Experimental Study of Ultrasound Hyperthermia Combined with CDDP on Human Oral Squamous Cell Carcinoma Xenografts in Nude Mice

Abstract:Objective To evaluate the interaction of ultrasound hyperthermia combined with chemotherapy by observing therapeutic efficacy of ultrasound hyperthermia combined with CDDP on human oral squamous cell carcinoma xenografts in nude mice and the tolerance of normal tissue. Methods Anesthetized nude mice with hypodermically injection. Tca8113 carcinomas underwent one of the following treatments: (a) localized ultrasound hyperthermia; (b) CDDP chemotherapy; (c) ultrasound hyperthermia combined with CDDP (CDDP was given 15 min prior to hyperthermia; (d)Sham-treated animals served as controls. Tumors were investigated for: (a) tumor volume growth; (b) tumor control rate; (c) interaction between hyperthermia and chemotherapy. Results Both ultrasound hyperthermia and CDDP caused a delay in tumor growth when given individually. In contrast, ultrasound hyperthermia combined with CDDP resulted in a much more significant enhancement in tumor control, with an enhancement ratio of 1.2. Conclusion Ultrasound heating technology has significant advantages that allow for a more exact and reliable control of the therapeutic temperature, which met the demand of individualized remedy. Ultrasound hyperthermia combined with CDDP resulted in a pronounced tumoricidal effect, and there was synergistic effect between these two modalities.
