Effect of Acute Hypoxia and Intermittent Hypoxic Acclimatization on Erythropoietin Levels in Plasma and Hepatic and Renal Tissues of Rats

Abstract:Objective To explore the influences of acute hypoxia and intermittent hypoxia acclimatizationon erythropoietin (EPO) level in plasma and EPO gene expression of hepatic and renal tissues of rats. Methods Twenty-four Wistar rats were divided into three groups: normal control group (NC), intermittent hypoxia acclimatization group (IH) and acute hypoxia group(AH). Rats of AH group underwent 8000 m altitude exposure for 12 h, while rats of IH group were acclimatized intermittently(4 h per day) to 3000 m and 5000 m altitude for 2 weeks respectively before 8000 m altitudeexposure. EPO plasma level and EPO gene expressions in tissues were measured with MTT and reverse transcription-polymerase chain reaction analysis (RT-PCR) respectively. Results As compared with those in NC group, plasma EPO level in rats of AH group was significantly higher (P0.05), and EPO gene expression in hepatic and renal tissues was also obviously elevated (P0.05). The blood RBC count in IH group was significantly higher than that of NC group (P0.05), while the plasma EPO level and EPO gene expression in renal and hepatic tissues from rats of IH group were all apparently lower than those of AH group (P0.05). Conclusions Intermittent hypoxia acclimatizationcan enhance the hypoxia tolerance of rats and inhibit hypoxia-induced increment of EPO level in plasma and EPO gene expression in hepatic and renal tissues.
