramming errors or incomplete reprogramming. These errors or incompleteness present as the abnormal expression of imprinted gene Xist, abnormal DNA methylation, and abnormal histone modification. In this review, we summarize the main factors that influence the low development efficiency of mammalian cloned embryos to provide theoretical reference for the research and practice of improving somatic cell cloning efficiency.
Study of Murine Reconstructed Embryo by Somatic Nuclear Transfer

Abstract:Objective To develop nuclear transplantation of embryonic stem cells in mice.Methods Through microinjection, ICR mouse fibroblasts nucleus was transferred into the cytoplasm of C 57 mouse enucleated oocyte to reconstruct embryo by micromanipulation. The reconstructed embryos were treated with chemical activation or three different pulse electric currents, then cultured in oviduct or in vitro. The cultured embryos were collected three days later to identify embryonic development. Results 71.91% of transferred enucleated oocytes survived. Chemical activation rate was 75.09%.The embryonic fusional potential was the best by pulse current 60V(80μs, five times activation) than by pulse current 70V or 80V(80μs,any times activation),and the embryonic development potential was better by pulse current 70V(80μs, two times activation) than by pulse current 60V or 80V(80μs,any times activation).Conclusion The nuclear transfer method is with success basically.

Abstract:Nuclear transfer was carried out with mammary epithelial cells,fetal fibroblasts and cumulus-granulosa cells by method of intracytoplast injection and electro-fusion respectively.The influence of cell types,cell refrigeration and the methods of reconstruction of donor cells to developmental ability of cloned embryos were compared.The donor cells were treated with/without refrigeration,and were synchronized at G_0/G_1 stage by serum deprivation before nuclear transfer.Results:(1)The developmental ability of cloned embryos originated from mammary epithelial cells(blastocyst rate: 7.14%) was significantly lower than those from fetal fibroblasts(16.19%) and cumulus-granulosa cells(19.01%)(P0.05).(2) There was no significant difference among three kinds of cell(P0.05) mentioned above in the influence of cell refrigeration on developmental ability of cloned embryos.(3) The rate of reconstruction with electro-fusion(69.52%)(P0.01)was significantly higher than that with intracytoplast injection(59.20%) to mammary epithelial cells.However,there was no significant difference between these two methods(P0.05) for fetal fibroblasts.For cumulus-granulosa cells,the rate with intracytoplast injection(82.17%) was significantly higher than that with electro-fusion(50.99%)(P0.05),and the rate of forming blastula was a little higher than that with electro-fusion,but not significantly different.Conclusions: Fetal fibroblasts and cumulus-granulosa cells were superior to sustain the developmental ability of cloned embryos.Cell refrigeration had no significant effect on the reconstruction of cloned embryos.Comparing the ways of reconstructing embryos,it was suggested that electro-fusion was better for mammary epithelial cells,but intracytoplasm injection was better than electro-fusion for cumulus-granulosa cells.Both methods were fit to fetal fibroblasts.

Abstract:In this study,with bovine auricle fibroblasts used as nuclear donor,the effects of nuclear donor with serum starvation vs.contact inhibition,frozen vs.non-frozen,as well as the age and sex of animals from which nuclear donor cells were derived on the efficiency of nuclear transfer were investigated and compared.The results showed that the rates of blastocysts derived from nuclear donor cells pretreated with contact inhibition or non-freezing were significantly higher than those derived from nuclear donor cells pretreated with serum starvation or frozen respectively(P<0.05).Moreover,when female cells were used as nuclear donors,the blastocyst rate of cloned embryos was significantly higher than those derived from male cells(P<0.05).However,the age of animal from which donor cells derived could not affect the efficiency of nuclear transfer(P>0.05).When adult female bovine auricle fibroblasts were pretreated with contact inhibition and were not frozen,the results indicated those were significantly beneficial to the development of nuclear transfer embryos.

Abstract:The effects of donor cell cycle and different passage on interspecies nuclear transfer were discussed in this research.Bovine oocytes stained by hochest33342 were enucleated as receptor and human fibroblast in different cell cycle and passage were used as donor for nuclear transfer.After electric fusion,these reconstructed embryos were activated by 6-DAMP+ethanol and cultured in SOFaa medium.The fusion and development rates of the treatment in the 20th passaage cell without starvation(P<0.01) were significantly lower than that in the 20th passage with starvation(P<0.05),and were significantly lower than that in the 8th passage with starvation and without starvation(P<0.01).There were no significant differences in 8-cell embryo rates and morula-blastocyst rates in four treatments.The result indicates that starvation treatment may not be the essential factor for low passage donor cell and when donor cell is in high passage,starvation treatment can be the important way to improve the efficiency of interspecies nuclear transfer by increasing fusion rate and development rate significantly.
